Metformin alleviates LTA-induced inflammatory response through PPARγ/MAPK/NF-κB signaling pathway in bovine mammary epithelial cells

Mastitis is a co.mon inflammatory co. mammary infection;that causes significant eco.omic loss in dairy *** the interesting co.nection between metformin’s anti-inflammatory function and mastitis model induced by LTA in pbMECs,our objective was to prove that metformin was beneficial in suppressing proinflammatory response induced by LTA through modulation of mitogen-activated protein kinase(MAPK)and nuclear factor kappa B(NF-κB)signaling pathways and activation of peroxisome proliferator-activated receptor-γ(PPARγ)in *** proliferation of cells and mRNA expression were measured using EdU assay and quantitative reverse transcriptase-polymerase chain reaction(qRT-PCR).Immunoblotting and immunofluorescence analysis were co.ducted to evaluate the expression of target proteins in inflammatory and anti-inflammatory responses to metformin and ***,pbMECs were allowed to treat with the PPAR antagonist GW9662,and inflammatory markers were detected in the *** results showed that LTA co.centration at 100μg/mL significantly stimulated the MAPK14,IL-6 and IL-1βmRNA expressions co.pared to the co.trol cells(Pco.lectively indicated that metformin act to attenuate LTA-stimulated inflammatory response in pbMECs by suppressing MAPK and NF-κB activation via a mechanism partially dependent on PPARγ*** results suggested that metformin co.ld function as an anti-inflammatory drug in the treatment of mastitis.

Porcine epidemic diarrhea(PED)caused by porcine epidemic diarrhea virus(PEDV)is one of the most devastating diseases in the global pig industry due to its high mortality rate in *** vaccines can effectively enhance the gut-mammary gland-secretory IgA axis to boost lactogenic immunity and passive protection of nursing piglets against PEDV *** 2017 to 2021,we co.lected 882 diarrhea samples from 303 farms in China to investigate the epidemiology of *** result showed that about 52.15%(158/303)of the farms were positive for PEDV with an overall detection rate of 63.95%(564/882)of the *** S1 fragments of S gene from 104 strains were sequenced for the phylogenetic analysis.A total of 71 PEDV strains(68.27%)sequenced in this study were clustered into the predominant G2c subgroup,while the newly-defined G2d strains(9.62%)were identified in three provinces of *** NH-TA2020 strain of G2c subgroup was isolated and cultured,and its infection to piglets caused watery diarrhea within 24 h,indicating its strong *** administration of NH-TA2020 strain to pregnant gilts stimulated high levels of IgA antibody in *** piglets fed by the gilts above were challenged with NH-TA2020 strain or CH-HeB-RY-2020 strain from G2d subgroup,and the clinical symptoms and virus shedding were significantly reduced co.pared to the mock *** findings suggest that G2c subgroup is the predominant branch circulating in China from 2017 to *** administration of NH-TA2020 enhances maternal IgA and lactogenic immune responses,which co.fer protection against the homologous and emerging G2d PEDV strains challenges in neonates.

Isolation and Identification of Haemophilus parasuis SD02 strain

A bacterial strain was isolated from the sick pigs suspiciously infected by polyserositis and arthritis in a pig farm in shandong Province, and identified through morphological observation, culture traits, bioehemical characteristics and PCR amplifieation. Additionally, primers were de- signed acco.ding to the 16S rRNA sequence of Haemophihzs parasuis, and the bacterial strain was amplified by PCR. The amplified fragments of approximately 1 400 bp was sequenced, and aligned with the sequence in GenBank. The results showed that it shared the homology of 97%-99% with the 16S rRNA sequence of foreign ***, and co.firmed as *** （HPS）. The strain was determined as serotype 4 through serotype identification. The strain was named SD02.

Separation and Preliminary Identification of Intestinal Probiotics of Laying Hens

[Objective] The paper was to isolate and identify probiotics in the intestine of laying hens. [Method] The intestinal probiotics in laying hens at peak period were isolated using co.ventional separation methods; the physical and chemical properties of target strains and in vitro antibacterial effects were measured. Moreover, the safety test of chicks was co.ducted. [Result] Four strains of lactobacillus were isolated from the jejunum of laying hens, including Lactobacillus plantarum, Bacillus acidophilus, L. delbrueckii subsp. Delbrueckii and L. delbrueckii subsp. Lactis, and their inhibition zone diameters were 18.30, 16.07, 11.45, 17.26 mm, respectively. One strain of Lactobacillus, L. brevis, was isolated from the cecum, with the inhibition zone diameter of 10.26 mm. Three strains of bacillus were isolated from the cecum, including Bacillus subtilis, B. cereus and B. licheniformis, and their inhibition zone diameters were 9.25, 8.46 and 8.37 mm, respectively. Daily drinking 2 billion units of viable bacteria was the safe dosage for chicks. [co.clusion] Eight strains of probiotics had certain inhibitory effect on Escherichia co.i O;, and had no toxic side effects to chicks. Lactobacillus had strong antibacterial effect on E. co.i O;, while the antibacterial effect of bacillus was relatively weak.

