Community structure of Lumbricidae in beech woodland of the Bieszczady National Park, Southeast Poland

As ecosystem engineers,earthworms play a key role in the soil ***,due to increasing anthropogenic pressure,soil organisms,including earthworms,are being threatened by habitat *** this study,we undertook a qualitative and quantitative investigation of earthworms of the family Lumbricidae in four types of Carpathian beech woodland(Fagetum carpaticum),characterized by their understory vegetation(I,*** drymejae;Ⅱ,***;Ⅲ,***;and IV,***),in the Bieszczady National Park(Eastern Carpathians,Southeast Poland).At each investigated site,soil monoliths(25 cm×25 cm×25 cm)were examined by hand *** were expelled from deep soil layers using a weak formalin solution(0.4%).Depending on the phytocoenosis,7 species of Lumbricidae were identified at each of the sites I,Ⅱ,andⅢand 10 at site *** IV(***)differed significantly(P<0.05)from the other three sites with respect to earthworm biomass(59.71±39.53 g m^(-2))and the Shannon-Wiener diversity index(0.52±0.12).Although present three decades ago,the deep-burrowing species Octodrilus transpadanus and the litter-dwelling species Dendrobaena octaedra were not found at site IV in the present *** suspect that these two species may have been displaced by the invasive Lumbricus terrestris,which was not found at the same site in the *** observations warrant further investigation to verify the predicted effects of an invasive earthworm,including the potential effects on soils and other fauna and flora,which have been documented in numerous countries.

Mapping the metabolic responses to oxaliplatin-based chemotherapy with in vivo spatiotemporal metabolomics

Adjuvant chemotherapy improves the survival outlook for patients undergoing operations for lung metastases caused by colorectal cancer (CRC). However, a multidisciplinary approach that evaluates several factors related to patient and tumor characteristics is necessary for managing chemotherapy treatment in metastatic CRC patients with lung disease, as such factors dictate the timing and drug regimen, which may affect treatment response and prognosis. In this study, we explore the potential of spatial metabolomics for evaluating metabolic phenotypes and therapy outcomes during the local delivery of the anticancer drug, oxaliplatin, to the lung. 12 male Yorkshire pigs underwent a 3 h left lung in vivo lung perfusion (IVLP) with various doses of oxaliplatin (7.5, 10, 20, 40, and 80 mg/L), which were administered to the perfusion circuit reservoir as a bolus. Biocompatible solid-phase microextraction (SPME) microprobes were combined with global metabolite profiling to obtain spatiotemporal information about the activity of the drug, determine toxic doses that exceed therapeutic efficacy, and conduct a mechanistic exploration of associated lung injury. Mild and subclinical lung injury was observed at 40 mg/L of oxaliplatin, and significant compromise of the hemodynamic lung function was found at 80 mg/L. This result was associated with massive alterations in metabolic patterns of lung tissue and perfusate, resulting in a total of 139 discriminant compounds. Uncontrolled inflammatory response, abnormalities in energy metabolism, and mitochondrial dysfunction next to accelerated kynurenine and aldosterone production were recognized as distinct features of dysregulated metabolipidome. Spatial pharmacometabolomics may be a promising tool for identifying pathological responses to chemotherapy.

Vapour Treatment Method Against Other Pyro-and Hydrometallurgical Processes Applied to Recover Platinum From Used Auto Catalytic Converters

Today more and more cars are produced every year. All of them have to be equipped with catalytic converters, the main role of which is to obtain substances harmless to the environment instead of exhausted gases. Catalytic converters contain platinum group metals （PGM） especially platinum, palladium and rhodium. The price of these metals and their increasing demand are the reasons why today it is necessary to recycle used auto catalytic converters. There are many available methods of recovering PGM metals from them, especially platinum. These methods used mainly hydrometallurgical processes; however pyrometallurgical ones become more and more popular. The article presents results of the research mainly concerning pyrometallurgical processes. Two groups of research were carried out. In the first one different metals such as lead, magnesium and copper were used as a metal collector. During the tests, platinum went to those metals forming an alloy. In other research metal vapours were blown through catalytic converter carrier （grinded or whole）. In the tests metals such as calcium, magnesium, cadmium and zinc were applied. As a result white or grey powder （metal plus platinum） was obtained. The tables present results of the research. Processing parameters and conclusions are also shown. To compare efficiency of pyrometallurgical and hydrometallurgical methods catalytic converter carrier and samples of copper with platinum obtained from pyrometallurgical method were solved in aqua regia, mixture of aqua regia and fluoric acid.

