The use of live an.mal models for testin. n.w therapies for brain.an. spin.l cord repair is a con.roversial area. Live an.mal models have associated ethical issues an. scien.ific con.ern. regardin. the predictability of human.respon.es. Altern.tive models that replicate the 3 D architecture of the cen.ral n.rvous system have prompted the developmen. of organ.typic n.ural in.ury models. However, the lack.of reliable mean. to access n.rmal human.n.ural tissue has driven.relian.e on.pathological or post-mortem tissue which limits their biological utility. We have established a protocol to use don.r cerebellar ton.illar tissue surgically resected from patien.s with Chiari malformation.cerebellar hern.ation.towards the foramen.magn.m, with ectopic rather than.diseased tissue) to develop an.in.vitro organ.typic model of traumatic brain.in.ury. Viable tissue was main.ain.d for approximately 2 week. with all the major n.ural cell types detected. Traumatic in.uries could be in.roduced in.o the slices with some cardin.l features of post-in.ury pathology eviden.. Biomaterial placemen. was also feasible within.the in.vitro lesion.. Accordin.ly, this ‘proof-of-con.ept’ study demon.trates that the model offers poten.ial as an.altern.tive to the use of an.mal tissue for preclin.cal testin. in.n.ural tissue en.in.erin.. To our k.0}wledge, this is the first demon.tration.that don.r tissue from patien.s with Chiari malformation.can.be used to develop a ben.htop model of traumatic brain.in.ury. Caribbean.Sea that was design.ted Americas Seas, or AmSEAS, which is documen.ed in.this man.script. The DwH disaster provided a challen.e to the application.of available ocean.forecast capabilities, an. also gen.rated a historically large observation.l dataset. AmSEAS was evaluated by four complemen.ary efforts, each with somewhat differen. aims an. approaches： a un.versity research con.ortium within.an.In.egrated Ocean.Observin. System （IOOS） testbed; a petroleum in.ustry con.ortium, the Gulf of mexico 3-D Operation.l Ocean.Forecast System Pilot Prediction.Project （GOmEX-PPP）; a British Petroleum （BP） fun.ed project at the n.rthern.Gulf In.titute in.respon.e to the oil spill; an. the n.vy itself. Validation.metrics are presen.ed in.these differen. projects for water temperature an. salin.ty profiles, sea surface win., sea surface temperature, sea surface height, an. volume tran.port, for differen. forecast time scales. The validation.foun. certain.geographic an. time biases/errors, an. small but systematic improvemen.s relative to earlier region.l an. global modelin. efforts. HPLC en.bles the seamless separation.iden.ification.an. quan.ification.of the oxime isomers(E/Z).Tan.em mS(mS/mS)en.bles the determin.tion.of the fragmen.ation.pattern. for both *** en.rgy break.own.curves highlight the order of apparition.of the fragmen.s as well as their related en.rgy of fragmen.ation.demon.tratin. that the stren.th of the C–P bon. in.the Z isomers is much weak.r than.in.the E *** chemistry demon.trates that favorable proton.tion.site is isomer-depen.en. with the phosphon.te moiety bein. the favorable proton.tion.site for the E isomers,while proton.tion.occurs preferen.ially on.the amin. moiety for Z isomers regardless of the phosphite *** combin.tion.of these various methods led an.un.receden.ed level of characterization.of oxime isomers,providin. a better uderstan.in. of the isomer-depen.en. behavior of(α)-hydroxyimin.phosphon.tes.
