背景:在纤维化过程中,转化生长因子(TGF-β1)通过活化下游Smad2/3信号起正调节作用,但通过上调Smad7起抑制作用。另外我们发现上调Smad7可抑制NF-κB炎症信号通路而控制炎症。本实验通过研究TGF-β/Smad的信号,特别是Smad7基因的失调引致糖尿病肾损害的发病机制,并运用超声微波基因疗法探讨Smad7靶向治疗糖尿病肾病的新方法。第一部分研究:TGF-β/Smad的激活与糖尿病肾损害有密切关系方法1:大量临床研究表明,C反应蛋白(CRP)被认为是糖尿病的危险因素,但作用机制并不清楚。我们利用CRP转基因和野生型小鼠建立糖尿病肾病模型,观察CRP在成模24周小鼠的糖尿病肾病中的作用。结果1:CRP转基因的糖尿病小鼠比野生型糖尿病出现更严重的微量蛋白尿,增加肾损伤蛋白-1,促进巨噬细胞和T细胞浸润、炎症因子(Il-1β,TNF-α)表达,加重肾纤维化(胶原Ⅰ、Ⅲ、Ⅳ)。进一步研究发现CRP转基因糖尿病小鼠肾脏炎症和纤维化与TGF-β/Smad信号通路进一步活化有密切关系。由于TGF-β/Smad的活化受Smad7的负调控,所以我们假设Smad7在糖尿病肾损害起关键的负调控作用。这一假设在第二部分实验中得以证实。第二部分研究:Smad7基因敲除加重而Smad7基因高表达抑制糖尿病肾损害方法2:利用Smad7基因敲除小鼠研究该基因在糖尿病肾病中的作用,并用超声微泡技术导入Smad7基因到糖尿病大鼠肾病模型中,来研究Smad7基因的保护和治疗作用。结果2:研究发现Smad7基因敲除的糖尿病鼠比野生型糖尿病鼠出现更严重的肾损害,包括加重了微量蛋白尿、肾脏纤维化指标(胶原Ⅰ,Ⅳ和纤连蛋白)、肾脏炎症指标(Il-1β,TNF-α,MCP-1,ICAM-1和巨噬细胞侵润)。进一步研究发现:Smad7基因敲除加重糖尿病小鼠肾脏炎症和纤维化与促进NF—κB炎症信号通路和TGF-β1/Smad纤维化信号通路激活有关。为验证上述发现,我们应用超声微波将Smad7高表达基因转入糖尿病大鼠肾内。实验发现,Smad7基因疗法对血糖水平无影响,但能显著抑制糖尿病鼠的微量蛋白尿形成,降低TGF-β1/Smad介导的肾脏纤维化和NFκB调控的肾脏炎症反应。结论:Smad7的下调可促进糖尿病肾损害。增加NF—κB炎症信号和TGF-β1/Smad纤维化信号通路激活可能是下调Smad7促进糖尿病肾损害的主要机理。超声微波Smad7基因导入技术为糖尿病肾病的靶向治疗提供新的疗法。

At present,a variety of vaccines have been approved,and existing antiviral drugs are being tested to find an effective treatment for coronavirus disease 2019(COVID-19).However,no standardized treatment has yet been approved by the World Health *** virally encoded chymotrypsin-like protease(3Cl^(pro))from severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),which facilitates the replication of SARS-CoV in the host cells,is one potential pharmacological target for the development of antiSARS *** search engines,such as Web of Science,Google Scholar,Scopus and PubMed,were used to retrieve data on the traditional uses of medicinal plants and their inhibitory effects against the SARS-CoV 3Cl^(pro).Various pure compounds,including polyphenols,terpenoids,chalcones,alkaloids,biflavonoids,flavanones,anthraquinones and glycosides,have shown potent inhibition of SARS-CoV-2 3Cl^(pro) activity with 50% inhibitory concentration(IC_(50))values ranging from 2-44μg/***,most of these active compounds,including xanthoangelol E(isolated from Angelica keiskei),dieckol 1(isolated from Ecklonia cava),amentoflavone(isolated from Torreya nucifera),celastrol,pristimerin,tingenone and iguesterin(isolated from Tripterygium regelii),tannic acid(isolated from Camellia sinensis),and theaflavin-3,3’-digallate,3-isotheaflav1in-3 gallate and dihydrotanshinone I(isolated from Salvia miltiorrhiza),had IC_(50)values of less than 15μg/*** mechanistic studies of several active compounds revealed that their mode of inhibition was dose-dependent and competitive,with K_(i)values ranging from 2.4-43.8μmol/*** the significance of plant-based compounds and the many promising results obtained,there is still need to explore the phytochemical and mechanistic potentials of plants and their *** medicinal plants could serve as an effective inexpensive nutraceutical for the general public to help manage COVID-19.

