LiF@SiO2 nanocapsules for controlled lithium release and osteoarthritis treatment

Electrolytes can be taken orally or intravenously as supplements or thera- peutics. However, their therapeutic window may exceed the serum toxicity threshold, making systemic delivery a poor option. Local injection is also not adequate due to rapid diffusion of electrolytes. Here, we solved this issue with a nanocapsule technolog~ comprising an electrolyte nanocrystal as the drug filling and a silica sheath to regulate drug release rates. In particular, we prepared LiF@SiO2 nanocapsules and investigated their potential as a delivery system for lithium, which was shown in recent studies to be an effective therapeutic agent for osteoarthritis （OA）. We demonstrated that LiF@SiO2 can extend lithium release time from minutes to more than 60 h. After intra- articular （i.a.） injection into a rat OA model, the nanocapsules reduced the Osteoarthritis Research Society International （OARSI） score by 71% in 8 weeks while inducing no systemic toxicity. Our study opens new doors for improved delivery of electrolyte therapeutics, which have rarely been studied in the past.

Efficacy and safety of once daily tacrolimus compared to twice daily tacrolimus after liver transplantation

BACKGROUND Once daily tacrolimus regimen was found to exhibits similar bioavailability,safety and efficacy properties compared to twice-daily tacrolimus in kidney transplantation *** To compare the efficacy and safety of once-daily prolonged release tacrolimus compared to twice-daily tacrolimus in liver transplantation *** MEDLINE,EMBASE,CENTRAL databases were searched for clinical trials until December *** outcome measured as the rate of treatment failure indicated by biopsy-proven acute rejection,Serum creatinine,graft loss,or *** reviewers independently selected studies,collected data and assessed risk of *** results are reported as risk ratio with 95%confidence interval(CI)for dichotomous *** Seven studies included with 965 *** the included studies were of moderate quality according to the risk of bias assessment using Cochrane Risk of Bias ***-proven acute rejection was reported in four studies,and pooled analysis of those studies indicated similar rejections in both twice daily and once daily tacrolimus groups(risk ratio:1.06,95%CI:0.84-1.34,n=758,I2=0%)and also we found no significant difference between both groups for renal outcome(serum creatinine;mean difference,0.001 mg/dL,95%CI:-0.042 to 0.043,n=846,I2=18.6%).Similarly,there was similar number of adverse events such as hypertension,headache,back pain,blood related disorders,infections and nausea observed in both *** The analysis findings confirm that both once daily and twice daily tacrolimus formulations are comparable in terms of efficacy and safety outcomes.

The traditional medicinal plant,and endangered species Aristolochia delavayi(Aristolochiaceae)is an endemic species in China and occurs in the warm and dry areas along the Jinsha *** is also a specific host of the larvae of Byasa daemonius,a vulnerable *** this study,15 pairs of polymorphic microsatellite primers of *** were designed and screened based on the Simple Sequence Repeats(SSR)loci found by using the results of genome *** on these 15 SSR markers,the genetic diversity and structure of 193 individuals from ten natural populations were analyzed in *** comparison to other endemic and endangered plants in the region,the population of *** possess a relatively high genetic diversity(He=0.550,I=1.112).AMOVA analysis showed that 68.4%of the total genetic diversity was within populations and 31.6%of the variation occurred among *** was a significant genetic differentiation among natural populations of *** detectable,with low gene flow(Nm=0.591).This might be attributed to geographical barriers and limited seed *** test the isolation by distance(IBD),we performed Mantel test,which showed a significant correlation between the geographic and genetic *** order to cope with the possible biases caused by IBD,we additionally performed Bayesian genetic cluster analyses and principal coordinate analysis(PCoA).The final cluster analysis revealed three groups with distinct geographical *** fragmentation and limited gene flow between these populations may be the main reasons for the current genetic *** conservation of this species,we suggest to divide its populations into three protection management units,with subsequent focus on the Yongsheng and Luquan populations which experienced a genetic bottleneck event in the past.

