Enhanced antibacterial activity of silica nanorattles with ZnO combination nanoparticles against methicillin-resistant Staphylococcus aureus

Silica nanorattles(SNs) with zinc oxide(ZnO) combination nanoparticles are reported to inhibit methicillin-resistant Staphylococcus aureus(MRSA) for the first time. SNs loaded with ZnO nanoparticles,which can produce free radicals, can cause severe damage to bacteria. ZnO nanoparticles not only provide free radicals in the combined nanostructures, which can inhibit the growth of bacteria, but also form nanorough surfaces with an irregular distribution of spikes on the SNs, which can enhance their adhesion to bacteria. Nanorough silica shell surfaces maintain the high activity and stability of small-sized ZnO nanoparticles and gather ZnO nanoparticles together to enhance production, which improves the efficiency of free radicals against the cytomembranes of bacterial cells. The enhanced adhesion of ZnO@SN nanoparticles to MRSA cells shortens the effective touching distance between free radicals and MRSA, which also improves antibacterial activity. As we expected, the ZnO@SN nanoparticles exhibit a better antibacterial effect than free ZnO nanoparticles against MRSA in vitro and in vivo. We also demonstrate that SNs loaded with ZnO nanoparticles can accelerate wound healing in MRSA skin inflammation models. This method of multilevel functionalization will be potentially applicable to the antibacterial field.

Silica nanorattle with hollow and mesoporous structure has been proven to be an excellent drug carrier.However,how to control the drug release from silica nanorattle is still a challenge.In this study,we designed two methods,in-situ polymerization method and water in oil in water(W/O/W) double emulsion method,to coat a nanosized poly(lactic-co-glycolic acid)(PLGA) layer onto the surface of silica nanorattle for controlling the drug release behavior.Hydrophobic antitumor drug docetaxel was loaded into the PLGA coated silica nanorattle(PLGA@SN).The drug release profile,cellular uptake and cytotoxicity on human liver cancer HepG2 cells were evaluated to prove that the PLGA layer plays an effective role in tuning the drug delivery.

ZrMOF@CdTe nanoparticles(NPs)with high fluorescence were synthe-sized by hydrothermal method.The morphology,particle size distribution,compositions,fluorescence properties and stability of the synthesized ZrMOF@CdTe were analyzed via the characterization by TEM,ICP-AES and fluorescence spectrophotometry,and the effects of the reaction time and pH value on the fluorescent property of ZrMOF@CdTe NPs were discussed.The results show that ZrMOFs could maintain its morphology and structure well during the process of loading CdTe quantum dots.With the increase of the loading reaction time,the red-shifted emission peaks of ZrMOF@CdTe NPs appear,and their fluorescence gradually changes from green to red color.With the increase of the pH value and temperature of the hydrothermal reaction,the fluorescence of ZrMOF@CdTe NPs was also consistent with the red-shifted change.The fluorescent property of ZrMOF@CdTe NPs could be remained for more than 3 months.Therefore,ZrMOF@CdTe NPs synthesized by the hydrothermal method have the characteristics of simple operation,adjustable fluorescent color and high stability,and the potential application in the fields of biological detection and sensing is expected.

With the rapid development of nanotechnology,nano-drug carrier systems in biomedicine have received extensive attention from researchers.By adsorbing or coating the drug in/on a nano-sized carrier,this system improves the drug’s stability and selectivity in the body,reducing the toxicity and side effects of the drug.Thus these carrier systems have broad application prospects in the field of biomedicine.A common nano-drug carrier system includes inorganic nano-carriers,organic nano-carriers,and metal nano-carriers.Inorganic nano-carriers have the advantages of good stability,controllable size,larger specific surface area,and unique photothermal electromagnetic properties.

Despite being a common therapy for hepatocellular carcinoma(HCC),insufficient thermal ablation can leave behind tumor residues that can cause recurrence.This is believed to augment M2 inflammatory macrophages that usually play a pro-tumorigenic role.To address this problem,we designed D-mannose-chelated iron oxide nanoparticles(man-IONPs) to polarize M2-like macrophages into the antitumor Ml phenotype.In vitro and in vivo experiments demonstrated that man-IONPs specifically targeted M2-like macrophages and accumulated in peri-ablation zones after macrophage infiltration was augmented under insufficient microwave ablation(MWA).The nanoparticles simultaneously induced polarization of pro-tumorigenic M2 macrophages into antitumor M1 phenotypes,enabling the transformation of the immunosuppressive microenvironment into an immunoactivating one.Post-MWA macrophage polarization exerted robust inhibitory effects on HCC progression in a well-established orthotopic liver cancer mouse model.Thus,combining thermal ablation with man-IONPs can salvage residual tumors after insufficient MWA.These results have strong potential for clinical translation.

