Due to the lack of effective assessment method,overheated milk commodities are often marketed as pasteurized milk on the market,which was sold in high price by ***,this article aims to establish an approach based on metabolomics to monitor thermal processing temperature of bovine *** data of bovine milk samples heated at temperatures ranging from 60℃to 150℃were achieved by ultra-performance liquid chromatography-high-resolution mass spectrometry(UPLC-HRMS)platform,followed by multivariate data analysis.A regular variation pattern of chemical composition as temperature rises was pictured,furthermore.N^(∈)carboxymethyl lysine(CML),N^(∈)-carboxyethyl lysine(CEL),*** and lysinoalanine(LAL)were identified as 5 of the most contributed compounds to discriminate pasteurized and ultra-high-temperature(UHT)*** the comprehensive study on their content changes,we concluded that the optimal temperature range was 90-100℃for the generation of CML and CEL in this experiment,moreover.110~120℃for LAL.S0~100℃for pentosidine and 130-140℃for ***,a predicted rule to discriminate pasteurized and UHT milk was preliminarily established based on the ratios of CML/***/pentosidine and CML/*** could be applied in food labelling authentication of commercial bovine milk after further validation.

Mercury ion(Hg^(2+)),a highly noxious of heavy metalion,has detrimental effects on the ecological environment and human ***,we have developed an exonuclease III(Exo III)assisted catalytic hairpin assembly formation of a trivalent G-quadruplex/hemin DNAzyme for colorimetric detection of Hg^(2+).A hairpin DNA(Hr)was designed with thymine-Hg^(2+)-thymine pairs that catalyzed by Exo III is prompted to happen upon binding Hg^(2+).A released DNA fragment triggers the catalytic assembly of other three hairpins(H1,H2,and H3)to form many trivalent G-quadruplex/hemin DNA enzymes for signal *** developed sensor shows a dynamic range from 2 pM to 2μM,with an impressively low detection limit of 0.32 pM for Hg^(2+)*** a sensor also has good selectivity toward Hg^(2+)detection in the presence of other common metal *** strategy shows the great potential for visual detection with portable type.

Without considering the ecosystem-depen- dence of agricultural production, irrational use of agricultural technologies could bring only short-term economic benefits but leave long-term environmental deterioration. If some agricultural lands have to be abandoned because of these technologies such as chemical films or groundwater depletion, it will aggravate the burden of remaining lands for maintaining or enhancing production. Thus, agricultural production should be a part of public services, requiring the consideration of interests of different stakeholders and sustainability.

Laser-enhanced thermal effect of moderate intensity focused ultrasound on bio-tissues

For avoiding extra-damage to healthy tissues surrounding the focal point during high intensity focused ultrasound(HIFU) treatment in medical therapy, to reduce the ultrasonic intensity outside the focal point is expected. Thus, the heating processes induced by moderate intensity focused ultrasound(MIFU) and enhanced by combined irradiation of laser pulses for bio-tissues are studied in details. For fresh bio-tissues, the enhanced thermal effects by pulsed laser combined with MIFU irradiation are observed experimentally. To explore the mechanisms of these effects, several tissue-mimicking materials composed of agar mixed with graphite powders are prepared and studied for comparison, but the laser-enhanced thermal effects in these mimicking materials are much less than that in the fresh bio-tissues. Therefore, it is suggested that the laser-enhanced thermal effects may be mainly attributed to bio-activities and related photo-bio-chemical effects of fresh tissues.

Determination of Melamine in Dairy Products and Melamine Tableware by Inhibition Electrochemiluminescent Method

An electrochemiluminescent （ECL） method has been developed for the determination of melamine based on the inhibition of luminol ECL. A significant luminol ECL can be found at 1.47 V in the phosphate buffer solution at high pHs and low potential scan rates, this ECL signal can be inhibited obviously by melamine. The decrease of ECL intensity was linearly proportional to the logarithm of melamine concentration in the range of 1--100 ng/mL （R2~0.9911） and with the detection limit of 0.1 ng/mL. The method has been applied successfully to determine melamine in dairy products and melamine tableware, the recoveries were in the range of 98.5%--103.7% and 95.5%--106.0%, respectively. The mechanism of the inhibition effect was also proposed, the active oxygen （O2- ） generated from the electrooxidation of OH- reacted with luminol anion （L^-） to generate light emission, and the present of melamine can eliminate the active oxygen, which cause the decrease of the ECL intensity.

Eighteen sex hormones in antler velvet were determined by high performance liquid chromatography tandem mass *** solid phase extraction was applied to eliminating the matrix *** experimental conditions were examined and *** the optimal conditions,the proposed method provides the good linearities and determination limits（0.2―1.0 μg/kg） of the analytes *** recoveries ranging from 72.3% to 149.5% were obtained for the target analytes at two concentration *** method was applied to the determination of eighteen sex hormones in different kinds of antler velvet samples and the obtained results are *** results indicate that the proposed method is suitable for the determination of sex hormones in antler velvet samples.

