In-vitro antioxidant and anti-inflammatory activities of ethanol stem-bark extract of Blighia sapida ***

Blighia sapida(***)***(Family Sapindaceae)is a branchless straight bole approximately 15 m in *** study evaluated the antioxidant and anti-inflammatory activities of ethanol extract and fractions of *** stem-bark using in vitro *** extract and its fractions were investigated for 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical scavenging activity,ferric reducing antioxidant power(FRAP),total antioxidant capacity(TAC),and quantitative phenolic and flavonoid ***-inflammatory activity was evaluated using albumin denaturation and membrane stabilization *** extract and its fractions exhibited radical scavenging and anti-inflammatory *** ethyl acetate fraction possessed maximum phenolic and flavonoid contents(136.67±1.55 gallic acid equivalent mg/g and 75.76±4.03 quercetin equivalent mg/g,respectively).Antioxidant studies revealed that the ethyl acetate fraction displayed superior activity with an IC_(50)=0.09±0.03 mg/mL DPPH,and values of 146.96±3.81 ascorbic acid equivalent(AAE)mg/g and 359.20±4.98 AAE mg/g for FRAP and TAC,***,the anti-inflammatory activity was revealed by inhibition of heatinduced albumin denaturation and red blood cell membrane stabilization at concentrations of 200 -1000 mg/mL and 50-250 mg/mL,*** ethanol extract and fractions exhibited antioxidant and anti-inflammatory activities,with ethyl acetate fraction showing superior activity,which could be attributed to secondary metabolites,mainly phenolic ***,the antioxidant and antiinflammatory activities of *** can be exploited by ethnomedicinal users.

Antithrombotic effects of the effective components group of Xiaoshuantongluo formula in vivo and in vitro

The present study was designed to investigate the antithrombotic effects and underlying mechanisms of the effective components group(ECG) of Xiaoshuantongluo recipe(XECG) and to further verify the rationality and feasibility of ECG-guided methodology in traditional Chinese medicine(TCM) research. The arterial thrombosis model induced by ferric chloride(FeC l3) oxidation and the venous thrombosis model induced by inferior vena cava ligation were established to evaluate the antithrombotic potential of XECG. Our results indicated that XECG significantly prolonged the time to occlusion, activated partial thromboplastin time(APTT), and prothrombin time(PT), and markedly inhibited adenosine diphosphate(ADP)-induced platelet aggregation in the 20% Fe Cl3-induced arterial thrombosis model. The superoxide dismutase(SOD) activity was significantly increased and the levels of malondialdehyde(MDA) and nitric oxide(NO) were dramatically decreased in the plasma of arterial thrombosis rats after XECG treatment for 12 days. Furthermore, XECG markedly reduced the weight of thrombus formed by inferior vena cava ligation. Additionally, XECG exhibited 2,2-diphenyl-1-picrylhydrazyl(DPPH) free radical scavenging activity and protective effect on mitochondrial lipid peroxidation. In summary, XECG played an important role in the prevention of thrombosis through interacting with multiple targets, including inhibition of platelet aggregation and coagulation and repression of oxidative stress. The ECG-guided methodology was validated as a feasible tool in TCM research.

Peroxynitrite scavenging Activities of Resveratrol and Piceid

In vitro antioxidant activities of resveratrol and piceid against peroxynitrite（ONOO-） were examined by the inhibition of 3-nitrotyrosine *** was used as a positive *** and piceid exhibited high ONOO--scavenging activities in a concentration dependent *** antioxidant activities（the concentration of test compound required to yield a 50% inhibition of tyrosine nitration,IC 50） of resveratrol and piceid against ONOO-were（48.34±0.97） and（74.69±1.49） μmol/L,*** with that of trolox[（105.40±1.16） μmol/L],their scavenging activities were 2.2-and 1.5-fold higher for resveratrol and *** of nitroresveratrol as shown by UV-Vis spectroscopy and liquid chromatography-tandom mass spectrometry（LC-MS/MS） analysis indicates that resveratrol could directly scavenge ONOO-via nitration *** results demonstrate that foods and medicinal herbs with resveratrol and piceid as stronger ONOO-scavengers are valuable ingredients and have healthy application in preventing humans from peroxynitrite-mediated oxidative damage by scavenging peroxynitrite efficiently.

