Protective effects of umbilical cord mesenchymal stem cell exosomes in a diabetic rat model through live retinal imaging

AIM:To assess the protective effect of human umbilical cord mesenchymal stem cell exosomes(hucMSC-Exs)in a diabetic rat model by using a variety of retinal ***:hucMSCs were subjected to differential ultracentrifugation for the collection of exosomes,and transmission electron microscopy(TEM),nanoparticle tracking analysis(NTA)using a NanoSight analysis system and Western blotting(WB)were used to analyze the expression of surface marker proteins such as CD63,CD9 and ***(STZ)was injected into the intraperitoneal cavity to establish a diabetic *** were divided into a normal group,diabetic group and hucMSC-Ex *** fluorescein angiography(FFA),optical coherence tomography(OCT)and other live imaging methods were used to observe the fundus of the ***,the eyeballs of rats from each group were collected for hematoxylin-eosin(HE)staining to further analyze the retinal ***:Through TEM,NTA and WB,we successfully isolated *** FFA and OCT confirmed that hucMSC-Exs effectively prevented early retinal vascular damage and thickening of the ***,HE staining of rat retinal sections revealed that exosomes effectively alleviated retinal structure disruption caused by ***:hucMSC-Exs have a protective effect on the retina in diabetic rat through FFA,OCT and HE staining.

Paclitaxel-etoposide-carboplatin/cisplatin versus etoposidecarboplatin/cisplatin as first-line treatment for combined small-cell lung cancer: a retrospective analysis of 62 cases

Objective: To compare the efficacy and adverse effects of paclitaxel-etoposide-carboplatin/cisplatin(TEP/TCE) regimen with those of etoposide-carboplatin/cisplatin(EP/CE) regimen as first-line treatment for combined small-cell lung cancer(CSCLC).Methods: A retrospective study was conducted on 62 CSCLC patients who were treated at tianjin Medical University Cancer Institute and Hospital from July 2000 to April 2013 and administered with TEP/TCE regimen(n=19) or EP/CE regimen(n=43) as first-line CSCLC treatment. All patients received more than two cycles of chemotherapy, and the response was evaluated every two cycles. The primary endpoint was overall survival(OS), and the secondary endpoints were progression-free survival(PFS), objective response rate(ORR), disease control rate(DCR), and adverse effects. Results: ORR between the TEP/TCE and EP/CE groups showed a statistical difference(90% vs. 53%, P=0.033). Both groups failed to reach a statistical difference in DCR(100% vs. 86%, P=0.212). The median PFS and OS of the TEP/TCE group were slightly longer than those of the EP/CE group, although both groups failed to reach a statistical difference(10.5 vs. 8.9 months, P=0.484; 24.0 vs. 17.5 months, P=0.457). However, stratified analysis indicated that the PFS of patients with stages III and IV CSCLC showed marginally significant difference between the TEP/TCE and EP/CE groups(19.5 vs. 7.6 months; P=0.071). Both rates of grade IV bone marrow depression and termination of chemotherapy in the TEP/TCE group were significantly higher than those in the EP/CE group(26.3% vs. 7.0%, P=0.036; 31.6% vs. 14.7%, P=0.004). Conclusion: The TEP/TCE regimen may not be preferred for CSCLC, and this three-drug regimen requires further exploration and research. To date, the EP/CE regimen remains the standard treatment for CSCLC patients.

In recent years,vehicle re‐identification has attracted more and more *** to learn the discriminative information from multi‐view vehicle images becomes one of the challenging problems in vehicle re‐identification *** example,when the viewpoint of the image changes,the features extracted from one image may be lost in another image.A two‐branch network with pyramid‐based local and spatial attention global feature learning(PSA)is proposed for vehicle re‐identification to solve this ***,one branch learns local features at different scales by building pyramid from coarse to fine and the other branch learns attentive global features by using spatial attention ***,pooling operation by using global maximum pooling(GMP)for local features and global average pooling(GAP)for global feature is ***,local feature vectors and global feature vector extracted from the last pooling layer,respectively,are employed for identity re‐*** experimental results demonstrate that the proposed method achieves state‐of‐the‐art results on the VeRi‐776 dataset and VehicleID dataset.