Sucrose phosphate synthase(SPS)is a rate-limiting enzyme that works in conjunction with sucrose-6-phosphate phosphatase(SPP)for sucrose synthesis,and it plays an essential role in energy provisioning during growth and development in plants as well as improving fruit ***,studies on the systematic analysis and evolutionary pattern of the SPS gene family in apple are still *** the present study,a total of seven MdSPS and four MdSPP genes were identified from the Malus domestica genome GDDH13 *** gene structures and their promoter cis-elements,protein conserved motifs,subcellular localizations,physiological functions and biochemical properties were analyzed.A chromosomal location and gene-duplication analysis demonstrated that whole-genome duplication(WGD)and segmental duplication played vital roles in MdSPS gene family *** Ka/Ks ratio of pairwise MdSPS genes indicated that the members of this family have undergone strong purifying selection during ***,three SPS gene subfamilies were classified based on phylogenetic relationships,and old gene duplications and significantly divergent evolutionary rates were observed among the SPS gene *** addition,a major gene related to sucrose accumulation(MdSPSA2.3)was identified according to the highly consistent trends in the changes of its expression in four apple varieties(‘Golden Delicious’,‘Fuji’,‘Qinguan’and‘Honeycrisp’)and the correlation between gene expression and soluble sugar content during fruit ***,the virus-induced silencing of MdSPSA2.3 confirmed its function in sucrose accumulation in apple *** present study lays a theoretical foundation for better clarifying the biological functions of the MdSPS genes during apple fruit development.

Cystathionine-γ-lyase(CSE),an enzyme associated with hydrogen sulfide(H2S)production,is an important endogenous regulator of *** domain-containing protein 3(JMJD3)is implicated in the immune response and ***,we investigated the potential contribution of JMJD3 to endogenous CSE-mediated inflammation in rheumatoid arthritis(RA).Upregulated CSE and JMJD3 were identified in synovial fibroblasts(SFs)from RA patients as well as in the joints of arthritic *** down CSE augmented inflammation in IL-1β-induced SFs by increasing JMJD3 *** addition,CSE−/−mice with collagen-induced arthritis(CIA)developed severe joint inflammation and bone ***,overexpressing CSE inhibited JMJD3 expression by the transcription factor Sp-1 and was accompanied by reduced inflammation in IL-1β-treated ***,JMJD3 silencing or the administration of the JMJD3 inhibitor GSK-J4 significantly decreased the inflammatory response in IL-1β-treated SFs,mainly by controlling the methylation status of H3K27me3 at the promoter of its target ***-J4 markedly attenuated the severity of arthritis in CIA *** conclusion,suppressing JMJD3 expression by the transcription factor Sp-1 is likely responsible for the ability of CSE to negatively modulate the inflammatory response and reduce the progression of RA.

L-Palmitoylcarnitine(L-PC)is an important endogenous fatty acid *** classical biological functions are involved in the regulations of membrane molecular dynamics and theβ-oxidation of fatty *** plasma long-chain acylcarnitines showed the association of venous thrombosis,implying anticoagulant activity of the metabolites and inspiring us to investigate if and how L-PC,a long-chain acylcarnitine,takes part in *** we show that L-PC exerted anti-coagulant effects by potentiating the enzymatic activities of plasmin and tissue plasminogen activator(tPA).L-PC directly interacts with plasmin and tPA with an equilibrium dissociation constant(KD)of 6.47×10^(-9)and 4.46×10^(-9)M,respectively,showing high *** mouse model,L-PC administration significantly inhibited FeCl_(3)-induced arterial *** also mitigated intracerebral thrombosis and inflammation in a transient middle cerebral artery occlusion(tMCAO)mouse model.L-PC induced little bleeding *** results show that L-PC has anti-thrombotic function by potentiating plasmin and tPA.

