In silico docking analysis of beta-defensin 20 against cation channel sperm-associated protein 1–4 to predict its role in the sperm maturation

Beta-defensin 20(DEFB20)is widely expressed in the epididymis with gene features involved in epididymal sperm maturation.However,the action mechanism and function of DEFB20 in sperm maturation are still unclear.One of the important roles of beta-defensin is the ion channel activity.The cation channel sperm-associated protein(CatSper)alpha is an ion channel protein found on the sperm surface.This study aimed to investigate the interaction between DEFB20 and CatSper1–4 protein in relation to the sperm maturation process.Protein sequences were obtained from the National Center for Biotechnology Information(NCBI).Protein modeling and validation were carried out by using the Robetta modeling server and the Ramachandran plot method.Rosetta web server was used for the docking analysis.The results revealed a natural interaction between DEFB20 and CatSper1–4.The interaction occurred at the cation channel(close to the casein kinase II),ion transport protein,and kinase c phosphorylation of the CatSper1–4 active site.The DEFB20 region interacting with CatSper2–4 was the beta-defensin domain,while with CatSper1 was the non-beta-defensin domain.Based on the analysis,DEFB20 may interact with CatSperαsubunits,particularly CatsSper1,to affect ion channel activity during sperm maturation.

Pterostilbene is a natural compound that can be found in various food plants such as blueberries,grapes,and peanuts.It has also been reported to be extracted from Pterocarpus indicus,a tree species native to India and Southeast Asia.Pterostilbene exhibits various pharmacological activities such as antioxidants,anti-proliferation,anti-microbial,and anti-inflammatory activities with favorable pharmacokinetic properties,such as high oral bioavailability and longer half-life.The anti-inflammatory effect of pterostilbene has been reported to contribute to its therapeutic effects in many chronic inflammatory diseases.Besides,pterostilbene has anti-cancer activity on various types of cancers due to its ability to induce cell cycle arrest and apoptosis.Hence,in this review,we discuss the anti-inflammatory and anti-cancer activities of pterostilbene in preclinical studies.

Obesity-induced inflammation in white adipose tissue,characterized by increased macrophage infiltration and associated with macrophage population shift from anti-inflammatory M2 to proinflammatory M1 macrophages,largely contributes to obesity-induced insulin resistance and influences T2 DM pathogenesis.GSK3,a serine/threonine kinase,has been reported to participate in various cellular processes.We sought to examine the potential mechanism by which GSK3,a serine/threonine kinase implaicated in various cellular processes,modulates obesity-induced visceral adipose tissue(VAT) inflammation.Male C57 BL/6 J mice were fed a high-fat diet for 10 weeks while being treated with control or GSK3 inhibitors.RNA-Sequencing results using VAT demonstrated that GSK3 inhibitor treatment reversed obesity-specific expression of genes associated with inflammation.Consistently,GSK3 inhibition reduced M1 macrophages and increased M2 macrophages in VAT,while decreasing inflammatory monocytes.These anti-inflammatory effects of GSK3 inhibition were found to be driven,at least in part,by inhibiting production of apoptosis inhibitor of macrophage(AIM) in macrophages via inactivating STAT3 to reduce free fatty acid and chemokine level produced from VAT to suppress circulatory monocyte chemotaxis.Our findings suggest that GSK3 may act as an important regulator of obesity-induced inflammation and characterize the novel role of GSK3 in shifting macrophage polarization and reinforce its therapeutic potential for obesity-induced inflammation and its associated diabetes.

Human pluripotent stem cells(hPSCs)are important resources for cell-based therapies and pharmaceutical applications.In order to realize the potential of hPSCs,it is critical to develop suitable technologies required for specific applications.Most hPSC technologies depend on cell culture,and are critically influenced by culture medium composition,extracellular matrices,handling methods,and culture platforms.This review summarizes the major technological advances in hPSC culture,and highlights the opportunities and challenges in future therapeutic applications.

The need for novel therapeutics against human cancers such as leukemias and solid tumors is well recognized.Human T cells are poised to make a fundamental change in the therapeutic approach.T-cell interaction with a tumor cell is a critical event and primarily driven by T-cell receptor(TCR)recognition of peptide in the pocket HLA.However,among TCR-based T-cell therapies,either TCR mismatching or the low density of major histocompatibility complex causes tumor cells to escape from the immune response.TCR molecules have low binding affinities,preventing their recognitions.Undoubtedly,antibody therapeutics is an effective treatment for cancer.As the new generation of monoclonal antibodies,TCR-like antibodies can mimic TCR recognition but are not susceptible for mechanisms of tumor evasion from the immune response.As chimeric antigen receptor(CAR)structure expressed on the surface of T cells,TCR-like antibodies can confer antigen specificity to T cells.The new TCR-like CAR may be important to drive new technologies of adoptive cell therapy,in particular,T-cell therapy,and open possibilities to target endogenous tumor-specific antigens.

Alzheimer’s disease(AD)is a progressive neurodegenerative disease in which patients exhibit gradual loss of memory that impairs their ability to learn or carry out daily tasks.Diagnosis of AD is difficult,particularly in early stages of the disease,and largely consists of cognitive assessments,with only one in four patients being correctly diagnosed.Development of novel therapeutics for the treatment of AD has proved to be a lengthy,costly and relatively unproductive process with attrition rates of>90%.As a result,there are no cures for AD and few treatment options available for patients.Therefore,there is a pressing need for drug discovery platforms that can accurately and reproducibly mimic the AD phenotype and be amenable to high content screening applications.Here,we discuss the use of induced pluripotent stem cells(iPSCs),which can be derived from adult cells,as a method of recapitulation of AD phenotype in vitro.We assess their potential use in high content screening assays and the barriers that exist to realising their full potential in predictive efficacy,toxicology and disease modelling.At present,a number of limitations need to be addressed before the use of iPSC technology can be fully realised in AD therapeutic applications.However,whilst the use of AD-derived iPSCs in drug discovery remains a fledgling field,it is one with immense potential that is likely to reach fruition within the next few years.

“Obesity is a disease.” This is the declaration by the American Medical Association in June, 2013. The purpose of this article is to provide an overview of options for patients who meet the criteria of obesity. Primary care nurse practitioners are urged to confront obesity head on, with early, individualized intervention to reduce associated morbidity and mortality. Options for weight loss and maintenance are abundant and often confusing or unattractive to the patient. Working with patients to achieve realistic, reasonable, and patient-tailored goals for weight loss and exercise programs are paramount to achieving a healthy weight and lifestyle.
