Objective To investigate the factors that influence the potential for cryoembryo implantation and multiple pregnancy. Methods In this retrospective study, a total of 93 7 thawing cycles （859 couples） in which 3286 d 3-embryos were thawed. Rates of implantation, clinical pregnancy and multiple conception following FET were observed. Results There were significant differences in female age （P〈0.05） and number of good quality embryos （P〈0. 05） between cycles that resulted in pregnancy and those did not. There was a trend toward decreasing rates of implantation, clinical pregnancy and multiple pregnancy with increasing female age. Compared with transferring 1 good quality embryo, clinical pregnancy rate of transferring 2 and 3 good quality embryos was increased significantly （P〈0. 001）, there was no significant difference in clinical pregnancy rate between transferring 2 and 3 good quality embryos. Multiple pregnancy rate was increased significantly in the group of transferring 3 good quality embryos （P〈0.05）,but there was no significant differences in multiple pregnancy rate between transferring 1 and 2 good quality embryos. Younger women （≤ 30 years） also had a significantly higher multiple pregnancy rates （28.13%） than the older ones（〉35 years）（13.64%）. With an increase in age from ≤ 30 years to〉40 years, clinical pregnancy rate declined from 47 61% to 25.00%.Conclusion Female age and the number of gooa quality embryos transferred are important factors influencing the clinical and multiple pregnancy rate, reducing the number of good quality embryos transferred may decrease the rate of multiple pregnancy but do not affect the clinical pregnancy rate.

To investigate the relationship between the expression of cyclooxygenase-2 （COX-2） and menstrual cycle, the regulatory effects of 17-β-estradiol （E2） and medroxyprogesterone acetate （MPA） on the expression of COX-2 in cervical cancer Hela cells were examined. Cervical cancer specimens were obtained from 47 pre-menopausal patients. The phase of menstrual cycle was determined by case history and HE staining of uterine endometrium. COX-2 was immunohistochemically stained by SABC staining and the staining intensity was determined with computerized image analysis system. Hela cells were incubated with alcohol, E2, E2＋MPA, MPA for 12, 24 and 48 h respectively. The expression of COX-2 in Hela cells was detected by Western blotting and reverse transcriptase-polymerase chain reaction （RT-PCR）. Our results showed that the expression of COX-2 was significantly higher during proliferative phase than secretory phase （P〈0.05）, but there was no difference in the positive rate between proliferative phase and secretory phase （P〉0.05）. Incubation with E2 could significantly enhance the expression of COX-2 continually. On the contrary, E2＋MPA and MPA alone could decrease the expression of COX-2 as compared with the control and E2 group （P〈0.05 and P〈0.01 respectively）. It is concluded that the expression of COX-2 in cervical cancer of pre-menopausal patients and Hela cells was regulated by estrogen/progestogen.

Obtaining high-quality embryos is one of the key factors to improve the clinical pregnancy rate of assisted reproductive technologies(ART).So far,the clinical evaluation of embryo quality depends on embryo ***,the clinical pregnancy rate is still ***,new indicators are needed to further improve the evaluation of embryo *** studies have shown that the decrease of sperm-specific protein actin-like 7A(ACTL7A)leaded to low fertilization rate,poor embryo development,and even *** aim of this study was to study whether the different expression levels of ACTL7A on sperm can be used as a biomarker for predicting embryo *** this study,excluding the factors of severe female infertility,a total of 281 sperm samples were collected to compare the ACTL7A expression levels of sperms with high and low effective embryo rates and analyze the correlation between protein levels and in-vitro fertilization(IVF)laboratory *** results indicated that the ACTL7A levels were significantly reduced in sperm samples presenting poor embryo ***,the protein levels showed a significant correlation with fertilization outcomes of ***7A has the potential to be a biomarker for predicting success rate of fertilization and effective embryo and the possibility of embryo *** conclusion,sperm-specific protein ACTL7A has a strong correlation with IVF laboratory outcomes and plays important roles in fertilization and embryo development.

s To study the influence of insulin on IGF Ⅰ and IGFBP Ⅰ secretion of the hum an endometrial stromal cells Methods Late proliferative phase endometrial stromal cells were isolated from endometriu m tissues and then cultured for 24 h in Hams F 12 only as a control and in Hams F 12 with different concentrations of estradiol (E2) and insulin (INS) as trea ted groups Simultaneously, the endometrial stromal cells from late secretory phase endometrium were cultured for 24 h in Hams F 12 only as a control and in Hams F 12 supplemented with different concentrations of progesterone (P) and i nsulin as treated groups After 24 h of culturing, the mediums were collected f or either IGF Ⅰ or IGFBP Ⅰ assays Result The concentrations of IGF Ⅰ in medium from cultured endometrial stromal cells i n the proliferative phase were 0 78±0 47 ng/ml in the hormone free control g roup; 1 44±0 59 ng/ml and 1 39± 0 33 ng/ml in 100 pg/ml E2 group and 20 μU /ml INS group, which was higher than that of the control group ( P <0 05 and P <0 01, respectively) The IGF Ⅰ concentration in the 100 μU/ml INS group was 2 03±0 53 ng/ml, which was higher than that of the 20 μU/ml INS group ( P <0 01) Levels of IGF Ⅰ in the 100 pg/ml E2 plus 20 μU/ml INS gro up was 2 18±0 36 ng/ml, which was significantly higher than that of the 20 μU/ml INS and 100 pg/ml E2 group ( P <0 01), but lower than that of the 100 pg/ml E2 p lus 100 μU/ml INS group (3 42±0 75 ng/ml), P <0 01 The concentration o f IGFBP Ⅰ in medium from cultured endometrial stromal cells in the secretory ph a se was 2 50±1 39 ng/ml in the hormone free control group and 5 44±2 09 ng /ml in the 10 pg/ml P group, which was significantly higher than that of the con trol ( P <0 01) IGFBP Ⅰ concentration in 20 μU/ml INS group was 0 1 6±0 58 ng/ml, which was lower compared with control, but higher compared with the 100 μU/ml INS group ( P <0 01)