The paper was to study the inhibitory effect of reco.binant freeze-drying chicken interferon against Newcastle disease virus （NDV） F48E10. Nine-day-old chicken embryos were inoculated with reco.binant freeze-drying chicken interferon via allaotoic sack, while 10-day-old chicken embryos were inoculated with NDV F48E10, and in vivo protective efficacy of interferon on chichen embryos was studied. The results showed that the reco.binant freeze-drying chicken interferon at the dose of 1.28 mg/embryo reached the protection ratio of 90% on chicken embryo infected by F48E10.

Determination of Berberine Hydrochloride co.tent in Qingwen Baidu Granule by HPLC

The paper was to establish a simple and sensitive HPLC method for determination of berbefine hydrochloride co.tent in Qingwen Baidu granule. The HPLC system co.sisted of Stable Bond - C18 co.umn （4.6 × 150 mm, 5um） and with a mobile phase of acetonitrile -0.05mol/L potassium dihydrogen phosphate solution （50：50,v：v） mixture. Berberine hydrochloride was detected at the wavelength of 345 nm, with a flow rate of 1.0 ml/min and a co.umn temperature of 30 ℃. The excipients and solvents in the granule co.ld be well separated from the drug under such a designated chromatogram co.dition and did not interfere with the assay. A good linear relationship was found between peak area and the co.centration of berberine hydrochloride in the range of 16.0 -48.0 I^g/mL. The average reco.ery of berberine hydrochloride was 99.09%. The established approach was specific, accurate, reliable, prompt, sensitive and applicable, and co.ld be used to co.trol the quality of Qingwen Baidu granule.

With the long -term and irrational use of veterinary drugs and drug additives in the process of animal feeding, the veterinary drug resi- dues in animals and their additives can be ingested into the human body and pose potential threat to human health. Detection of veterinary drug resi- dues are extremely important practical significance to protect the eco.ogical environment and human health. In recent years, liquid chromatography -tandem mass spectrometry has been widely used in vetednary drug residue detection because of its applicability, qualitative and quantitative fea- tures as well as high sensitivity. This article summarized the application of liquid chromatography -tandem mass spectrometry in detection veterinary drug residues from feed, animal products, arid udne as well as illicit drugs and harmful additives.

Simultaneous Determination of Sulfamonomethoxine Sodium and Trimethoprim in co.pound Sulfamonomethoxine Sodium Propolis Solution by High Performance Liquid Chromatography(HPLC)

[Objective] The paper aimed to provide a basis for quality co.trol of co.pound sulfamonomethoxine sodium propolis solution. [Method] High per- formance liquid chromatography （HPLC） method was used to determine the co.tents of sulfamonomethoxine sodium and trimethoprim simultaneously in co.pound sulfamonomethoxine sodium propolis solution. [Result] Sulfamonomethoxine sodium and trimethoprim showed a linear relationship within the co.centration of 10 - 500 μg/mL under the following chromatographic co.ditions： Kromasil C18 co.umn （250 mm × 4.6 mm, 5 μm）, mobile phase 0.02 mol/L phosphoric acid solution- methanol（80：20, V/V）, detection wavelength 270 nm, flow rate 1.0 mL/min, and co.umn temperature 30℃. The average reco.eries were 99.3% and 99.4%, respectively. [ co.clusion] The method is accurate and feasible, and can determine sulfamonomethoxine sodium and trimethoprim simultaneously in co.pound sul- famonomethoxine sodium propolis solution.

Phenotypic and Genetic Characteristics of a Bordetella hinzii Isolated from a Rex Rabbit

A Bordetella hinzii strain, isolated from the lung of a dead breast-feeding infant rex rabbit with a respiratory infection, was used as study object to investigate its phenotypic and genetic characteristics. The strain co.ld grow on Macco.key agar and was gram-negative, oxidase-positive, catalase-positive, short rodshaped, and it can produce alkali from malonate. Its 6S rRNA sequence was partially identical with B. hinzii （AF177667）. Animal experiments revealed nasal muco.s hyperemia, tracheal co.gestion and bronchopneumonia in 28-day old rex rabbits. To sum up, this Bordetella hinzii isolate was deemed as a novel causative agent of respiratory disease in rex rabbit.

Pathogenicity of Escherichia co.i O_(123) from Rex Rabbit on White Mice

[Objective]The paper was to analyze the pathogenesis of Escherichia co.i O123 from rex rabbit. [Method]E. co.i O123 isolated from rabbit liver with diarrhea symptom in scale rex rabbit farm was intraperitoneally injected into 18- 22 g Kunming mice,and its pathogenicity was determined by clinical symptoms and pathological examination. [Result]When the inoculation co.centration was about 8. 5 × 107 CFU /mL,Kunming mice appeared the clinical symptoms of drooping spirit,diarrhea and gathering,and the mortality reached 50%. Anatomical examination found that intestinal wall was thinning and intestinal muco.a was bleeding. [co.clusion]E. co.i from rex rabbit has strong pathogenicity,and establishing animal model with Kunming mice to study its pathogenesis is of great reference significance for diagnosis and prevention of E. co.i disease of rex rabbit.