Metal-organic frameworks(MOFs)as a class of porous functional materials have attracted more and more attention for biomedical applications.To date,MOFs have been developed for bioimaging based on a series of advantages such as the large surface areas,high porosity,fluorescence functionalities and good biocompatibility.It is worth noting that organic or inorganic fluorescent materials such as fluorescent dyes,quantum dots,metal nanoclusters and nanosheets can combine MOFs or be encapsulated in MOFs to form fluorescent nanocomposites for excellent imaging function.Importantly,excellent imaging capabilities are of great significance in living cells and in vivo for detection,diagnosis and cancer therapy.In this review,we focus on the recent research of the bioimaging in living cells and in vivo based on various MOF-based nanocomposites and their potential biological clinical applications.

Although surgical resection and chemotherapy were widely applied in tumor therapy, the dysfunction of normal cells resulted in the side effects (such as anorexia, nausea). MW thermal therapy is a non-invasive anticancer strategy under the help of MW sensitizer, with the safety and higher efficacy. Zirconium metal-organic framework nanocubes (ZrMOF NCs) modified with polyethylene glycol, were prepared via one pot method and carbodiimide technique, resulting in their large specific surface area and porosity.Our results showed that non-ion-loaded ZrMOF NCs in 0.9% NaCl solution exhibited better heating effect,higher than that in pure water, due to the robust collision among the ions under MW irradiation. The in vivo experiments confirmed that ZrMOF-PEG NCs + MW group exhibited the higher temperature in the tumor region than that of only MW treatment, suggesting a better MW thermal therapeutic anticancer efficacy. This work provides a new preparation strategy of biosafety nanomaterial as MW sensitizer for enhancing MW thermal anticancer therapy.

Metal ions are physiologically essential,but excessive metal ions may cause severe risk to plants and animals.Here,we prepared gold nanoclusters（Au NCs） protected by 11-mercaptoundecanoic acid（11-MUA）,which have excellent fluorescence properties for the detection of metal ions.The results showed that the copper ions（Cu^（2＋）） and iron ions（Fe^（3＋）） in the solution have obvious quenching effect on the fluorescence intensity of Au NCs.The detection range of Fe^（3＋） was 0.8-4.5 mmol/L（R^2= 0.992） and 4.5-11.0 mmol/L（R^2= 0.997）.And Cu^（2＋） has a lower linear range（0.1-1.0 mmol/L,R2= 0.993）.When EDTA was added into the reaction system,it was observed that the quenching effect of Cu^（2＋） and Fe^（3＋）on Au NCs showed different phenomenon.Then,the effect of metal ions on the fluorescence of Au NCs was investigated.The selective detection of Cu（2＋） was achieved by EDTA masking of Fe^（3＋）.In addition,we realized the metal ions detection application of Au NCs in the serum

Hollow mesoporous nanomaterial is a kind of promising new drug delivery system due to their unique hollow structures.In order to evaluate the toxicity of silica nanorattle (SN) particles in vivo,40 female mice were used in this study to investigate the acute toxicity and oxidative damage.Mice were intravenously injected with SN suspension in sterile 5% glucose at 40,80 and 240 mg/kg,respectively.The control group was administrated with equal-volume 5% glucose.Weight,feed intake,hematology analysis,blood biochemical assay and histopathology diagnosis were examined.The activities of SOD,GSH and CAT were measured as well.The results demonstrated that the levels of ALT and AST in the mice treated with 240 mg/kg SN increased significantly as compared with the control group (P<0.05),whereas the contents of BUN and CREA changed unremarkably.The activity of SOD induced by SN in the liver decreased significantly (P<0.05).In summary,this study revealed that liver was the target organ of the SN.It also can be concluded that activity of SOD played an important role in liver injury caused by SN.