Gated transport measurements are the backbone of electrical characterization of nanoscale electronic devices. Scanning gate microscopy (SGM) is one such gating technique that adds crucial spatial information, accessing the localized properties of semiconductor devices. Nanowires represent a central device concept due to the potential to combine very different materials. However, SGM on semiconductor nanowires has been limited to a resolution in the 50-100 nm range. Here, we present a study by SGM of newly developed III–V semiconductor nanowire InAs/GaSb heterojunction Esaki tunnel diode devices under ultra-high vacuum. Sub-5 nm resolution is demonstrated at room temperature via use of quartz resonator atomic force microscopy sensors, with the capability to resolve InAs nanowire facets, the InAs/GaSb tunnel diode transition and nanoscale defects on the device. We demonstrate that such measurements can rapidly give important insight into the device properties via use of a simplified physical model, without the requirement for extensive calculation of the electrostatics of the system. Interestingly, by precise spatial correlation of the device electrical transport properties and surface structure we show the position and existence of a very abrupt (<10 nm) electrical transition across the InAs/GaSb junction despite the change in material composition occurring only over 30-50 nm. The direct and simultaneous link between nanostructure composition and electrical properties helps set important limits for the precision in structural control needed to achieve desired device performance.

Methodological impact of starch determination on starch content and ileal digestibility of starch in grain legumes for growing pigs

