Carbon monoxide（CO） has recently been well-established as a promising therapeutic modality against cancer. The development of transition-metal-free and tumor-targeted delivery platforms for CO is highly demanded but remains very sporadic. Herein, we developed a polyprodrug system for the co-delivery of CO and a chemotherapeutic agent combretastin A-4（CA-4） to the tumor site based on a chemiexcitation mechanism. Such a poly-prodrug successfully achieved the co-activation of CO and CA-4 in response to H2O2at the tumor site both in vitro and in vivo. Importantly, a synergist effect between CO and CA-4 was observed in suppressing tumor growth in a mouse tumor model. It is anticipated that such a poly-prodrug system could be potentially adapted for the co-delivery of CO and another drug payload for combinational therapy for other indications.

Organic electrochemical transistors have emerged as a solution for artificial synapses that mimic the neural functions of the brain structure,holding great potentials to break the bottleneck of von Neumann ***,current artificial synapses rely primarily on electrical signals,and little attention has been paid to the vital role of neurotransmitter-mediated artificial *** is a key neurotransmitter associated with emotion regulation and cognitive processes that needs to be monitored in real time to advance the development of disease diagnostics and *** provide insights into the development of artificial synapses with neurotransmitter involvement,this review proposes three steps towards future biomimic and bioinspired neuromorphic *** first summarize OECT-based dopamine detection devices,and then review advances in neurotransmitter-mediated artificial synapses and resultant advanced neuromorphic ***,by exploring the challenges and opportunities related to such neuromorphic systems,we provide a perspective on the future development of biomimetic and bioinspired neuromorphic systems.

Hydrogen sul fi de(H_2S) is recognized as one of three gasotransmitters together with nitric oxide(NO) and carbon monoxide(CO). As a signaling molecule, H_2S plays an important role in physiology and shows great potential in pharmaceutical applications. Along this line, there is a need for the development of H_2S prodrugs for various reasons. In this review, we summarize different H_2S prodrugs, their chemical properties, and some of their potential therapeutic applications.

AIM: To investigate how the saturated and unsaturated fatty acid composition influences the susceptibility of developing acute pancreatitis. METHODS: Primary pancreatic acinar cells were treated with low and high concentrations of different saturated and unsaturated fatty acids, and changes in the cytosolic Ca2+ signal and the expression of protein kinase C(PKC) were measured after treatment. RESULTS: Unsaturated fatty acids at high concentrations, including oleic acid, linoleic acid, palmitoleic acid, docosahexaenoic acid, and arachidonic acid, induced a persistent rise in cytosolic Ca2+ concentrations in acinar cells. Unsaturated fatty acids at low concentrations and saturated fatty acids, including palmitic acid, stearic acid, and triglycerides, at low and high concentrations were unable to induce a rise in Ca2+ concentrations in acinar cells. Unsaturated fatty acids at high concentrations but not saturated fatty acids induced intra-acinar cell trypsin activation and cell damage and increased PKC ***: At sufficiently high concentrations, unsaturated fatty acids were able to induce acinar cells injury and promote the development of pancreatitis. Unsaturated fatty acids may play a distinctive role in the pathogenesis of pancreatitis through the activation of PKC family members.

Hydrogen sulfide(H2S) is commonly referred to as the third gasotransmitter with firmly established physiological roles. Prodrug approaches have been broadly applied to deliver H2S for various applications and mechanistic studies. Since S-persulfidation and glutathionylation are known to be important in cellular signaling by sulfur species, there have been interests in developing donors of persulfide and glutathione persulfide as well. In this review, we discuss the recent development in area of prodrugs for various sulfur species.

Click chemistry has been proven to be very useful in drug *** to the availability of a large number of click reactions with a various characteristics,selection of appropriate chemistry for a given application is often not a trivial *** review is written for pharmaceutical researchers who are interested in click chemistry applications and yet may not be click chemistry *** this,the review gives an overview of available click reactions organized by application ***,the general understanding of click reactions being fast and high yielding sometimes overshadows the need to analyze reaction kinetics in assessing suitability of a given reaction for certain *** this,we highlight the need to analyze the relationship among reaction kinetics,concentration effects,and reaction time scales,knowing that lack of such analysis could easily lead to ***,possible issues such as chemical stability with various click reagents are also discussed to aid experimental *** examples and extensive references are also provided to aid in-depth understanding of technical *** hope this review will help those interested in using click chemistry in drug delivery to select the appropriate reactions/reagents and minimize the number of pitfalls.