Characterization and QTL identification in eggplant introgression lines under two N fertilization levels

The long-term immunological effects of alloferon and its analogues in the mealworm Tenebrio mofitor

The subject of this article is a search for the long-term immunological effects of alloferon and 3 structural analogues of alloferon, which were earlier characterized by the highest pro-apoptotic activity in Tenebrio molitor. The differences in the actions of these peptides on immune response were observed. Alloferon increased nodulation and significantly phenoloxidase activity in the hemolymph of experimentally infected T. molitor. However, [Phe（p-NH2）^1 ]- and [Phe（p-OMe） ^1 ]-alloferon strongly inhibited cellular and humoral defense of the mealworm against Staphylococcus aureus infection. One day after injection of these peptides, the specific biochemical and morphological hallmarks of apoptosis in bacteria-challenged hemocytes were visible; in contrast, 3 days after peptides injection in all hemocytes, caspase activation was not observed. However, these new, circulating hemocytes differed from the control and the peptide-untreated bacteria-challenged hemocytes. They had an increased adhesion that led to a separation of viable, anucleated fragments of hemocytes that retain the ability to adhere and to form long filopodia. The peptide-induced separation ofhemocyte fragments may resemble the formation ofplatelets in mammals and perhaps play a role in sealing wounds in insects. The results of in vivo studies may suggest a long half-life of studied peptides in the hemolymph of mealworm. Moreover, we showed the importance of the N-terminal histidine residues at position one of the alloferon molecule for its immunological properties in insects. The results obtained here show that alloferon plays pleiotropic functions in insects.

Evaluation of three sets of advanced backcrosses of eggplant with wild relatives from different gene pools under low N fertilization conditions

The development of new cultivars with improved nitrogen use efficiency(NUE)is key for implementing sustainable agriculture *** wild relatives(CWRs)provide valuable genetic resources for breeding programs aimed at achieving this *** this study,three eggplant(Solanum melongena)accessions together with their advanced backcrosses(ABs;BC3 to BC5 generations)were evaluated for 22 morpho-agronomic,physiological,and NUE traits under low nitrogen(LN)fertilization *** ABs were developed with introgressions from the wild relatives Solanum insanum,Solanum dasyphyllum,and Solanum *** AB population comprised a total of 25,59,and 59 genotypes,respectively,with overall donor wild relative genome coverage percentages of 58.8%,46.3%,and 99.2%.The three *** recurrent parents were also evaluated under control(normal)N *** of N fertilization in the parents resulted in decreased chlorophyll content-related traits,aerial biomass,stem diameter,and yield and increased NUE,nitrogen uptake efficiency(NUpE),and nitrogen utilization efficiency(NUtE).However,the decrease in yield was moderate,ranging between 62.6%and 72.6%.A high phenotypic variation was observed within each of the three sets of ABs under LN conditions,with some individuals displaying improved transgressive characteristics over the recurrent *** the single primer enrichment technology 5 k probes platform for high-throughput genotyping,we observed a variable but high degree of recurrent parent genome recovery in the ABs attributable to the lines recombination,allowing the successful identification of 16 quantitative trait loci(QTL).Different allelic effects were observed for the introgressed QTL *** candidate genes were identified in the QTL regions associated with plant growth,yield,fruit size,and NUE-related *** results show that eggplant materials with introgressions from CWRs can result in a dramatic impact in eggplant breedi

MYB34, MYB51, and MYB122 transcription factors are known as decisive regulators of indolic glucosinolate （IG） biosynthesis with a strong impact on expression of genes encoding CYP79B2 and CYP79B3 enzymes that redundantly convert tryptophan to indole-3-acetaldoxime （IAOx）. This intermediate represents a branching point for IG biosynthesis, and pathways leading to camalexin and indole-carboxylic acids （ICA）. Here we investigate how these MYBs affect the pathogen-triggered Trp metabolism. Our experiments indicated that these three MYBs affect not only IG production but also constitutive biosynthesis of other IAOx- derived metabolites. Strikingly, the PENETRATION 2 （PEN2）-dependent IG-metabolism products, which are absent in myb34/51/122 and pen2 mutants, were indispensable for full flg22-mediated induction of other IAOx-dedved compounds. However, germ induction and accumulation of ICAs and camalexin upon path- ogen infection was not compromised in myb34/51/122 plants, despite strongly reduced IG levels. Hence, in comparison with cyp79B2/B3, which lacks all IAOx-derived metabolites, we found myb34/51/122 an ideal tool to analyze IG contribution to resistance against the necrotrophic fungal pathogen Plectosphaerella cucumerina. The susceptibility of myb34/51/122 was similar to that of pen2, but much lower than susceptibility of cyp79B2/B3, indicating that MYB34/51/122 contribute to resistance toward P. cucumerina exclu- sively through IG biosynthesis, and that PEN2 is the main leaf myrosinase activating IGs in response to microbial pathogens.
