Hepcidin expression in colon during trinitrobenzene sulfonic acid-induced colitis in rats

AIM:To investigate hepcidin expression,interleukin-6(IL-6)production and iron levels in the rat colon in the presence of trinitrobenzene sulfonic acid(TNBS)-induced ***:In rats,we evaluated the severity of colitis induced by repeated TNBS administration using macroscopic and microscopic scoring systems and myeloperoxidase activity *** colonic levels of hepcidin,tumor necrosis factor alpha(TNF-α),IL-10 and IL-6 were measured by Enzyme-Linked Immunosorbent Assay,and hepcidin-25 expression and iron deposition were analyzed by immunohistochemistry and the Prussian blue reaction,***-3 phosphorylation was assessed by Western blot *** parameters,iron and transferrin levels,and transferrin saturation were also ***,the ability of iron,pathogen-derived molecules and IL-6 to induce hepcidin expression in HT-29 cells was ***:Repeated TNBS administration to rats resulted in macroscopically and microscopically detectable colon lesions and elevated colonic myeloperoxidase ***-25 protein levels were increased in colonic surface epithelia in colitic rats(10.2±4.0pg/mg protein vs 71.0±8.4 pg/mg protein,P<0.01).Elevated IL-6 levels(8.2±1.7 pg/mg protein vs 14.7±0.7 pg/mg protein,P<0.05),TNF-αlevels(1.8±1.2pg/mg protein vs 7.4±2.1 pg/mg protein,P<0.05)and Stat-3 phosphorylation were also *** alterations in iron homeostasis,hepcidin levels and anemia were not detected in colitic *** deposition in the colon was only observed during *** gene expression was increased in HT-29 cells after IL-6 and lipopolysaccharide[a toll-like receptor 4(TLR-4)ligand]***,ferric citrate and peptidoglycan(a TLR-2 ligand)were unable to alter the in vitro expression of hepcidin in HT-29 ***:Colitis increased local hepcidin-25 expression,which was associated with the IL-6/Stat-3 signaling *** increase in local iron sequestration was also

Human Beta Defensins 1,2 and 3 Produced by Amniochorion Membranes Is Similar in Term and Preterm Delivery

Amniochorion membranes were collected from 25 pregnant women at preterm labor, in the presence or not of Preterm Premature Rupture of Membranes (PPROM) and 27 pregnant women at term in the presence at labor, in order to quantify the expression and to evaluate the immunoreactivity of human beta defensins (HBD)1, HBD2, HBD3 and HBD4 in amniochorion membranes from pregnancies complicated by spontaneous prematurity. The HBDs were evaluated by immunohistochemistry, real time quantitative PCR and ELISA. Statistical analyses were performed using Chi-squared and Mann Whitney tests. There was no significant difference in HBDs expression between study and control groups: HBD1 (Md = 0.62 (0.0 - 105.0) vs Md = 0.80 (0.02 - 25.0);p = 0.85), HBD2 (Md = 0.17 (0.0 - 5.2) vs Md = 0.0 (0.0 - 43.2);p = 0.16), HBD3 (Md = 0.11 (0.0 - 140.5) vs Md = 0.06 (0.0 - 972.1);p = 0.91). Also, HBD1, HBD2 and HBD3 protein expression was not significant different between the groups: HBD1 (1.32 pg/mL (0.0 - 1.85) vs 1.08 pg/mL (0.04 - 2.22);p = 0.67), HBD2 (0.00 pg/mL (0.0 - 1.74) vs 0.02 pg/mL (0.0 - 1.24);p = 0.69), HBD3 (0.04 pg/mL (0.0 - 1.05) vs 0.09 pg/mL (0.0 - 1.05);p = 0.63). The immunoreactivity of HBD1, HBD2 and HBD3 was observed in amnion, chorion and decidua cells from preterm and term pregnancies. Amniochorion membranes are sources of HBD1, HBD2 and HBD3 and their expressions are similar in term and preterm pregnancies.

