There are two source plants for the traditional Chinese medicine Murrayae Folium et Cacumen(MFC)in Chinese Pharmaco.oeia,*** exotica *** ***(L.)***,a chemical co.parison of *** and *** by high performance liquid chromatography(HPLC)fingerprint analysis co.pled with chemometrics and network pharmaco.ogy was *** main peaks in the fingerprints were identified by liquid chromatography co.pled with ion trap/time-of-flight mass spectrometry(LC-IT-TOF-MS)and authenticated by *** chemometrics results showed that the HPLC fingerprints of these two species were clearly divided into two categories using hierarchical cluster analysis(HCA)and principal co.ponent analysis(PCA),and a total of 13 significantly differentiated markers were screened out by orthogonal partial least squares-discriminant analysis(OPLS-DA).However,the following network pharmaco.ogy analysis showed that these discriminated markers were found to act via many co.mon targets and metabolic pathways,indicating the possibly similar pharmaco.ogical effects and mechanisms for *** and *** above results provide valuable evidence for the equivalent use of these two plants in clinical ***,the chromatographic fingerprint analysis co.pled with chemometrics and network pharmaco.ogy supplies an efficient approach for the co.parative analysis of multi-source TCMs like MFC.

Ilex asprella(*** Arn.)*** Benth is one of the most important traditional Chinese medicines in southern *** seeds of Ilex asprella usually have extremely low germination due to their dormancy characteristics,which severely impacts the efficiency of seedling raising and increases labor *** this study,to improve the seed germination of ***,the effects of germination substrate,hormone,winnowing,and stratification treatments on the seed germination of *** were *** results of the germination matrix showed that the highest germination percentage of 45.2%was achieved under the 20℃/10℃day/night temperature and vermiculite germination medium *** results of hormone treatments revealed that 100–400 mg/L of gibberellin(GA)and 50–100 mg/L of salicylic acid(SA)were found to be effective in releasing the dormancy of *** ***,winnowing co.ld effectively eliminate unsaturated seeds and impurities,thus improving the seed germination of ***,warm temperature(15℃)stratification co.ld expand the temperature range of ***’s seed germination,which was beneficial for seed germination of *** and for seed nursery at room temperature in production *** present study obtained a method to break dormancy and increase seed germination in ***,thereby forming a groundwork for improving the efficiency of large-scale planting of ***.

Using nerve guide co.duits (NGCs) to promote the regeneration of PNI is a feasible alternative to *** with NGCs made of single material,co.posite NGCs have a greater development *** previous research has co.firmed that poly(D,L-lactic acid)/β-tricalcium phosphate/hyaluronic acid/chitosan/nerve growth factor (PDLLA/β-TCP/HA/CHS/NGF) NGCs have excellent physical and chemical properties,which can slowly release NGF and support cell adhesion and *** this study,PDLLA/β-TCP/HA/CHS/NGF NGCs were prepared and used to bridge a 10 mm sciatic nerve defect in 200-250 g Sprague-Dawley (SD) rat to verify the performance of the NGCs in *** improvements in nerve regeneration were observed after using the PDLLA/β-TCP/HA/CHS/NGF NGCs based on gross post-operation observation,triceps wet weight analysis and nerve histological *** vivo studies illustrate that the PDLLA/β-TCP/HA/CHS/NGF sustained-release NGCs can effectively promote peripheral nerve regeneration,and the effect is similar to that of autograft.

Shenfu Injection (SFI) is a well-defined Chinese herbal formulation that is obtained from red ginseng and processed aco.ite root. The main active co.stituents in SFI are ginsenosides and aco.itum alkaloids. In this work, ginsenosides (ginsenoside Rg 1, ginsenoside Rbl and ginsenoside Rc) and aco.itum alkaloids (benzoylmesaco.ine and fuziline) were used as the index co.ponents to explore the pharmaco.inetic behavior of SFI. A selective and sensitive HPLC MS/MS method was developed for the quantification of ginsenosides and aco.itum alkaloids in dog plasma and was used to characterize the pharmaco.inetics of the five index co.ponents after intravenous drip of three different dosages of SFI in beagle dogs. The pharmaco.inetic properties of the index co.ponents were linear over the dose range of 2-8 mL/kg. Antiviral Oral Liquid(AOL)is an adult medicine in the Chinese Pharmaco.oeia used to treat upper respiratory infections such as *** has shown promising clinical efficacy in relieving flu-like symptoms such as fever,inflammation,and pharyngalgia both in adults and ***,the instruction manual does not specify the exact usage and dosage of AOL for *** this article,we set 6 dosage ranges:0.2,0.5,0.7,0.9,1.1,1.4 mL/kg/d,acco.ding to its dosage for adults and the co.version method between adult and children *** six animal models were used to evaluate the effectiveness of AOL in different *** results indicated that AOL co.ld reduce the lung index,virus load,and expression of proinflammatory cytokines in the *** co.ld improve pathological changes and prolong the survival time of mice infected by the Influenza A virus(H1N1)A/PR/8/34 strains at 0.5–0.9 mL/kg/d co.centrations in different *** four dose groups of 0.7–1.4 mL/kg/d co.ld significantly inhibit the ear shell swelling caused by xylene and reduce the rabbit body temperature induced by lipopolysaccharide(Pco.ld inhibit the increase of peritoneal capillary permeability induced by glacial acetic acid(P<0.01).AOL at 0.7 and 0.9 mL/kg/d reduced the painful writhing times in young mice induced by glacial acetic(P<0.01).These results indicated that the optimal dose of AOL in antiviral,antipyretic,anti-inflammatory,and analgesic effects is 0.7 mL/kg/d.

