Melittin enhanced intracellular trehalose with synergistic membrane stabilization of macromolecular protectants for cell cryopreservation

cryopreservation plays an essential role in biobanking and cell therapy,but the physiological toxicity of traditional cryoprotectants such as glycerol and dimethyl sulfoxide(DMSO)has raised safety issues for biomedical ***,a nonreducing disaccharide that accumulates in desiccation-or cold-tolerant organisms,has been considered as a biocompatible ***,a naturally occurring membrane-active cationic peptide,melittin,was utilized to facilitate membraneimpermeable trehalose entry into cells for effective cell ***(ethylene glycol)and poly(vinyl pyrrolidone)were applied as macromolecular protectants to improve the stabilization of cell *** the optimal protocol,the postthaw recovery of human red blood cells in freezing bags at a hematocrit of~50%could achieve 82.9%with favorable cell morphologies and physiological ***,the cryosurvival of L929 fibroblasts reached 84.3%compared to the conventional method using 10%(v/v)*** short,this work by using trehalose and melittin provides a biocompatible solvent-free approach for long-term cryostorage of cells.

cell cryopreservation has evolved as an important technology required for supporting various cell-based applications,such as stem cell therapy,tissue engineering,and assisted *** times have witnessed an increase in the clinical demand of these applications,requiring urgent improvements in cell ***,cryopreservation technology suff ers from the issues of low cryopreservation effi ciency and cryoprotectant(CPA)*** of advanced biotechnology tools can signifi cantly improve post-thaw cell survival and reduce or even eliminate the use of organic solvent CPAs,thus promoting the development of ***,based on the diff erent cryopreservation mechanisms available,we provide an overview of the applications and achievements of various biotechnology tools used in cell cryopreservation,including trehalose delivery,hydrogel-based cell encapsulation technique,droplet-based cell printing,and nanowarming,and also discuss the associated challenges and perspectives for future development.

Numerous studies have shown that cell replacement therapy can replenish lost cells and rebuild neural circuitry in animal models of Parkinson’s *** of midbrain dopaminergic progenitor cells is a promising treatment for Parkinson’s ***,transplanted cells can be injured by mechanical damage during handling and by changes in the transplantation ***,we developed a one-step biomanufacturing platform that uses small-aperture gelatin microcarriers to produce beads carrying midbrain dopaminergic progenitor *** beads allow midbrain dopaminergic progenitor cell differentiation and cryopreservation without digestion,effectively maintaining axonal integrity in ***,midbrain dopaminergic progenitor cell bead grafts showed increased survival and only mild immunoreactivity in vivo compared with suspended midbrain dopaminergic progenitor cell ***,our findings show that these midbrain dopaminergic progenitor cell beads enhance the effectiveness of neuronal cell transplantation.

Discussion on Serum Technique and cryopreservation Technique in the Culture of Liver Sinusoidal Endothelial cells

Hepatic sinusoidal endothelial cells are a kind of highly differentiated cells in hepatic sinusoids, which play an important role in the occurrence and development of liver fibrosis. Hepatic sinusoidal endothelial cells are often used in the study of liver fibrosis. In the process of culturing liver sinusoidal endothelial cells, the treatment of serum for culture and cryopreservation of cells are relatively complicated and error-prone. If the technical details of serum treatment and cell cryopreservation are not handled properly, it will have a more obvious effect on the effect of hepatic sinusoidal endothelial cell culture. We have gained experience in the theoretical study of liver fibrosis and the operation of cell culture experiments, and this paper summarized the details of liver sinusoidal endothelial cell culture such as the problems of serum preservation, thawing, precipitates and heat inactivation, as well as methods and precautions for cell cryopreservation.