Parkinson’s disease(PD)is one of the fastest-growing neurodegenerative disorders *** far,PD treatments only offer little clinical relief and cannot reverse or stop the disease *** cell(SC)therapy is a rapidly evolving technology that holds significant promise for enhancing current therapeutic ***-derived mesenchymal SCs(AD-MSCs)have many features such as easy harvest with minimal invasive techniques,high plasticity,non-immunogenicity,and no ethical issues,which have made them suitable choices for clinical applications in regenerative ***-MSCs are ideal tools to treat PD,as they have the potential to differentiate into functional dopaminergic neurons,and also could produce and secrete useful paracrine factors and extracellular vesicles,such as cytokines and growth factors,and thus promote the repair and regeneration of damaged nerve *** revealed that AD-MSCs induced angiogenesis,nerve regeneration,and memory and motor improvement in cellular and animal models of ***,clinical studies demonstrated the safety of AD-MSC transplantation in PD *** review provides a comprehensive and current summary of the therapeutic potential of AD-MSC transplantation for the treatment of PD,by highlighting the ability of cells to differentiate into functional dopaminergic neurons.

Objective:To investigate the pharmacodynamic material basis,mechanism of actions and targeted diseases of Salicornia europaea L.(SE)based on the network pharmacology method,and to verify the antidepressant-like effect of the SE extract by pharmacological ***:Retrieval tools including Chinese medicine(CM),PubMed,PharmMapper,MAS 3.0 and Cytoscape were used to search the components of SE,predict its targets and related therapeutic diseases,and construct the"Component-TargetPathway"network of SE for central nervous system(CNS)***,protein-protein interaction(PPI)network,Kyoto Encyclopedia of Genes and Genomes(KEGG)and Gene Ontology(GO)function annotation of depression-related targets were analyzed to predict the antidepressant mechanism of *** unpredictable mild stress(CUMS)model was used to construct a mouse model with depression-like *** the animals were randomly divided into 6 groups(n=10)including the normal group(nonstressed mice administered with distilled water),the CUMS group(CUMS mice administered with distilled water),the venlafaxine group(CUMS mice administered with venlafaxine 9.38 mg/kg),SE high-,medium-,and low-dose groups(CUMS mice administered with SE 1.8,1.35 and 0.9 g/kg,respectively).Then some relevant indicators were determined for experimental verification by the forced swim test(FST),the tail suspension test(TST)and open-field test(OFT).Dopamine(DA)concentration in hippocampus and cerebral cortex,IL-2 and corticosterone(CORT)levels in blood,and nuclear factor E2 related factor 2(Nrf2),kelch-like epichlorohydrin related protein 1(Keap1),NAD(P)H dehydrogenase[quinone]1(NQO1)and heme oxygenase-1(HO-1)levels in mice were measured by enzyme linked immunosorbent assay(ELISA)and Western blot respectively to explore the possible ***:The"target-disease"network diagram predicted by network pharmacology,showed that the potential target of SE involves a variety of CNS diseases,among which depression ac

In the era of Metaverse and virtual reality(VR)/augmented reality(AR),capturing finger motion and force interactions is crucial for immersive human-machine *** study introduces a flexible electronic skin for the index finger,addressing coupled perception of both state and process in dynamic tactile *** device integrates resistive and giant magnetoelastic sensors,enabling detection of surface pressure and finger joint *** e-skin identifies three phases of finger action:bending state,dynamic normal force and tangential force(sweeping).The system comprises resistive carbon nanotubes(CNT)/polydimethylsiloxane(PDMS)films for bending sensing and magnetoelastic sensors(NdFeB particles,EcoFlex,and flexible coils)for pressure *** inward bending resistive sensor,based on self-assembled microstructures,exhibits directional specificity with a response time under 120 ms and bending sensitivity from 0°to 120°.The magnetoelastic sensors demonstrate specific responses to frequency and deformation magnitude,as well as sensitivity to surface roughness during sliding and material *** system’s capability is demonstrated through tactile-based bread type and condition recognition,achieving 92%*** intelligent patch shows broad potential in enhancing interactions across various fields,from VR/AR interfaces and medical diagnostics to smart manufacturing and industrial automation.

Carbon monoxide(CO) as one of the three important endogenously produced signaling molecules, termed as "gasotransmitter," has emerged as a promising therapeutic agent for treating various inflammation and cellular-stress related diseases. In this review, we discussed CO’s evolution from a well-recognized toxic gas to a signaling molecule, and the effort to develop different approaches to deliver it for therapeutic application. We also summarize recently reported chemistry towards different CO delivery forms.