Qualitative and quantitative assessment of related substances in the co.pound Ketoco.azole and Clobetasol Propionate Cream by HPLC-TOF-MS and HPLC

Related substances in pharmaceutical formulations are associated with their safety, efficacy and stability. However, there is no overall study already published on the assessment of related substances in the co.pound Ketoco.azole and Clobetasol Propionate Cream. In this work, a reliable HPLC-TOF-MS qualitative method was developed for the analysis of related substances in this preparation with a quick and easy extraction procedure. Besides the active pharmaceutical ingredients, two co.pounds named ketoco.azole impurity B′ optical isomer and ketoco.azole impurity E were identified. Furthermore, a new HPLC method for qualitative and quantitative assessment on related substances and degradation products, which were found in the stability test, was established and validated. The single standard to determine multi-co.ponents method was applied in the quantitative analysis, which was an effective way for reducing co.t and improving accuracy. This study can provide a creative idea for routine analysis of quality co.trol of the co.pound Ketoco.azole and Clobetasol Propionate Cream.

Shikonin alleviates co.lagen-induced arthritis mice by inhibiting M1 macrophage polarization

OBJECTIVE: To investigate the effects of shikonin(SKN) on M1 and M2 polarization of macrophages both in vitro and in vivo. METHODS: co.lagen-induced arthritis(CIA) in male DBA/1 mice were treated with a dose of 4 mg/kg/day of SKN for 23 d(n = 6/group). The histopathology of inflamed joints in CIA mice was evaluated to test the antiarthritic effect of SKN. M1/M2 polarization of macrophages induced by lipopolysaccharide(LPS) and interferon(IFN)-γ or interleukin(IL)-4 and IL-13, were used to assess the effect of SKN(0.05, 0.1, and 0.2 μM). The effect of SKN on the protein expression of nitric oxide synthase, arginase, CD68, and CD206 was evaluated using western blot analysis. RESULTS: The results of this study revealed that SKN delayed the arthritis feet symptom sco.e, reduced the incidence rate of arthritis, and relieved the inflammation of joints in CIA mice. SKN inhibited M1 macrophage polarization but did not affect M2 macrophage polarization in the joints of CIA mice. Moreover, SKN inhibited M1 polarization induced by LPS and IFN-γ, but did not affect M2 polarization induced by IL-4 and IL-13. co.CLUSION: These findings suggest that SKN alleviated CIA through inhibiting M1 macrophage polarization and has great potential as a new drug for RA treatment.

OBJECTIVE:To evaluate the clinical efficacy and safety of Rebixiao(RBX)Chinese herbal tablets(CHT)and Chinese formula granules(CFG)in the treatment of acute gout arthritis(AGA).METHODS:This randomized,multicenter,double-blind,co.trolled trial included 165 AGA patients with the damp-heat symptom pattern who were randomly divided into an RBX CHT group and an RBX CFG group and treated for 7 d at three *** total effective rates of the joint symptom sco.e,Traditional Chinese Medicine(TCM)symptoms sco.e,erythrocyte sedimentation rate(ESR),and C-reactive protein(CRP)were used to evaluate the clinical *** assessments were also ***:Of the 165 enrolled patients,147 co.pleted the clinical *** was no difference in baseline between the two *** total effective rates of the joint symptom sco.e were 94.36%and 97.36%,and the total effective rates of the TCM symptoms sco.e were 95.77%and 97.36%in the CFG group and CHT group,*** statistical difference was found between the two groups(P>0.05).Additionally,ESR and CRP were similar in both groups(P>0.05).Furthermore,treatment efficacy regarding TCM and joint symptoms,the ESR,and CRP were co.sistent within each center and among the different centers(P>0.05).In addition,the incidence of adverse events was 4.22%and 2.63%in the CFG group and CHT group,respectively,and no difference was observed between the two groups(P>0.05).co.CLUSION:RBX CFG and CHT have significant and similar efficacy in the treatment of AGA,and CFG did not increase adverse side effects.
