Protective effects of polygonatum sibiricum against CCl_(4)-induced acute liver injury in rats through oxidative stress and mitochondrial apoptotic pathways

In the present study,we aimed to investigate the hepatoprotective effect of Chinese herbal medicine polygonatum sibiricum(PS).In this study,a rat acute liver injury(ALI)model was established by a single intraperitoneal injection of 50%CCl_(4) oil solution,and the rats were treated intragastrically with polygonatum sibiricum aqueous extract(PSAE).The results showed that PSAE significantly decreased the serum levels of ALT,AST and ALP,increased the activities of glutathione(GSH)and superoxide dismutase(SOD),decreased malondialdehyde(MDA)activity in hepatic tissue,and decreased the reactive oxygen species(ROS)level in *** expressions of Nrf2,NQO-1,HO-1,Bcl-2,Bcl-x L mRNA,and HO-1 proteins were elevated,and the expression of p53 mRNA was *** conclusion,PSAE exerted a powerful protective action against CCl_(4)-induced ALI in rats via effectively regulating the expressions of Nrf2-Keap1-ARE related genes and proteins,and inhibiting hepatocyte *** outcomes provided evidence that PS had apparent hepatoprotective effect.

[Objectives]The paper was to establish the quality standard for freeze-dried tablets of polygonatum sibiricum and to explore the antitumor activity of its extract diosgenin.[Methods]Taking freeze-dried powder samples of *** from 4 different producing areas as materials,and referring to the quality standard of *** in the Chinese Pharmacopoeia(2020 edition),the contents of total ash,moisture,extract,total sugar and diosgenin were determined by total ash determination method,drying method,hot dipping method,0.2%anthrone-sulfuric acid method and HPLC,*** antitumor activities of diosgenin against A431(human epidermal carcinoma cells),H1975(human lung adenocarcinoma cells)and Ramos(human B lymphoblastoma cells)were investigated by MTT assay.[Results]The moisture content of the samples was 2.8%-4.7%(not more than 18.0%);the total ash content was 1.9%-3.4%(not more than 4.0%);the ethanol-soluble extract content was 72.99%-78.99%(not less than 45.0%);and the total sugar content was 7.95%-9.94%(not less than 7.0%).The lowest content of diosgenin was 0.18%,and diosgenin was significantly resistant to A431.[Conclusions]The content determination method established in the study is simple,accurate and reproducible.

In vitro Inhibitory Activity of Shisandra chinensis and polygonatum sibiricum against Vibrio harveyi and Its Biofilms

[Objective] This study aimed to analyze the in vitro inhibitory activity of Shisandra chinensis and polygonatum sibiricum against Vibrio harveyi and its biofilms. [Result] By agar diffusion test, in vitro inhibitory activity of 5. chinensis and P. sibiricum against V. harveyi was investigated. The minimal inhibitory concentration （ MIC） and minimal bactericidal concentration （MBC） of 5. chinensis and P. sibiricum against V. harveyi were determined by doubling dilution meth-od. The inhibitory activity of 5. chinensis and P. sibiricum on the formation of V. harveyi biofilms was evaluated by modified MTT assay. [ Result ] Both 5. chinen-sis and P. sibiricum had inhibitory activity against V. harveyi. The inhibition zone diameter of 5. chinensis against V. harveyi was 17. 95 mm; MIC and MBC of 5. chinensis were both 3.125 mg/ml. The inhibition zone diameter of P. sibiricum against V. harveyi was 12. 22 mm; MIC and MBC of P. sibiricum were 3.125 and 6.250 mg/ml, respectively. When the concentration was higher than 6. 25 mg/ml, 5. chinensis decoction had extremely significant inhibitory activity against V. harveyi （P 〈 0. 01） ; when the concentration was higher than 3. 125 mg/ml, P. sibiricum had extremely significant inhibitory activity against V. harveyi （P 〈0. 01）. [ Conclusion] 5. chinensis and P. sibiricum could significantly inhibit V. harveyi and its biofilms.

Research progress in immune regulation mechanism of Polygonati Rhizoma polysaccharide

polygonatum sibiricum Hua(Asparagaceae) is an important perennial Chinese herb used for treatment of diabetes and asthma. Anhui Province is a leading producer of P. sibiricum in China, with approximately 100 ha of commercial production. In a survey from 2013 to 2014, a pink mold on the fruit was also frequently observed(at 5 to 10% incidence) in Qingyang County, Anhui Province, which comprised approximately 10 ha of production. Fruits exhibited white mycelium and salmon-colored spores on the surface, and severe internal rot. To isolate the causal organism, fragments(2 × 2 mm) were excised from 35 symptomatic fruits, surface-sterilized in1.5% sodium hypochlorite for 1 min, rinsed three times with sterilized water, plated on potato dextrose agar(PDA)medium, and incubated at 25°C for 7 days. Morphologically similar fungal isolates were recovered(n = 20) on 35 fruit collected from 20 commercial orchards, each of which produced pink to white fungal colonies with hyaline hyphae, simple or branched conidiophores, and conidia in basipetal chains(frequently clustered). Conidia were ellipsoidal or obpyriform. Almost all spores were two-celled(the upper cell slightly larger). Conidia measured14.2 to 17.9 × 6.9 to 11.2 μm. According to these cultural characteristics, the fungal isolates were identified as Trichothecium roseum. To validate the species identification, isolate QY-52 was selected for molecular identification by amplifying and sequencing the ribosomal DNA internal transcribed spacer(rDNA-ITS) region using the universal primer pair ITS4 and ITS5. Sequencing of the PCR product revealed 100% similarity with ITS rDNA sequences of T. roseum in GenBank(EU552162.1, JQ434579.1, and JN942882.1). A pathogenicity test was conducted using isolate QY-52 on six asymptomatic fruits of 10 3-year-old P. sibiricum plants, in which each fruit was wounded with a pushpin at one site/fruit. 黄精(polygonatum sibiricum)是药食同源植物,耐阴使其成为林下栽培的理想经济作物。从黄精自然分布区中选取10个种源,在北京燕山南麓的怀柔区展开种源试验,栽培于北京地区造林工程的栾树林下,测定根茎质量增加值、根茎干质量、根茎鲜质量、新根茎长度、新根茎直径、根茎体积、叶片面积、多糖含量共8项生物指标。经过统计分析,确定来自河北遵化和陕西略阳的黄精地理种源在北京表现最佳。本研究对黄精林下栽培的种源选择和黄精育种提供了有益的基础数据,有助于北京林下黄精生态产业的发展。