Nutritional assessment in cirrhotic patients with hepatic encephalopathy

Hepatic encephalopathy(HE) is one of the worst complications of liver disease and can be greatly influenced by nutritional status. Ammonia metabolism, inflammation and muscle wasting are relevant processes in HE pathophysiology. Malnutrition worsens the prognosis in HE, requiring early assessment of nutritional status of these patients. Body composition changes induced by liver disease and limitations superimposed by HE hamper the proper accomplishment of exams in this population, but evidence is growing that assessment of muscle mass and muscle function is mandatory due to the role of skeletal muscles in ammonia metabolism. In this review, we present the pathophysiological aspects involved in HE to support further discussion about advantages and drawbacks of some methods for evaluating the nutritional status of cirrhotic patients with HE, focusing on body composition.

The Archean Eon was a time of geodynamic *** evidence of these transitions come from igneous/metaigneous rocks,which dominate cratonic segments *** data for granitoids from an Archean basement inlier related to the Southern S?o Francisco Craton(SSFC),are integrated with geochronological,isotopic and geochemical data on Archean granitoids from the *** rocks are divided into three main geochemical groups with different ages:(1)TTG(3.02–2.77 Ga);(2)mediumto high-K granitoids(2.85–2.72 Ga);and(3)A-type granites(2.7–2.6 Ga).The juvenile to chondritic(Hf-Nd isotopes)TTG were divided into two sub-groups,TTG 1(low-HREE)and 2(high-HREE),derived from partial melting of metamafic rocks similar to those from adjacent greenstone *** compositional diversity within the TTG is attributed to different pressures during partial melting,supported by a positive correlation of Dy/Yb and Sr/Zr,and batch melting *** proposed TTG sources are geochemically similar to basaltic rocks from modern island-arcs,indicating the presence of subduction processes concomitant with TTG ***~2.85 Ga to 2.70 Ga,the dominant rocks were K-rich *** are modeled as crustal melts of TTG,during regional metamorphism indicative of crustal *** compositional diversity is linked to:(i)differences in source composition;(ii)distinct melt fractions during partial melting;and(iii)different residual mineralogies reflecting varying P–T ***-collisional(~2.7–2.6 Ga)A-type granites reflect rifting in that they were closely followed by extension-related dyke swarms,and they are interpreted as differentiation or partial melting products of magmas derived from subduction-modified *** sequence of granitoid emplacement indicates subduction-related magmatism was followed by crustal thickening,regional metamorphism and crustal melting,and post-collisional extension,similar to that seen in younger Wilson *** is compelling evide

Hepatic encephalopathy(HE)is a cognitive disturbance characterized by neuropsychiatric *** occurs in acute and chronic hepatic disease and also in patients with portosystemic *** presence of these portosystemic shunts allows the passage of nitrogenous substances from the intestines through systemic veins without liver ***,the embolization of these shunts has been performed tocontrol HE manifestations,but the presence of portal vein thrombosis is considered a *** this presentation we show a cirrhotic patient with severe HE and portal vein thrombosis who was submitted to embolization of a large portosystemic *** report:a 57 years-old cirrhotic patient who had been hospitalized many times for persistent HE and hepatic coma,even without precipitant *** had a wide portosystemic shunt and also portal vein *** abdominal angiography confirmed the splenorenal shunt and showed other *** larger shunt was embolized through placement of microcoils,and the patient had no recurrence of overt *** was a little increase of esophageal and gastric varices,but no endoscopic treatment was *** portosystemic shunts are frequent causes of recurrent HE in cirrhotic patients,portal vein thrombosis should be considered a relative contraindication to perform a shunt ***,in particular cases with many shunts and severe HE,we found that one of these shunts can be safely embolized and this procedure can be sufficient to obtain a good HE *** conclusion,we reported a case of persistent HE due to a wide portosystemic shunt associated with portal vein *** the patient had other shunts,she was successfully treated by embolization of the larger shunt.

