Ischemic accumulation of succinate induces Cdc42 succinylation and inhibits neural stem cell proliferation after cerebral ischemia/reperfusion

Ischemic accumulation of succinate causes cerebral damage by excess production of reactive oxygen species. However, it is unknown whether ischemic accumulation of succinate affects neural stem cell proliferation. In this study, we established a rat model of cerebral ischemia/reperfusion injury by occlusion of the middle cerebral artery. We found that succinate levels increased in serum and brain tissue(cortex and hippocampus) after ischemia/reperfusion injury. Oxygen-glucose deprivation and reoxygenation stimulated primary neural stem cells to produce abundant succinate. Succinate can be converted into diethyl succinate in cells. Exogenous diethyl succinate inhibited the proliferation of mouse-derived C17.2 neural stem cells and increased the infarct volume in the rat model of cerebral ischemia/reperfusion injury. Exogenous diethyl succinate also increased the succinylation of the Rho family GTPase Cdc42 but repressed Cdc42 GTPase activity in C17.2 cells. Increasing Cdc42 succinylation by knockdown of the desuccinylase Sirt5 also inhibited Cdc42 GTPase activity in C17.2 cells. Our findings suggest that ischemic accumulation of succinate decreases Cdc42 GTPase activity by induction of Cdc42 succinylation, which inhibits the proliferation of neural stem cells and aggravates cerebral ischemia/reperfusion injury.

Human embryonic stem cells （heSCs） can self-renew indefinitely and differentiate into all cell types in the human body. Therefore, they are valuable in regenerative medicine, human developmental biology and drug discovery. A number of heSC lines have been derived from the chinese population, but limited of them are available for research purposes. here we report the derivation and characterization of two heSC lines derived from human blastocysts of chinese origin. These heSCs express alkaline phosphatase and heSC-specific markers, including Oct4, Nanog, SSEA-3, SSEA-4, TRA- 1-60 and TRA-1-81. They also have high levels of telomerase activity and normal karyotypes. These ceils can form embryoid body in vitro and can be differentiated into all three germ layers in vivo by teratoma formation. The newly established heSCs will be distributed for research purposes. The availability of heSC lines from the chinese population will facilitate studies on the differences in heSCs from different ethnic groups.

Aging biomarkers are a combination of biological parameters to(i)assess age-related changes,(ii)track the physiological aging process,and(iii)predict the transition into a pathological *** a broad spectrum of aging biomarkers has been developed,their potential uses and limitations remain poorly *** immediate goal of biomarkers is to help us answer the following three fundamental questions in aging research:How old are we?Why do we get old?And how can we age slower?This review aims to address this ***,we summarize our current knowledge of biomarkers developed for cellular,organ,and organismal levels of aging,comprising six pillars:physiological characteristics,medical imaging,histological features,cellular alterations,molecular changes,and secretory *** fulfill all these requisites,we propose that aging biomarkers should qualify for being specific,systemic,and clinically relevant.

Calciphylaxis is a rare disease characterized histologically by microvessel calcification and microthrombosis,with high mortality and no proven ***,we reported a severe uremic calciphylaxis patient with progressive skin ischemia,large areas of painful malodorous ulcers,and mummified *** of the worsening symptoms and signs refractory to conventional therapies,treatment with human amnion-derived mesenchymal stem cells(hAMSCs)was *** release inspections of hAMSCs,efficacy,and safety assessment,including cytokine secretory ability,immunocompetence,tumorigenicity,and genetics analysis in vitro,were *** further performed acute and long-term hAMSC toxicity evaluations in C57BL/6 mice and rats,abnormal immune response tests in C57BL/6 mice,and tumorigenicity tests in neonatal Balbc-nu nude *** the preclinical research,the patient was treated with hAMSCs by intravenous and local intramuscular injection and external supernatant application to the *** followed up to 15 months,the blood-based markers of bone and mineral metabolism improved,with skin soft tissue regeneration and a more favorable profile of peripheral blood mononuclear *** biopsy after 1-month treatment showed vascular regeneration with mature noncalcified vessels within the dermis,and 20 months later,the re-epithelialization restored the integrity of the damaged *** infusion or local treatment-related adverse events ***,this novel long-term intravenous combined with local treatment with hAMSCs warrants further investigation as a potential regenerative treatment for uremic calciphylaxis due to effects of inhibiting vascular calcification,stimulating angiogenesis and myogenesis,anti-inflammatory and immune modulation,multidifferentiation,re-epithelialization,and restoration of integrity.