Characterizing the antibiotic resistance genes in a river catchment： Influence of anthropogenic activities

Previous studies on environmental antibiotics resistance genes（ARGs） have focused on the pollution sources such as wastewater treatment plants, aquaculture and livestock farms,etc. Few of them had addressed this issue in a regional scale such as river catchment. Hence,the occurrence and abundances of 23 ARGs were investigated in surface water samples collected from 38 sites which located from the river source to estuary of the Beijiang *** them, 11 ARGs were frequently detected in this region and 5 ARGs（sul Ⅰ, sul Ⅱ, tet B,tet C, and tet W） were selected for their distribution pattern analysis. The abundances of the selected ARGs were higher in the upstream（8.70 × 10^6 copies/ng DNA） and downstream areas（3.17 × 10^6 copies/ng DNA） than those in the midstream areas（1.23 × 10^6 copies/ng DNA）, which was positively correlated to the population density and number of pollution sources. Pollution sources of ARGs along the Beijiang River not only had a great impact on the abundances and diversity, but also on the distribution of specific ARGs in the water samples. Both sul Ⅰ and sul Ⅱ were likely originated from aquaculture farms and animal farms,tet W gene was possibly associated with the mining/metal melting industry and the electric waste disposal and tet C gene was commonly found in the area with multiple pollution ***, the abundance of tet B was not particularly related to anthropogenic impacts. These findings highlight the influence of pollution sources and density of population on the distribution and dissemination of ARGs at a regional scale.

The sting of red imported fire ant （RIFA） could cause serious allergic response in fraction of people. These allergic reactions are mainly caused by its venom, especially venom allergen Sol i 1-4. To produce large amount of RIFA venom allergen Sol i 4 for diagnosis of RIFA allergy and allergen-specific immunotherapy, the gene encoding this protein was amplified and cloned into the prokaryotic expression vector pET43, la. The recombinant plasmid was used to transform competent cells and the recombinant proteins were expressed in E. coll. SDS-PAGE and Western blotting analysis indicated that high-level expression of Sol i 4 protein was successfully achieved. Allergenic activity analysis of the recombinant allergen Sol i 4 was then performed on rabbit. The result showed that the recombinant protein obtained had significant allergenic activity. It indicated that the recombinant allergen Sol i 4 of RIFA venom was successfully expressed in E. coli, which provided foundation for further developing therapeutic and diagnosis reagents of RIFA allergy.

Primary aromatic amines （PAAs） are substances with toxicity and suspected human carcinogenicity. A facile method for highly sensitive detection of PAAs using surface-enhanced Raman spectroscopy （SERS） is reported. The immobilization of Au nanoparticles （AuNPs） on the glycidyl methacrylate-ethylene dimethacrylate （GMA-EDMA） materials makes the suhstrate a closely packed but not aggregated Au NP arrays which provides a prominent SERS enhancement. Four PAAs with different substituent groups, namely, p-toluidine, p-nitroaniline, benzidine and 4,4-methylene-bis-（2-chloroaniline） have been successfully identified and quantified. High sensitivity and good linear relationship between SERS signals and concentrations of PAAs are obtained for all four PAAs.

Stable Isotope Dilution Analysis of Gibberellin Residues in Tomato Paste by Liquid Chromatography-Tandem Mass Spectrometry

An accurate and sensitive method for the simultaneous determination of gibberellic acid（GA3）, gibberellin A4（GA4） and gibberellin A7（GA7） residues in tomato paste was developed by coupling solid phase extraction to high performance liquid chromatography-tandem mass spectrometry（LC-MS/MS） with electrospray ionization based stable isotope dilution analysis（SIDA）. The isotope labeled internal standard can compensate for the losses during the extraction and cleanup steps and for discrimination due to ion suppression. After extraction from methanol, hydrophile lipophilic balance（HLB） solid phase extraction（SPE） column was tested for the capacity of the cleanup of the tomato paste in compared with C18 SPE column which is the common way to the detection of GAs, and the former gained better result. Spiked experiments were performed in the non-contaminated tomato pastes and the recoveries of GA3, GA4 and GA7 were 42.6%―75.0% in external standard method（ESM） and 91.1%―103.8% in internal standard method（ISM） respectively. The validities of this method were investigated and good analytical performance for the three GAs was obtained, including low limits of method detection（2 ng/g for GA3 and GA4, 0.3 ng/g for GA7）, excellent linear dynamic ranges（5―500 ng/g for GA3 and GA4, 1―100 ng/g for GA7） and good relative standard deviation ranges（4.8%―9.4% for the intra-day test and 3.5%―11.9% for the inter-day test）.
