OBJECtIVE:to investigate the relationship between antiviral restriction factor Sterile Alpha Motif and Histidine-Aspartic acid domain-containing protein 1(SAMHD1)expression and t cell activation,furthermore,identifying objective indexes of lung-spleen Qi deficiency symptom ***:We assessed the profile of t lymphocyte subsets,characteristics of SAMHD1 and human leukocyte antigen DR(HLA-DR)expression in lungspleen Qi deficiency *** the same time,people living with human immunodeficiency virus/acquired immune deficiency syndrome(HIV/AIDS)(PLWHA)without obvious clinical symptoms and healthy donors in this area were used as ***:Immunohematologic indexes lower CD4 count,lower CD4/CD8 ratio and higher SAMHD1 level were found in lung-spleen Qi deficiency ***,we demonstrated a positive relationship between SAMHD1 and HLA-DR level as well as with interferon factor in lung-spleen Qi deficiency syndrome and patients without obvious clinical signs and symptoms ***:these data indicated the positive relationship between SAMHD1 and t cell activation which further elucidated the role of SAMHD1 in cellular immune ***,combination of t lymphocyte subsets counts and SAMHD1 level may be used as clinical and biological reference basis for the differentiation and diagnosis of HIV/AIDS traditional Chinese medicine syndromes.

Immunomodulatory effects and mechanisms of plant alkaloid tetrandrine in autoimmune diseases

Autoimmune diseases characterized by activation of immune effector cells and damage of target organs are currently treated with a combination of several disease-modifying antirheumatic drugs (DMARDs) that preserve different immunomodulatory mechanisms. Such a combination treatment strategy not only provides synergistic effects but also reduces side effects from individual drug. tetrandrine (tet), purified from a creeper Stephania tetrandra S Moore, is a bis-benzylisoquinoline alkaloid and has been used to treat patients with silicosis, autoim-mune disorders, and hypertension in China’s mainland for decades. the accumulated studies both in vitro and in vivo reveal that tet preserves a wide variety of immunosuppressive effects. Importantly, the tet-mediated immu-nosuppressive mechanisms are evidently different from some known DMARDs. the synergistic effects have also been demonstrated between tet and other DMARDs like FK506 and cyclosporin. these results highlight tet a very potential candidate to be consid

Effect of Xinfeng capsule on pulmonary function in a adjuvant arthritis rat model

Effect of cold-dryness on pulmonary and immunologic function in chronic obstructive pulmonary disease model rats

OBJECtIVE： to study the effects of cold-dryness on pulmonary and immunologic function of peripheral t-lymphocytes in chronic obstructive pulmonary disease （COPD） model rats, and to provide references for the prevention and treatment of cold-dryness COPD in the Xinjiang region. MEtHODS： the COPD model was established with an elastase drip into the trachea combined with smoking. the cold-dryness COPD model was developed by stressing with a cold-dry environment. Success of the model was determined by observation of pathologic lung sections. Rats were sacrificed by exsanguination from the femoral artery and changes of peripheral blood CD4＋, CD8＋, and CD4＋/CD8＋ were detected by flow cytometryo Data were analyzed with SAS 11.5 statistical software. RESULtS： On the ninetieth day after ending the ex- periment, Peak expiratory flow in the cold-dryness COPD group was lower than that in the COPD and normal control groups （P〈0.01）. the time of inspiration in the cold-dryness COPD group was higher than that in the COPD and normal groups （P〈0.05）. time of expiration （te） in the cold-dryness COPD group was higher than that in the COPD and normal groups （P〈0.01）. 50% tidal volume expiratory flow （EFS0） in the cold-dryness COPD group was lower than that in the COPD and normal groups （P〈 0.01）, and EFS0 in the COPD group was lower than that in the normal group （P〈0.05）. CD4＋ content of peripheral blood in the cold-dryness COPD group was lower than that in the COPD and the normal groups （P〈0.05）. CD8＋ content in the cold-dryness COPD and COPD groups was higher than that in the normal control group （P〈0.01）, and CD8＋ content in the cold-dryness COPD group was higher than that in the COPD group （P〈0.01）. CD4＋/CD8＋ in the cold-dryness COPD group and the COPD group was lower than that in the normal control group （P〈0.01）, and CD4＋/CD8＋ in the cold-dryness COPD group was lower than that in the COPD group （P〈0.05）. Hepatitis E virus chimeric DNA vaccine elicits immunologic response in mice