Oxidative stress mitigation, kinetics of carbohydrate-enzymes inhibition and cytotoxic effects of flavonoids-rich leaf extract of Gazania krebsiana(Less.): An in vitro evaluation

Objective: To investigate the free radical scavenging, antidiabetic, kinetics of enzyme inhibition and cytotoxic potentials of flavonoid-rich extract of Gazania krebsiana leaves using standard methods. Methods: Antioxidant activity of the flavonoids was investigated at scavenging free radicals such as 1,1-diphenyl-2-picryl hydrazyl, nitric oxide, hydroxyl radical, reducing capabilities, 2,2-azinobis(3-ethylbenzothiazoline-6) sulphonic acid as well as metal chelating capability at different concentrations(125-1 000 μg/m L) while the antidiabetic activity was evaluated via the inhibition and kinetics of carbohydrate digestive enzymes including α-amylase, α-glucosidase, sucrase and maltase. Brine shrimp lethality assay was also employed to examine the cytotoxic effects at various concentrations(125-2 000 μg/m L). Results: The study showed the best antioxidant activity in 2,2-azinobis(3-ethylbenzothiazoline-6) sulphonic acid, hydroxyl radical, nitric oxide and metal chelating with IC50 values of(1 089.00±19.29),(719.40±5.35),(633.10±5.31),(1 116.00±22.92) μg/m L, respectively; as compared with butylated hydroxyl anisole(standard) with IC50 of 1 166, 747, 639 and 1 363 μg/m L, respectively. Similarly, strong inhibition of α-glucosidase as well as moderate inhibition of sucrase and maltase enzyme activities which were significantly(P<0.05) better than acarbose were also observed in the study; though acarbose activity against α-amylase was better than the extract. The kinetics or mode of inhibition of α-amylase by the extract revealed an uncompetitive inhibition, competitive for α-glucosidase and non-competitive inhibitions for both sucrase and maltase, while the result of the lethality assay showed a potent cytotoxic effect with LC50 value of 359 μg/m L. Conclusions: The results obtained from this study are suggestive of the antioxidative, antidiabetic and cytotoxic potentials of flavonoid-rich extract of Gazania krebsiana.

Protective effect of rhizome extracts of the herb, vacha (Acorus calamus) against oxidative damage: An in vivo and in vitro study

The rhizome of Acorus calamus,an herb widely used in Indian system of medicine for many ailments including epilepsy,mental illness and rheumatism,was subjected to soxhlet extraction to elucidate antioxidant property of different solvent extracts using in vitro *** benzene extract was most potent in scavenging hydroxyl and superoxide radicals and in reducing 1,1-diphenyl-2-picryl hydrazyl and ferric reducing antioxidant *** addition the benzene extract prevented oxidative damage to DNA and *** was also effective in preventing stress-induced decrease in total plasma anti-oxidant activity as determined in vivo using rat model wherein stress was induced by exposing to restraint and forced *** minimum effective dose of the benzene extract was 5 mg/kg body weight(oral),and at this dose,its effect was similar to the same dose of a standard anti-oxidant,ascorbic *** study for the first time,clearly demonstrates a potent anti-oxidant activity of *** combining in vitro and in vivo ***,the therapeutic value of this herb maybe due to its anti-oxidant property.©2016 Beijing Academy of Food *** and hosting by Elsevier *** rights reserved.

Study on Response Surface Methodology Optimization for Extraction Process of Cyclocarya paliurus Extract and Its Blood Sugar-lowering Effect

[Objectives]This study was conducted to discuss the mechanism of the blood sugar-lowering effect of Cyclocarya paliurus extract.[Methods]With *** leaves as the raw material,the ethyl acetate-butyl acetate microwave extraction process of *** was *** the yield of *** extract as an evaluation index,single-factor experiments were carried out on such 6 factors as the dry *** leaf powder size,solid-to-liquid ratio,extraction time,microwave frequency,microwave power,and microwave extraction time,and the extraction and purification process was further optimized by response surface *** normal mice and the hyperglycemic mice modeled by alloxan were injected with *** extract to study the blood sugar-lowering effects of different groups of *** extract.[Results]The optimal process combination for the extraction of *** leaves was obtained:dry *** powder size 90-100 mesh,solid-to-liquid ratio 1∶20(g/ml),extraction time 192 min,microwave frequency 2500 MHz,microwave power 490 W,and extraction time 248 *** these conditions,the yield of *** extract reached the maximum,95.10%,reaching 99.83%of the predicted *** blood sugar-lowering test on mice showed that the *** extract had a good effect on lowering blood sugar,indicating that *** extract can improve the free radical scavenging capacity to a certain extent.[Conclusions]This study provides certain reference for the in-depth study of the physiological effects of *** extract and the comprehensive development and utilization of *** extract.