Background： Grain legumes represent a valuable energy source in pig diets due to their high starch content. The present study was conducted to determine the content and apparent ileal digestibility（AID） of starch in different grain legume cultivars for pigs by means of both a polarimetric and enzymatic method for starch ***： Three experiments were conducted with six barrows each which were fitted with ileal T-cannulas. In total, 18 diets including six different cultivars of faba beans（Vicia faba l.） and peas（Pisum sativum l.）, five different cultivars of lupins（lupinus luteus l., lupinus angustifolius l.）, and one diet with a soybean meal（SBM） were ***： The starch content of faba beans and peas was greater（P ＆lt; 0.05） when determined polarimetrically than enzymatically（438 vs. 345 g/kg dry matter（DM） in faba beans and 509 vs. 390 g/kg DM in peas, respectively）.Considerable lower starch contents were obtained in lupins and SBM, with 82 and 48 g/kg DM（analyzed polarimetrically） and ＆lt;1.1 and 3 g/kg DM（analyzed enzymatically）, respectively. Mean values for contents of neutral detergent fiber（NDF） and acid detergent fiber（ADF） in grain legumes ranged from 111 and 79 g/kg DM in peas to248 and 207 g/kg DM in lupins, respectively. Contents of condensed tannins in the colored flowered faba bean cultivars ranged from 2.1 to 7.4 g/kg DM. The AID of starch was greater（P ＆lt; 0.05） in pea than in faba bean cultivars,and using the polarimetric starch determination method resulted in greater（P ＆lt; 0.05） digestibility values than using enzymatic starch analysis（84 vs. 80% in faba beans and 86 vs. 83% in peas）. Moreover, AID of starch differed（P ＆lt; 0.05）within pea cultivars and starch digestibility in faba beans decreased linearly（P ＆lt; 0.05） as the content of condensed tannins increased. AIM: To investigate relapse predictors in chronic hepatitis C (CHC) patients with end-of-treatment response (ETR), after pegylated interferon-α (PegIFN-α) and ribavirin treatment. METHODS: In a retrospective study we evaluated a spectrum of predictors of relapse after PegIFN-α and ribavirin treatment in 86 CHC patients with ETR. Viral loads were determined with real-time reverse transcrip-tion polymerase chain reaction. Hepatitis C virus geno-typing was performed by sequencing analysis. Patients with genotype 1 were treated for 48 wk with 180 μg PegIFN-α2a or 1.5 μg/kg PegIFN-α2b once weekly plus ribavirin at a dosage of 1000 mg/d for those under 75 kg or 1200 mg/d for those over 75 kg. Patients with geno- types 2 and 3 were treated for 24 wk with 180 μgPegIFN-α2a or 1.5 μg/kg PegIFN-α2b once weekly plus ribavirin at a dosage of 800 mg/d. RESUlTS: In all ETR patients, binary logistic regression analysis identif ied absence of complete early virological response (cEVR) (OR 27.07, 95% CI: 3.09-237.26, P lkaline phosphatase (AlP) levels prior to therapy l (OR: 6.16, 95% CI: 2.1-18.03, P 26 kg/m2 (OR: 8.27, 95% CI: 2.22-30.84, P lapse. When cEVR patients were analyzed exclusively, AlP prior to therapy l remained the only predictor of relapse. CONClUSION: lower levels of AlP prior to, during and after therapy seem to be associated with a higher risk of relapse in CHC patients with ETR.

AIM: To investigate whether chronic H pylori infection has the potential to induce pancreatitis in the Mongolian gerbil model, and whether it is dependent on an intact type Ⅳ secretion system. METHODS: Mongolian gerbils were infected with wild type (WT) H pylori typeⅠstrain B128 or its isogenic mutant B128 ?cagY (defective type Ⅳ secretion). After seven months of infection, H pylori was reisolated from antrum and corpus and H pylori DNA was analyzed by semi- nested polymerase chain reaction (PCR). Inflammation and histological changes were documented in the gastric antrum, corpus, and pancreas by immunohistochemistry. Cytokine mRNA, gastric pH, plasma gastrin, amylase, lipase, and glucose levels were determined. RESUlTS: The H pylori infection rate was 95%. Eight infected animals, but none of the uninfected group, developed transmural inflammation and chronic pancreatitis. Extensive interstitial fibrosis and inflammation of the pancreatic lobe adjacent to the antrum was confirmed by trichrome stain, and immuno- histochemically. Pro-inflammatory cytokine mRNA was significantly increased in the antral mucosa of all infected gerbils. In the corpus, only cytokine levels of WT-infected animals and those developing transmural inflammation and pancreatitis were significantly increased. levels of lipase, but not glucose or amylase levels, were significantly reduced in the pancreatitis group. H pylori DNA was detected in infected antral and corpus tissue,but not in the pancreas. CONClUSION: H pylori infection is able to induce chronic pancreatitis in Mongolian gerbils independently of the type Ⅳ secretion system, probably by an indirect mechanism associated with a penetrating ulcer.

We study the vertical distribution of the highly inclined galaxies from the Continuum Halos in Nearby Galaxies—an EVlA Survey(CHANG-ES).We explore the feasibility of photometrically deriving the HⅠdisk scale heights from the moment-0 images of the relatively edge-on galaxies with inclination>80°,by quantifying the systematic broadening effects and thus deriving correction equations for direct *** corrected HⅠdisk scale heights of the relatively edge-on galaxies from the CHANG-ES sample show trends consistent with the quasiequilibrium model of the vertical structure of gas *** procedure provides a convenient way to derive the scale heights and can easily be applied to statistical samples in the future.
