The persistent phosphor SrAl_(2)O_(4):eu^(2+),Dy^(3+)is the subject of numerous *** often neglected aspect is that in this phosphor,as well as in Sr_(4)Al_(14)O_(25):eu^(2+),Dy^(3+),there are two different Sr^(2)+sites which can be occupied by the dopant eu^(2+)ions,We first introduce a general scheme of possible energy transfers in these persistent phosphor materials including explicitly both europium ions,This scheme is used as a generic starting point to study experimentally specific *** illustrate this application with the study of the effect of excitation wavelength(444 and 382 nm)on the afterglow of differently doped SrAl_(2)O_(4):eu^(2+),Dy^(3+)samples,as well as on the emission decay *** the same excitation intensity under 444 nm excitation,the resulting afterglow intensity is stronger than under near UV *** 382 nm,eu^(2+)ions on both Sr^(2)+s ites in SrAl_(2)O_(4)are excited,but at room temperature the blue emission is quenched,leading to a loss of *** observed effects can further be associated with the ratio of eu^(2+)ions and trap states which are modulated by the concentrations of eu^(2+)and Dy^(3+)in SrAl_(2)O_(4),as well as by temperature,Increasing the nominal Dy^(3+)content from 0.1 mol%to 0.5 mol%with respect to Sr results in the doubling of the integrated afterglow intensity and confirms thus that Dy^(3+)ions are indeed involved in the trapping process,The concentration of trap states is much lower than the concentration of eu^(2+)ions,as even with low excitation densities,a plateau of integrated afterglow intensity(corresponding to the total number of accessible traps)is *** postulate that an important fraction of excited eu^(2+)ions can potentially transfer their energy to trap *** that all traps are filled or in a dynamical filling-depletion process under illumination(with thermal and/or optical depletion processes),for the remaining eu^(2+)a"normal"steady-state emission is observed

Liver sinusoidal endothelial and biliary cell repopulation following irradiation and partial hepatectomy

AIM: To investigate whether irradiation (IR) and partial hepatectomy (Ph) may prepare the host liver for nonparenchymal cell (NPC) ***: Livers of dipeptidyl peptidase(DPP)-deficient rats were pre-conditioned with external beam IR (25 Gy) delivered to two-thirds of the right liver lobules followed by a one-third Ph of the untreated lob-ule. DPP-positive liver cells (NPC preparations enriched for liver sinusoidal endothelial cells (LSeCs) and hepatocytes) were transplanted via the spleen into the recipient livers. The extent and quality of donor cell engraftment and growth was studied over a long-term interval of 16 wk after ***: host liver staining demonstrated 3 different repopulation types. Well def ined clusters of donor-derived hepatocytes with canalicular expression of DPP were detectable either adjacent to or in between large areas of donor cells (covering up to 90% of the section plane) co-expressing the endothelial marker platelet endothelial cell adhesion molecule. The third type consisted of formations of DPP-positive duct-like structures which co-localized with biliary epithelial ***: Liver IR and Ph as a preconditioning stimulus enables multiple cell liver repopulation by donor hepatocytes, LSeCs, and bile duct cells.

Fatty acid oxidation and autophagy promote endoxifen resistance and counter the effect of AKT inhibition in eR-positive breast cancer cells

Tamoxifen(TAM)is the first-line endocrine therapy for estrogen receptor-positive(eR+)breast cancer(BC).however,acquired resistance occurs in∼50%***,although the PI3K/AKT/mTOR pathway is a viable target for treatment of endocrine therapy-refractory patients,complex signaling feedback loops exist,which can counter the effectiveness of inhibitors of this ***,we analyzed signaling pathways and metabolism in eR+MCF7 BC cell line and their TAM-resistant derivatives that are co-resistant to endoxifen using immunoblotting,quantitative polymerase chain reaction,and the Agilent Seahorse XF *** found that activation of AKT and the energy-sensing kinase AMPK was increased in TAM and endoxifen-resistant ***,eRRα/PGC-1βand their target genes MCAD and CPT-1 were increased and regulated by AMPK,which coincided with increased fatty acid oxidation(FAO)and autophagy in TAM-resistant *** of AKT feedback-activates AMPK and eRRα/PGC-1β-MCAD/CPT-1 with a consequent increase in FAO and autophagy that counters the therapeutic effect of endoxifen and AKT ***,our results indicate increased activation of AKT and AMPK with metabolic reprogramming and increased autophagy in TAM-resistant *** inhibition of AKT and FAO/autophagy is necessary to fully sensitize resistant cells to endoxifen.

evaluation of disinfection byproducts for their ability to affect mitochondrial function

In the race to deliver clean water to communities through potable water reuse, disinfection and water quality assessment are and will continue to be fundamental factors. There are over 700 disinfection byproducts(DBPs) in water;evaluating each compound is practically impossible and very time consuming. A bioanalytical approach could be an answer to this challenge. In this work, the response of four major classes of DBPs toward mitochondrial membrane potential( Δψm) and cytoplasmic adenosine triphosphate(C-ATP) was investigated with human carcinoma(hep G2) cells. Within 90 min of cell exposure, only the haloacetic acid(hAA) mixture caused a cytotoxic response as measured by C-ATP. All four groups(haloacetonitriles(hans), trihalomethanes(ThMs), nitrosamines(NOAs), and hAAs) responded well to Δψm, R 2 > 0.70. Based on the half-maximum concentration that evoked a 50% response in Δψm, the response gradient was hans >> hAAs ~ ThM > NOAs. The inhibition of the Δψm by hans is driven by dibromoacetonitrile(DBAN), while dichloroacetonitrile(DCAN) did not cause a significant change in the Δψm at less than 2000 μM. A mixture of hans exhibited an antagonistic behavior on the Δψm compared to individual compounds. If water samples are concentrated to increase hAN concentrations, especially DBAN, then Δψm could be used as a biomonitoring tool for DBP toxicity.

