The sensing of hot and cold stimuli by dental neurons differs in several fundamental ways. These sensations have been characterized quantitatively through the measured time course of neural discharge signals that result from hot or cold stimuli applied to the teeth of animal models. Although various hypotheses have been proposed to explain the underlying mechanism, the ability to test competing hypotheses against experimental recorded data using biophysical models has been hindered by limitations in our understanding of the specific ion channels involved in nociception of dental neurons. Here we apply recent advances in established biophysical models to test the competing hypotheses. We show that a sharp shooting pain sensation experienced shortly following cold stimulation cannot be attributed to the activation of thermosensitive ion channels, thereby falsifying the so-called neuronal hypothesis, which states that rapidly transduced sensations of coldness are related to thermosensitive ion channels. Our results support a central role of mechanosensitive ion channels and the associated hydrodynamic hypothesis. In addition to the hydrodynamic hypothesis, we also demonstrate that the long time delay of dental neuron responses after hot stimulation could be attributed to the neuronal hypothesis-that a relatively long time is required for the temperature around nociceptors to reach some threshold. The results are useful as a model of how multiphysical phenomena can be combined to provide mechanistic insight into different mechanisms underlying pain sensations.

The immune microenvironment extensively participates in tumorigenesis as well as progression in osteosarcoma(OS).However,the landscape and dynamics of immune cells in OS are poorly *** analyzing single-cell RNA sequencing(sc RNA-seq)data,which characterize the transcription state at single-cell resolution,we produced an atlas of the immune microenvironment in *** results suggested that a cluster of regulatory dendritic cells(DCs)might shape the immunosuppressive microenvironment in OS by recruiting regulatory T *** also found that major histocompatibility complex class I(MHC-I)molecules were downregulated in cancer *** findings indicated a reduction in tumor immunogenicity in OS,which can be a potential mechanism of tumor immune *** note,CD24 was identified as a novel“don’t eat me”signal that contributed to the immune evasion of OS ***,our findings provide insights into the immune landscape of OS,suggesting that myeloid-targeted immunotherapy could be a promising approach to treat OS.

In order to investigate the behavioral responses of pregnant ewes to high ambient temperature in a loose housing system,eighty-seven pregnant ewes(Chinese little fat-tailed sheep,Ovis aries)from the same loose house were chosen,and nine ewes with the similar body weight and age were marked for later individual behavioral *** ambient temperature and relative humidity were recorded,and the individual behaviors of the pregnant ewes were observed continuously from 04:50 to 20:00 once every other day for 14 *** behavioral state of all the pregnant ewes was recorded once every hour from 5:00 to 20:00,the number of pregnant ewes selecting specific lying areas(designated as area A and area B),was recorded at 15 minute intervals from 12:00 to 17:00 *** final results show that the behavioral durations are significantly different in eating and drinking,rumination,rest,and locomotion for pregnant ewes(Plocomotion(Plocomotion behavior for the pregnant ewes in the loose housing systems under the high ambient temperature.

Bone scaffolds are critical in current implant and periodontal regeneration approaches. In this study, we prepared a novel composite type-I collagen and hydroxyapatite （HA）/β-tricaleium phosphate （TCP） scaffold （CHTS） by incorporating type-I collagen and bovine calcined bone granules, prepared as a mixture of 50% HA and 50% TCP, by freeze drying. We then characterized the CHTS and determined its cytotoxic effects. Additionally, ridge preservation experiments were carried out to evaluate the clinical effects of the CHTS. The results demonstrated that the composite scaffolds had good surface morphology and no cytotoxicity. Additionally, an in vivo experiment in an animal model showed that the CHTS performed equally as well as Bio-Oss Collagen, a widely used bone graft in ridge preservation. These findings revealed that the CHTS, which contained natural constituents of bone, could be used as a scaffold for bone regeneration and clinical use.

The big-headed turtle Platysternon megacephalum is a stream-dwelling species whose ecology is poorly known. We carried out field and laboratory investigations to determine field body temperatures and thermal preference of this species. In the field, the body temperatures of the turtles conformed to the water temperature, with little diel variation in either summer or au- tumn. Over the diel cycle, the mean body temperatures ranged from 20.8℃ to 22.2℃ in summer and from 19.3℃ to 21.2℃ in autumn; the highest body temperatures ranged from 22.1℃ to 25.0℃ in summer and from 20.6℃ to 23.8℃ in autumn. In the laboratory, the preferred body temperature （Tp） was 25.3℃ Food intake was maximized at 24.0℃, whereas locomotor perfor- mance peaked at 30.0℃. Consequently, Tp was closer to the thermal optimum for food intake than for locomotion. Therefore, this freshwater turtle has relative low field body temperatures corresponding to its thermal environment. In addition, the turtle prefers low temperatures and has a low optimal temperature for food intake [Current Zoology 59 （5）： 626-632, 20＇13].

Intramedullary nailing is a standard treatment for adult long bone fracture. Previous numerous animal, biomechanical and clinical studies have documented the short-term performance of the intramedullary nails;however, the long-term influence of the nails on the change of bone density distribution is still not fully *** to bone remodeling theory, a set of computational algorithms was implemented in the finite element model to predict the bone density distribution for retained intramedullary nail. The bone density was recorded to observe the progression of remodeling. The results show that the effect of intramedullary nail on the Gruen zone 4 and 5 of femur is small, while the effect on the Gruen zone 1, 2 and 6 is greater than other *** simulation indicates that intramedullary nail plays a significant role in the long-term bone density distribution after surgery.

Collective cell migration is extensively observed in embryo development and cancer *** these processes,the interactions between cells with distinct identities and fates are of importance for boundary formation and host defense against *** this paper,we explore the collective dynamics of a two-dimensional cell mixing monolayer consisting of non-tumorigenic mammary epithelial cells and breast cancer *** show that the epithelial–cancerous cell mixing system displays unique sorting *** epithelial cells aggregate to form scattered clusters,which perform random motion with simultaneous translation and rotation,strikingly distinct from the classical persistent random walk of individual migratory *** motility of cancer cells is markedly promoted by the epithelial clusters,exhibiting remarkable contact enhancement of locomotion.A discrete model based on the Johnson–Kendall–Roberts contact mechanics is proposed to identify the influence of intercellular interactions,active migration forces and cell–substrate friction forces on the collective cell *** findings could advance our understanding of many biological processes,such as cancer metastasis and tissue morphogenesis.