Objective Congenital intellectual disability（ID） is usually caused by the abnormity of genes or chromosomes. The ID-related gene mutations of chinese population which are different from others need systematic investigations. We screened the ID-related gene mutations from chinese ID children by sequencing, and identified some novel mutations of genes including fibroblast growth factor 13（FGF13）, which is a candidate gene for a syndromal X chromosome linked ID（XliD）. The genetic deletion of Fgf13 in mice results in neuronal migration defects and weaken learning and memory in our previous study. We studied the pathogenicity and related mechanisms of the mutations of Fgf13 in influencing ID phenotypes. Methods Whole genome sequencing and genetic analysis of family trees were performed to screen mutation sites. Cultured i PSCs-derived neural stem cells and neurons were used to investigate the molecular mechanisms. The construction of gene mutation transgenic mice and related behavioral tests were used to test the pathogenic phenotypes at various levels. Results（1） A single base mutation was occurred in Fgf13 gene of 3 ID boys, which caused down-regulation of the protein but not the m RNA expression of FGF13 in heK293 cells.（2） i PSCsderived neural stem cells from those mutation-carrying ID children further confirmed down-regulation of the protein but not the m RNA expression of FGF13, and a rescued single base mutation reversed those alterations.（3） Behavior tests showed that the knockin mice carrying the point mutation of Fgf13 gene resulted in the learning and memory defects similar with those in ID patients, and exhibited delayed migration and increased axonal branching of neurons in the developing brains. Conclusion Our study provides IDrelated pathogenic genes and related mechanisms of chinese population.

Burkitt淋巴瘤患者的临床特点及影响预后的因素分析

目的:探讨Burkitt淋巴瘤(BL)患者的临床特点并分析其疗效。方法:收集并回顾性分析27例BL患者的临床资料,对患者临床特征及实验室指标进行分析,同时进行生存和预后影响因素分析。结果:本研究患者以成人为主,中位发病年龄为30(15-83)岁,男性∶女性为3.5∶1,所有患者无EB病毒感染,92.6%患者存在结外器官受累,其中40.7%的患者起病时为白血病表现,Ⅲ-Ⅳ期占85.2%,IPI评分高危/高中危组占74.1%;分子生物学方面,5例患者进行了二代测序,均具有MYC基因突变,最常见突变为CCND3、ID3、TP53突变。治疗总有效率为85.2%,完全缓解率为63.0%,部分缓解率为22.2%,5年无进展生存率和5年总生存率均为76.6%,高剂量甲氨蝶呤治疗组疗效优于未使用高剂量甲氨蝶呤治疗组,其中LMB89方案治疗组完全缓解率为78.6%,总有效率为100%,5年生存率为92.9%,显著优于其他治疗方案组;无法耐受高剂量化疗的患者进行自体造血干细胞移植作为巩固治疗,具有生存获益。单因素分析显示,ECOG评分、血清乳酸脱氢酶大于500 U/L、白细胞水平、是否存在中枢系统受累、近期疗效及是否应用R-LMB89方案为影响患者预后的因素。结论:成人BL具有高度侵袭性,高剂量化疗LMB89方案在本研究中显示出较好的疗效和生存率,对于无法耐受高剂量甲氨蝶呤患者,一线自体造血干细胞移植作为巩固治疗为患者可供选择的治疗方案。
