1 Physicochemical a spects of multilayered polyelectrolyte f ilms multilayered polyelectrolyte films are obtained by al-ternated depositions on a solid surface of polyanions and polycations[1].The driving force for the build-up results from the charge excess which appears on the top of the film after each new polyelectrolyte *** film top becomes thus positively(respectively negatively)charged when the film is ending by a polycation(respec-tively polyanion).Various polyelectrolyte systems have been used to realize such architectures,in particular syn-thetic polyelectrolytes(poly-styrene sulfonate:PSS,poly-allylylamine:PAH,etc.);polypeptides(poly-glutamic acid:PGA,poly-lysine:PLL,etc.),polysaccharides[-](hyaluronane:HA,chitosan:Chi,etc.).

Systems describing the dynamics of proliferative and quiescent cells are commonly used as computational models, for instance for tumor growth and *** on the very earliest stages of hematopoiesis, stem cells and early progenitors, the authors introduce a new method, based on an energy/Lyapunov functional to analyze the long time behavior of solutions. Compared to existing works, the method in this paper has the advantage that it can be extended to more complex situations. The authors treat a system with space variable and diffusion, and then adapt the energy functional to models with three equations.

All of metabotropic glutamate-like receptors are dimers and share a cysteine-rich domain(CRD) which contains 9 high conserved cysteines,but how the CRD servers as a mediator in signal transduction from VFT to HD in vivo is still largely *** we first replaced the 9 conserved cysteines by alanine in mGluR2,and find the most remarkable pharmacology phenotype is the high constitutive activity of *** on a series of results, we have proved the spontaneous formed disulfide bridge between two new‘free’C519 in each monomer accounts for the constitutive activity of *** of cysteine for the residue which close to C519 and in the putative CRD-CRD interface to form analogous inter-monomer bridge also produces constitutive *** similar results are also gain in mGluR4,mGluR5 and *** into account the bioinformatics results,we demonstrate that the activation involves the horizontal nearness and the tangent rotation of two CRD in each monomer, thereby explain how agonist binding to VFT induces allosteric conformation change conduct to effector *** our study is also important for medical research and drug screening.

对 GABA R 调控的下游信号转导的早期研究表明,功能性的 GABA R 可与 G/o 型家族的 G 蛋白耦联。在神经细胞中突触前 GABA R 主要通过 G亚基抑制 Ca通道,突触后 GABA R 也通过 G亚基打开 K通道。另外,GABAR 通过 G/G和 G亚基激活或抑制腺苷酸环化酶(adenylyl cyclase,AC)的活性,从而增加或减少 cAmP 在细胞内的产量与浓度。但是对于 GABA R 对受体下游级联放大信号通路的调节途径与分子机制还不清楚。通过对原代培养小脑神经细胞的研究发现,GABA R 受体的特异性激活可以诱导 G/PI3K/ERK/ CREB 信号通路,激活的 pCREB 进入细胞核内调节下游基因的表达,特别值得注意的是 CGP7930能单独激活 GABA R 受体介导的 G/PI3K/ERK/CREB 信号通路,而 ERK/CREB 信号通路对细胞的长期效应(基因转录与表达,神经突触的可朔性等)有着密切的联系,并最终调节着神经系统的学习与记忆等功能。同时该研究发现 GABA R 的激活也可以诱导下游的 G/Src/PI3K/Akt 信号通路,在中枢神经系统中该信号通路可能调节着神经细胞的存活与迁移,轴突的形成,突触可塑性以及学习与记忆的形成等,其具体的生理功能还有待进一步研究。以上研究结果为进一步揭示 GABA R 受体激活机制与生理功能及相关神经疾病的病理机制等奠定了基础,同时也为深入揭示 GPCR 家族 C 受体变构药(如 CGP7930等)的药理意义提供了非常重要的实验依据。

Hyponatremia during a Severe Pneumococcal meningitis with Renal Salt Wasting: A Cerebral Salt Wasting Syndrome?

Background: The existence and prevalence of cerebral salt wasting and its differentiation from syndrome of inappropriate antidiuretic hormone have been controversial. This controversy stems from overlapping clinical and laboratory findings and an inability to assess the volume status of these patients. Objectives:To present a case of a cerebral salt wasting syndrome secondary to a bacterial mengitis and to emphasize the difficulty to assess the diagnosis. Case report: A 51-year-old male admitted to the ICU for a severe bacterial meningitis who developed, four days later, hyponatremia associated with hypovolemia due to a renal salt wasting. Clinical and biological parameters were collected;electrolytes balances and salt clearances were calculated to best investigate this hydroelectrolytic disorder. Patient's volume status and improvement after fluid and sodium replacement highly suggests the diagnosis of a cerebral salt wasting syndrome. Conclusion: A thorough analysis of this case showed that occurrence of hyponatremia and renal salt wasting after brain agression is not univocal underlying the fact that the diagnosis of cerebral salt wasting should be assessed only after a careful examination of the different determinants of the hydroelectrolytical balance.

The ruminal redox potential (Eh) can reflect the microbiological activity and dynamics of fermentation in the rumen. It might be an important indicator of rumen fermentation in combination with pH. However, the ruminal Eh has been rarely studied in dairy cows due to the difficulty of its measurement, and the relationship between ruminal Eh and pH is not clear. The objective of this study was to investigate the relationship between ruminal Eh and pH of dairy cows by meta-analysis of systematic measurements from different experiments. A database was constructed from 22 experiments on cannulated dairy cattle including 57 dietary treatments. The ruminal pH and Eh were measured without air contact between 0 and 8 h post-feeding. The results demonstrated a quadratic correlation between ruminal Eh and pH with a reliable within-animal variation (Eh = -1697 + 540.7 pH -47.7 pH2, nobservation = 70, nanimal = 26, P Eh, but not always to the same extent. Some of them still influenced the relationship between ruminal Eh and pH. While the mechanism of the interaction between ruminal Eh and pH remains to be elucidated, it would be interesting to associate Eh to microbial profile, ruminal VFA concentration and milk production performance in future studies.

Trastuzumab Re-Introduction with FOLFIRI for Treatment of HER2 Overexpression-Advanced Gastric Adenocarcinoma Following Failure of Other Trastuzumab-Based Chemotherapy Regimens

A 67-year-old man diagnosed with HER2 overexpression advanced gastric adenocarcinoma and metastasis to liver and lungs was admitted for tertiary care. He received a third line chemotherapy that consists of trastuzumab combined with FOLFIRI regimen (irinotecan plus 5-FU/LV) following a disease relapse after an initial successful response to a combination of 5FU + oxaliplatin and trastuzumab. The patient showed a favorable and prolonged response to it. In addition the chemotherapy was well tolerated and devoid of remarkable side effects. The response to trastuzumab + FOLFIRI was assessed clinically and through CT scan imaging and upper gastrointestinal endoscopy. This case report shows that firstly the combination of FOLFIRI and trastuzumab could be tested as another regimen in metastatic gastric cancer, and, secondly, that in this disease, like in metastatic breast cancer, the continuation of trastuzumab after an initial progression under this antibody could be tested in order to improve the efficacy of the treatment Trastuzumab re-introduction with FOLFIRI for treatment of HER2 overexpression-advanced gastric adenocarcinoma following failure of other trastuzumab-based chemotherapy regimens.
