A model of white blood cell membrane chromatography (WB-CMC) was established to screen active component from Atractylodes macrocephala Koidz. The component can antagonize Toll-like receptor 4 (TLR4) and inhibit inflammatory reaction. In the model of WB-CMC, cell membrane stationary phase (CMSP) was prepared by immobilizing the rabbit white blood cell membrane (WBCM) onto the surface of silica carrier and taxinol was used as a model molecule. The active component which can act on WBCM and its receptor (such as TLR4) as an effective target in A. macrocephala was determined by using a replacement experiment. The anti-inflammatory effects of the active component were tested by using pharmacological methods in vivo. The results indicated that the re-tention characteristics of atractylenolide I as active component was similar to that of taxinol in the model of WB-CMC. And so, atractylenolide I acted on the WBCM and TLR4 and its anti-inflammatory activity was related with antagonizing TLR4. Therefore, the interaction between the active component and WBCM and its receptor can be simulated by the model of WB-CMC in vitro. This model can be used to screen active components and to study effective characteristics for acting on definite targets.

To analyze the binding of glycyrrhetic acid (GA) to angiotensin II type I (AT1) receptor and to explore the mechanisms underlying the binding, primary cell culture of rat vascular smooth muscle cell (VSMC), radioactive ligand-receptor binding assay, lascer confocal scanning micro-scope (LCSM), Northern blot, 3H-TdR incorporation DNA assay were used in this study. The re-sults suggest that specific binding of GA to AT1 receptor (IC50 value was 35.0 mmol/L) increases intracellular [Ca2+]i of VSMC, activates transcription factor c-myc and promotes the proliferation of VSMC, therefore GA was probably an agonist of AT1 receptor, providing a new target for GA抯 pharmaceutical effects.

Estrogen receptor (ER), one member of nuclear hormone receptor (NR) family, is an estrogen-dependent transcriptional factor that plays an important role in development, progression and treatment of breast cancer. Transcriptional co-factors (co-activators and co-repressors) are critical for ER to transduce hormone and metabolic signaling to target genes. A number of functional and structural studies have elucidated the precise mechanisms of co-activator interaction with the ligand-inducible activation domain in ER via one and several LXXLL motifs (where X is any amino acid) known as NR-Box. By the yeast two-hybrid system we have identified a novel ER-a interacting protein ERIAP (Estrogen receptor Interacting and Activating Protein) which contains two consensus LXXLL motifs. ERIAP associated with ER-a in a ligand-dependent manner, as demonstrated by in vivo immunoprecipitation and in vitro GST capture assays. The two NR boxes were essential for ERIAP interaction with ER-a. Furthermore, ERIAP specifically enhanced ligand-mediated ER-a transcriptional activity in a dose-dependent fasion and increased the expression of estrogen-responsive gene pS2. Thus, our present findings indicate that ERIAP funcions as a new coactivator for ER-a transcriptional activity, which may play an important role in development and progression of breast cancer.

Mutations of the melanocortin-4 receptor (MC4R) gene are associated with the appetite,obesity and growth in pig,mice and human. But little is known about the function of chicken MC4R gene. In this study,F2 chicken resource population derived from broilers crossing to Silky was screened for the polymorphisms of the MC4R gene using PCR-single strand conformation polymor-phism (PCR-SSCP) and DNA sequencing methods. Four single nucleotide polymorphisms (SNPs) sites were found. The mutation (C→T) in the 5′ regulation region of chicken MC4R gene results in one more NF-E2 and cap transcription factor binding sites in the mutation allele than in the wild allele. One missense mutation (G→A) occurs in the coding region (61nt),which changes the glycine to arginine. Moreover,in the coding region there are 2 synonymous mutations,one G→T mutation at 315nt and one C→T mutation at 336nt. Least square analysis of the SNPs and carcass traits showed that BB,DD and FF genotypes are significantly associated with body weight,carcass weight (or half carcass weight),and leg muscle weight (P<0.05 or P<0.01). But no significant association between the geno-types and abdominal fat weight is found. The results present the evidence that the chicken MC4R gene can be selected as the major candidate gene for the carcass traits such as body weight and growth.