AIM： to construct the plasmid pcHEV23 containing fragments of HEV ORF2 and ORF3 chimeric gene and to assess its ability to elicit specific immunologic response in mice. MEtHODS： the gene encoding the structural protein of HEV ORF2 fragment and full-length ORF3 was amplified by PCR. the PCR products were cloned into an eucaryotic expression plasmid pcDNA3. the resulting plasmid pcHEV23 was used as a DNA vaccine to inoculate BALB/c mice intramuscularly thrice at a dose of 100 or 200 ug. Mice injected with empty pcDNA3 DNA or saline served as control and then specific immune responses in the mice were detected. RESULtS： After 2-3 times of inoculation, all mice injected with pcHEV23 had anti-HEV IgG seroconversion and specific t lymphocyte proliferation. the lymphocyte stimulation index in the group immunized with pcHEV23 （3.1＋0.49） was higher than that in the control group （0.787±0.12, P〈0.01）. None in the control group had a detectable level of anti-HEV IgG. CONCLUSION： DNA vaccine containing HEV ORF2 and ORF3 chimeric gene can successfully induce specific humoral and cellular immune response in mice.

Immunohistopathology of the contralateral testis of rats undergoing experimental torsion of the spermatic cord

Aim： to evaluate the immunohistopathological changes in the contralateral testis of rats after an experimental spermatic cord torsion. Methods： Male Sprague-Dawley rats of 45-50 days old were subjected to a 720° unilateral spermatic cord torsion for 10, 30 and 80 days （experimental group, E）, respectively or sham operation （control group, C）. Histopathology of the contralateral testis as well as germ cell apoptosis were studied using the terminal Deoxynucleotidyl transferase Biotin-dUtP Nick End Labeling （tUNEL） technique. the number of testicular lymphocytes, mast cells and macrophages, and the expression of tumor necrosis factor-α （tNF-α） and its receptor （tNFR1） in testicular cells of the contralateral testis were quantified by histochemistry and immunohistochemistry. tNF-α concentration in testicular fluid was determined by ELISA. Results： In the contralateral testis of rats from the E group, the maximal degree of damage of the germinal epithelium was seen 30 days after torsion. At this time we observed in the E group vs. the C group increases：（i） the number of testicular t-lymphocytes; （ii） the number of testicular mast cells and macrophages; （iii） the percentage of macrophages expressing tNF-α; （iv） tNF-α concentration in testicular fluid; （v） the number of apoptotic germ cells; and （vi） the number of tNFR1^＋ germ cells. Conclusion： Experimental spermatic cord torsion induces, in the contralateral testis, a focal damage of seminiferous tubules characterized by apoptosis and sloughing of germ cells. Results suggest humoral and cellular immune mediated testicular cell damage in which macrophages and mast cells seem to be involved in the induction of germ cell apoptosis through the tNF-α/tNFR1 system and in the modulation of the inflammatory process.

INtRODUCtIONthe t-cell dependent specific liver injury in mice induced by concanavalin A(ConA) is a newly cstablished experimental liver injury model,which is considered more eligible for the study on pathophysiology of several human liver discascs,such as viral hepatitis and autommune hepatitis[1-9].t cell activation and several cytokines release had been proven to play a critical role in ConA -induced liver injury[10-19].Cyclosprine A(CsA),an effective inhibitor of activation of t lymphocytc,hes been used widely in clinical treatment,especially in autoimmune diseases and organ transplantation[20-25].In this study,we investigated the possible effect of CsA on ConA-induced liver injury in Kunning mice.

Effect of compound Kushen injection on t-cell subgroups and natural killer cells in patients with locally advanced non-small-cell lung cancer treated with concomitant radiochemotherapy

OBJECtIVE: to observe effect of compound Kushen injection on t-cell subgroups and NK cells in patients with locally advanced non-small-cell lung cancer(NSCLC) treated with concomitant ***: We randomly divided 60 patients with locally advanced NSCLC who were treated at our hospital between May 2011 and May 2013 into a treatment group and a control group by *** treatment group(n = 30) received concomitant radiochemotherapy plus compound Keshen injection, and the control group(n = 30) received only ***: After treatment, levels of CD3+, CD4+,CD4 +/CD8 + and CD16 +/CD56 + cells had significantly increased, and CD8 + cells had significantly decreased, in the treatment group compared with both their pretreatment levels and with levels in the control group. In the control group, post-treatment levels of CD3 +, CD4 +, CD4 +/CD8 + and CD16+/CD56+ cells were not significantly changed from pretreatment levels. the two groups did not significantly differ in their rates of toxicity reactions(P > 0.05).CONCLUSION: Compound Kushen injections can increase immunologic function in patients with locally advanced non-small cell lung cancer who receive concomitant radiochemotherapy.