Sexual Diversity and Homophobia in Health Care Services: Perceptions of Homosexual and Bisexual Population in the Cross-Cultural Theory

The provision of care by health professionals seems to be related to cultural patterns in which these workers are inserted. The relationship between culture and care becomes a challenge for the provision of health care to homosexual and bisexual population. Thus, this study aimed to identify the perception of Gays, Lesbians, Bisexuals and Transgenders in relation to the care received from health professionals, in the light of Cross-Cultural Theory. This is a qualitative, ethnographic study, conducted with 30 members of sexual minorities in Juazeiro do Norte, Brazil. The data analysis lead to the elaboration of five categories based on of the Theory of Cultural Care Diversity and Universality. The participants emphasized the constant violation of their rights to universal and equitable access to the National Health System, stating that they have suffered from prejudice and exclusion, as result of a heteronormative culture. They highlighted that homophobia and dehumanization of care have a strong relation to sexual orientation. The transvestites are the most prone to humiliation and pejorative attitudes of professionals and such condition is a barrier to accessing services.

Assessment of fetus during second trimester ultrasonography using HDlive software:What is its real application in the obstetrics clinical practice?

AIM To show imaging results from application of fourdimensional(4D) ultrasound lightening technique(HDlive^(TM)) in clinical obstetrics practice. METHODS Normal and abnormal fetuses at second and third trimester of pregnancy undergoing routine scan with 4D HDlive^(TM)(5DUS) in the rendering mode are described. Realistic features of fetal structures were provided by 5DUS in the rendering mode. Normal anatomy as well as pathology like cleft lip, hypoplastic face, micrognathia, low-set ears, corpus callosum, arthrogryposis, aortic arch, left congenital diaphragmatic hernia are highlighted in this study. Anatomical details of the fetuses were provided by 5DUS with higher quality imaging modality compared to those obtained using conventional 2D/3D ultrasound. RESULTS Realistic views of fetal anatomy details were displayed by means of 5DUS in the rendering mode, with high image quality obtained either in low-risk or in high-risk obstetrics population. Corpus callosum, esophagus, and aortic arch were obtained in normal fetuses. Cleft lip, cleft lip and palate, micrognathia, hypoplastic face, lowset ears, arthrogryposis, left congenital diaphragmatic hernia, exomphalos, and clitoris hypertrophy were clearly rendered by 5DUS *** The use of 5DUS in the rendering mode, when clinical available, was diagnostic in a variety of congenital anomalies, aided understanding of the parents-tobe and improved prenatal counseling and perinatal management.

BACKGROUND Coronavirus disease 2019(COVID-19)pandemic is still evolving globally,and Brazil is currently one of the most affected *** is still debated whether patients with inflammatory bowel disease(IBD)are at a higher risk for developing COVID-19 or its *** To assess geographical distribution of IBD patients at the highest risk and correlate these data with COVID-19 mortality rates in *** The Brazilian IBD Study Group(Grupo de Estudos da Doença Inflamatória Intestinal do Brasil)developed a web-based survey adapted from the British Society of Gastroenterology *** included categories were demographic data and inquiries related to risk factors for complications from *** were categorized as highest,moderate or lowest individual *** Spearman correlation test was used to identify any association between highest risk and mortality rates for each state of the *** A total of 3568 patients(65.3%females)were *** participants were from the southeastern and southern regions of Brazil,and 84.1%were using immunomodulators and/or *** patients(55.1%)were at moderate risk,23.4%were at highest risk and 21.5%were at lowest risk of COVID-19 *** association between the proportion of IBD patients at highest risk for COVID-19 complications and higher mortality rates was identified in different Brazilian states(r=0.146,P=0.467).CONCLUSION This study indicates a distinct geographical distribution of IBD patients at highest risk for COVID-19 complications in different states of the country,which may reflect contrasting socioeconomic,educational and healthcare *** association between high risk of IBD and COVID-related mortality rates was identified.