Epstein-Barr virus (EBV) encoded latent membrane protein 1 (LMP1) is considered to be the major oncogenic protein of EBV encoded proteins, and also it has always been the core of the oncogenic mechanism of EBV. Traditional receptor theory demonstrates that cell surface receptors exert biological functions on the membrane, which neither enter into the nucleus nor directly affect the transcription of the target genes. But, advanced studies on nuclear transloca-tion of the epidermal growth factor receptor (EGFR) family have greatly developed our knowl-edge of the biological function of cell surface receptors. In this study, we used Tet-on LMP1 HNE2 cell line as a cell model, which is a dual-stable LMP1 integrated NPC cell line and the ex-pression of LMP1 in which could be regulated by Tet system. We found that LMP1 could regulate the nuclear translocation of EGFR in a dose-dependent manner from both quantitative and qualitative levels through the Western blot analysis and the immunofluorescent analysis with a laser scanning confocal microscope. We further demonstrated that the nuclear localization se-quence of EGFR played some roles in the location of the protein within the nucleus under LMP1 regulation, and the nuclear accumulation of EGFR regulated by LMP1 was in a ligand-independent manner. These findings provide a novel view that the regulation of LMP1 on the nuclear translocation of EGFR is critical for the process of nasopharyngeal carcinoma.

Ethylene plays important roles in plant growth, development and stress responses. Its receptor genes have been studied in dicots such as Arabidopsis, tobacco and tomato. However, no research has been reported for the ethylene receptors from monocots currently. In the present study, we cloned an ethylene receptor gene OSPK2 from rice and found that its encoded protein was divergent from the ethylene receptors from dicots. OSPK2 had a long extension in its N- terminal, followed by three transmembrane segments, a GAF domain, a putative kinase domain and a putative receiver domain. Although most of the domains were conserved, the expected phosphorylation site His and the phosphate receiver Asp have been replaced by Gln and Asn, respectively. This fact indicates that OSPK2 may not function as a histidine kinase in a phosphorelay manner, but rather play roles by other mechanism, probably through Ser/Thr kinase activity. The expression of the OSPK2 gene was investigated by RT-PCR method under different conditions. We found that this gene was apparently induced by wounding and PEG treatment, but not significantly affected by salt and ABA treatments. The differential expression of the OSPK2 gene may reflect its roles in mediating different abiotic stress responses, consistent with our previous studies on tobacco ethylene receptors.

Two multiamide calyx[4].arenas(5,6)were synthesized and characterized by IR,1H NMR,MS and elemental *** binding properties of receptors with some anions (p-O2NphOpO2-3,p-O2NphO-,H2PO-4,Ac-,CI-,Br- and I-)Were studied by UV-Vis *** results indicate that the tetraamide calix[4]arenas (5,6) have a good selectivity to the anions containing aromatic ring (p-O2NphOpO2-3,p-O2NphO-).The 1:1 complexes between host and guest were formed through multiple hydrogen bonding and π-π *** hosts 5 and 6 also show a definite binding ability for the anions (H2PO-4,Ac-,CI-) that have no ultraviolet absorption,which provides a simple method of spectrum detection for these anions.

Intracellular recordings were made from luminosity-type horizontal cells (LHCs) in the isolated superfused carp retina and the effect of AMPA (a-amino-3-hydroxy-5-methylisoxazole-4-propionic acid), a glutamate receptor agonist, on these cells was studied. AMPA suppressed the responses of LHCs driven by red-sensitive (R-) cones whereas it potentiated the responses driven by green-sensitive (G-) cones. The AMPA effect could be completely blocked by GYKI 53655, a specific AMPA receptor antagonist, indicating the exclusive involvement of AMPA-preferring receptors. The AMPA effect persisted in the presence of picrotoxin (PTX) or dihydrokainic acid (DHK), suggesting that the feedback from LHCs onto cones and glutamate transporters on cones may not be involved. It is suggested that there may exist different AMPA receptor subtypes with distinct characteristics on LHCs, which mediate signal transfer from R- and G-cones to LHCs, respectively.