Bioactive glasses(BGs)are promising bone substitute ***,under certain circumstances BGs such as the well-known 45S5 Bioglass®(composition in wt%:45.0 SiO2,24.5 Na2O,24.5 CaO,6.0 P2O5)act cytotoxic due to a strong increase in pH caused by a burst release of sodium ions.A potential alternative is a sodiumreduced fluoride-containing BG belonging to the CaO–MgO–SiO2 system,namely BG1d-BG(composition in wt%:46.1 SiO2,28.7 CaO,8.8 MgO,6.2 P2O5,5.7 CaF2,4.5 Na2O),that has already been evaluated in-vitro,in-vivo and in preliminary clinical *** further application,however,BG1d-BG should be compared to the benchmark amongst BGs,the 45S5 Bioglass®composition,to classify its effect on cell viability,proliferation and osteogenic differentiation of human mesenchymal stem cells(MSCs).Therefore,in this study,the biocompatibility and osteogenic potential of both BGs were investigated in an indirect and direct culture setting to assess the effect of the ionic dissolution products and the BGs’physical presence on the *** results indicated an advantage of BG1d-BG over 45S5 Bioglass®regarding cell viability and *** BGs induced an earlier onset of osteogenic differentiation and accelerated the expression of late osteoblast marker genes compared to the control *** conclusion,BG1d-BG is an attractive candidate for further experimental *** basic mechanisms behind the different impact on cell behavior should be assessed in further detail,*** further alteration of the BG compositions.

Level of Bruising and DFD Carcasses from Bulls of Arsi, Boran and Harar Cattle Breeds in Ethiopia

The objective of the study was to determine the level of Bruising and Dark Firm Dry on Arsi, Boran and Harar cattle breeds in Ethiopia. A total of 39 beef bulls from three different breeds (14 Boran, 14 Harar, and 11 Arsi cattle breeds) were used in this study. Based on the present finding, from 39 slaughtered beef bulls 66.7% was bruised and 30.77% produced DFD meat. Harar breed had the highest (39.1%) overall level of bruising while Boran and Arsi had the lowest level. However, Harar breed had the lowest level of DFD (14.29%). Age groups 3 - 6 and 6 - 9 years had the highest level of a bruise (29.70% and 28.10%, respectively) compared to age groups 2 - 3 years. The mean number of bruises per carcass was 1.96 ± 0.25 while the bruise score per carcass was 3.58 ± 0.68. A linear bruise was the most observed (33.33%) shape of a bruise. It was concluded that the level of bruising and DFD was higher compared to some other abattoir studies in other countries which indicated the possible presence of poor handling practice of cattle from farm to abattoir gates. To minimize the incidence of DFD and bruising, it was recommended that proper handling of cattle during loading, transport, unloading, at lairriage and during slaughter need to be practiced.

Non-conjugated terpolymer acceptors for highly efficient and stable lager-area all-polymer solar cells

All-polymer solar cells(all-PSCs)have made significant progress recently,but few studies have been conducted to investigate the lab-to-manufacturing translation from the spin-coating method to the printing ***,the random copolymerization method and non-conjugated backbone approach are integrated to manipulate the morphology and photoelectric properties of the active layer for large-area printed all-PSCs.A series of non-conjugated terpolymer acceptors PYSe-TC_(6)T(x)(x=5,10,and 20,refers to the molar ratio of TC_(6)T unit)are developed by covalently introducing non-conjugated unit TC_(6)T into the PYSe host bipolymer by random *** spin-coated PYSe-TC_(6)T(10)-based all-PSC demonstrates the best power conversion efficiency(PCE)of 13.54%,superior to the PYSe-based one(12.45%).More intriguingly,morphological studies reveal that a combination of the random polymerization and non-conjugated backbone strategy can effectively prevent the active layer from overaggregation and improve the film quality during the printing process,thereby minimizing the efficiency and technology gap between spin-coated small-area devices and blade-coated large-area *** directly using the same preparation condition of spin-coating,the blade-coated small-area(0.04 cm^(2))delivers a PCE of 12.83%and the large-area(1.21 cm^(2))device achieves a PCE of 11.96%with a small PCE *** PCE value and PCE loss are one of the most outstanding performances of the bladecoated *** findings reveal that a combination of the non-conjugated flexible backbone with random copolymerization to develop non-conjugated terpolymers is an attractive design concept to smoothly realize the lab-to-manufacturing translation.