Surface Modification of Polycarbonate Urethane by Covalent Linkage of Heparin with a PEG Spacer

Heparin was grafted onto polycarbonate urethane (PCU) surface via a three-step procedure utilizing α, ωdiamino-poly(ethylene glycol) (APEG, M n =2 000) as a spacer. In the first step, isocyanate functional groups were introduced onto PCU surface by the treatment of hexamethylene diisocyanate (HDI) in the presence of di-n-butyltin dilaurate (DBTDL) as a catalyst. In the second step, APEG was linked to the PCU surface to obtain the APEG conjugated PCU surface (PCU-APEG). In the third step, heparin was covalently coupled with PCU-APEG in the presence of N-hydroxysuccinimide (NHS) and 1-ethyl-3-(3-dimethylamidopropyl) carbodiimide (EDAC). The amount of heparin (1.639 μg/cm 2 ) covalently immobilized on the PCU-APEG surface was determined by the toluidine blue method. The modified surface was characterized by water contact angle, attenuated total reflection Fourier transform infrared spectroscopy (ATR-FTIR), X-ray photoelectron spectroscopy (XPS), and atomic force microscopy (AFM). The hemocompatibility was preliminarily studied by platelet adhesion test. The results indicated that heparin was successfully grafted onto the PCU surface, and meanwhile the hydrophilicity and hemocompatibility of the modified PCU surface were improved significantly compared with the blank PCU surface.

Design and Analysis of a 0.01-to-6GHz 31dBm-P1dB 31.5%-PAE Distributed Power Amplifier in 0.25-μm GaAs technology

This paper presents the design and analysis of a distributed power amplifier with 6-d B bandwidth from 10 MHz to 6 GHz. To meet the stringent targeted specification, the concurrent design and analysis are carefully performed with optimizations in both passive and active devices. The gate capacitive division technique is proposed and proven theoretically of bandwidth extension effect and power efficiency enhancement. To validate the theory, a prototype is designed in a 0.25-μm Ga As technology. The fabricated amplifier chip is packaged in an evaluation cavity of SMA connectors. The measurement shows an average power gain of 15 d B, OP1 d Band PSAT of 31 d Bm and 32.6 d Bm at 3 GHz, and PAE at OP1 d Band PSAT points are 31.5% and 43.7% respectively. To the best of authors’ knowledge, the amplifier achieves the highest Power-added efficiency(PAE) among the similar Ga As amplifiers.

Effect of terahertz pulse on gene expression in human eye cells

In recent years, the advances in terahertz applications have stimulated interest in the biological effects associated with this frequency range. We study the gene expression profile in three types of cells exposed to terahertz radiation,i.e., human ARPE-19 retinal pigment epithelial cells, simian virus 40-transformed human corneal epithelial cells, and human MIO-M1 Müller cells. We find that the gene expression in response to heat shock is unaffected, indicating that the minimum temperature increases under controlled environment. The transcriptome sequencing survey demonstrates that 6-hour irradiation with a broadband terahertz source results in specific change in gene expression and also the biological functions that are closely related to these genes. Our results imply that the effect of terahertz radiation on gene expression can last over 15 hours and depends on the type of cell.

Objective:The immunoscore,which is used to quantify immune infiltrates,has greater relative prognostic value than tumor,node,and metastasis(TNM)stage and might serve as a new system for classification of colorectal ***,a comparable immunoscore for predicting lung adenocarcinoma(LUAD)prognosis is currently ***:We analyzed the expression of 18 immune features by immunohistochemistry in 171 *** relationship of immune marker expression and clinicopathologic factors to the overall survival(OS)was analyzed with the Kaplan-Meier method.A nomogram was developed by using the optimal features selected by least absolute shrinkage and selection operator(LASSO)regression in the training cohort(n=111)and evaluated in the validation cohort(n=60).Results:The indicators integrated in the nomogram were TNM stage,neuron-specific enolase,carcino-embryonic antigen,CD8 center of tumor(CT),CD8 invasive margin(IM),Fox P3 CT,and CD45 *** calibration curve showed prominent agreement between the observed 2-and 5-year OS and that predicted by the *** simplify the nomogram,we developed a new immune-serum scoring system(I-SSS)based on the points awarded for each factor in the *** I-SSS was able to stratify same-stage patients into different risk *** combination of I-SSS and TNM stage had better prognostic value than the TNM stage ***:Our new I-SSS can accurately and individually predict LUAD prognosis and may be used to supplement prognostication based on the TNM stage.