Novel technique of insulin loading into porous carriers for oral delivery

The increasing demand for oral macromolecule delivery encouraged the development of mi-croencapsulation technologies to protect such drugs against gastric and enzymatic degra-dation. however, microencapsulation often requires harsh conditions that may jeopardizetheir biological activity. Accordingly, many trials attempted to load macromolecules intoporous drug carriers to bypass any formulation induced instability. In this study, we pre-pared chitosan coated porous poly(d, l-lactide-co-glycolide)(PLGA) microparticles(MPs)loaded with insulin using a novel loading technique; double freeze-drying. The resultsshowed a significant increase in drug loading using only 5 mg/ml initial insulin concen-tration and conveyed a sustained drug release over uncoated MPs. Furthermore, SeM andconfocal microscopy confirmed pore blocking and insulin accumulation within the MPs re-spectively. The oral pharmacodynamic data on rats also proved the preservation of insulinbioactivity after formulation. Finally, the new coating technique proved to be efficient inproducing robust layer of chitosan with higher insulin loading while maintaining insulinactivity.

Genetic assessment of inbred chicken lines indicates genomic signatures of resistance to Marek's disease

Background: Marek’s disease(MD) is a highly contagious pathogenic and oncogenic disease primarily affecting chickens. however, the mechanisms of genetic resistance for MD are complex and not fully understood. MD-resistant line 63and MD-susceptible line 72are two highly inbred progenitor lines of White Leghorn. Recombinant Congenic Strains(RCS) were developed from these two lines, which show varied susceptibility to ***: We investigated genetic structure and genomic signatures across the genome, including the line 63and line72, six RCSs, and two reciprocally crossed flocks between the lines 63and 72(F1 63× 72and F1 72× 63) using Affymetrix~? Axiom~? hD 600 K genotyping array. We observed 18 chickens from RCS lines were specifically clustered into resistance sub-groups distributed around line 63. Additionally, homozygosity analysis was employed to explore potential genetic components related to MD resistance, while runs of homozygosity(ROh) are regions of the genome where the identical haplotypes are inherited from each parent. We found several genes including SIK, SOX1, LIG4, SIK1 and TNFSF13B were contained in ROh region identified in resistant group(line 63and RCS), and these genes have been reported that are contribute to immunology and survival. Based on FSTbased population differential analysis, we also identified important genes related to cell death and anti-apoptosis, including AKT1, API5, CDh13, CFDP and USP15,which could be involved in divergent selection during inbreeding ***: Our findings offer valuable insights for understanding the genetic mechanism of resistance to MD and the identified genes could be considered as candidate biomarkers in further evaluation.

TRPC3 is required for the survival, pluripotency and neural differentiation of mouse embryonic stem cells(meSCs)

Transient receptor potential canonical subfamily member 3(TRPC3) is known to be important for neural development and the formation of neuronal networks. here, we investigated the role of TRPC3 in undifferentiated mouse embryonic stem cells(meSCs) and during the differentiation of meSCs into neurons. CRISPR/Cas9-mediated knockout(KO) of TRPC3 induced apoptosis and the disruption of mitochondrial membrane potential both in undifferentiated meSCs and in those undergoing neural differentiation. In addition, TRPC3 KO impaired the pluripotency of meSCs. TRPC3 KO also dramatically repressed the neural differentiation of meSCs by inhibiting the expression of markers for neural progenitors, neurons, astrocytes and *** together, our new data demonstrate an important function of TRPC3 with regards to the survival, pluripotency and neural differentiation of meSCs.

Tetracycline-inducible protein expression in pancreatic cancer cells: effects of CapG overexpression

AIM: To establish stable tetracycline-inducible pancreatic cancer cell lines. MeThODS: Suit-2, MiaPaca-2, and Panc-1 cells were transfected with a second generation reverse tetracycline-controlled transactivator protein (rtTA2S-M2), under the control of either a cytomegalovirus (CMV) or a chicken β-actin promoter, and the resulting clones were ***: Use of the chicken (β-actin) promoter proved superior for both the production and maintenance of doxycycline-inducible cell lines. The system proved versatile, enabling transient inducible expression of a variety of genes, including GST-P, CYP2e1, S100A6, and the actin capping protein, CapG. To determine the physiological utility of this system in pancreatic cancer cells, stable inducible CapG expressors were established. Overexpressed CapG was localised to the cytoplasm and the nuclear membrane, but was not observed in the nucleus. high CapG levels were associated with enhanced motility, but not with changes to the cell cycle, or cellular proliferation. In CapG-overexpressing cells, the levels and phosphorylation status of other actin-moduating proteins (Cofilin and ezrin/Radixin) were not altered. however, preliminary analyses suggest that the levels of other cellular proteins, such as ornithine aminotransferase and enolase, are altered upon CapG induction. CONCLUSION: We have generated pancreatic-cancer derived cell lines in which gene expression is fully controllable.
