Per-and polyfluoroalkyl substances(PFAS)are persistent organic pollutants of concern be-cause of their ubiquitous presence in surface and ground water;analytical methods that can be used for rapid comprehensive exposure assessment and fingerprinting of PFAS are *** the fires at the Intercontinental Terminals Company(ITC)in deer Park,TX in 2019,large quantities of PFAS-containing firefighting foams were *** release of these substances into the Houston Ship Channel/Galveston Bay(HSC/GB)prompted con-cerns over the extent and level of PFAS contamination.A targeted liquid chromatography-tandem mass spectrometry(LC-MS/MS)-based study of temporal and spatial patterns of PFAS associated with this incident revealed presence of 7 species;their levels gradually de-creased over a 6-month *** the targeted LC-MS/MS analysis was focused on about 30 PFAS molecules,it may have missed other PFAS compounds present in firefighting ***,we utilized untargeted LC-ion mobility spectrometry-mass spectrome-try(LC-IMS-MS)-based analytical approach for a more comprehensive characterization of PFAS in these water *** analyzed 31 samples from 9 sites in the HSC/GB that were collected over 5 months after the *** data showed that additional 19 PFAS were detected in surface water of HSC/GB,most of them decreased gradually after the *** features detected by LC-MS/MS correlated well in abundance with LC-IMS-MS data;however,LC-IMS-MS identified a number of additional PFAS,many known to be compo-nents of firefighting *** findings therefore illustrate that untargeted LC-IMS-MS improved our understanding of PFAS presence in complex environmental samples.

The importance of timely diagnosis of aspirin-exacerbated respiratory disease for patient health and safety

Backgroud:Aspirin-exacerbated respiratory disease(AERd)is a difficult-to-treat syndrome where timely diagnosis and initiation of disease-specific therapies are pertinent to improved patient ***:To characterize the most common timeline for development of the clinical triad[asthma,nasal polyposis,and reactions to nonsteroidal anti-inflammatory drugs(NSAIds)],identify barriers to prompt diagnosis of AERd,and describe indications for an aspirin challenge to facilitate accurate ***:Six hundred ninety-seven patients with diagnosed AERd and history of at least one sinus surgery to remove nasal polyps were identified in the Brigham and Women’s Hospital AERd *** reported age at disease onset of asthma,nasal polyposis,and age of first NSAId reaction were obtained from 2013 to 2019 at ***:Of the 697 patients identified,diagnosis of asthma preceded diagnosis of nasal polyposis and first NSAId reaction,although there was considerable variability between ***:Prompt diagnosis of AERd is important for patient and provider education and improved care of this difficult-to-treat population of *** diagnostic aspirin challenge in patients without historical reactions to NSAIds who have an otherwise compatible clinical history,specifically in patients who take daily low-dose aspirin,leukotriene modifiers,avoid NSAIds,or who are severely symptomatic at baseline where it would be difficult to identify an acute worsening of symptoms.

Human enteric viruses–potential indicators for enhanced monitoring of recreational water quality

Recreational waters contaminated with human fecal pollution are a public health concern, and ensuring the safety of recreational waters for public use is a priority of both the Environmental Protection Agency(EPA) and the Centers for disease Control and Prevention(CdC). Current recreational water standards rely on fecal indicator bacteria(FIB) levels as indicators of human disease risk. However present evidence indicates that levels of FIB do not always correspond to the presence of other potentially harmful organisms, such as viruses. Thus, enteric viruses are currently tested as water quality indicators, but have yet to be successfully implemented in routine monitoring of water quality. This study utilized enteric viruses as possible alternative indicators of water quality to examine 18 different fresh and offshore recreational waters on O‘ahu, Hawai‘i, by using newly established laboratory techniques including highly optimized PCR, real time PCR, and viral infectivity assays. All sample sites were detected positive for human enteric viruses by PCR including enterovirus, norovirus genogroups I and II, and male specific FRNA coliphage. A six time-point seasonal study of enteric virus presence indicated significant variation in virus detection between the rainy and dry seasons. Quantitative PCR detected the presence of norovirus genogroup II at levels at which disease risk may occur, and there was no correlation found between enteric virus presence and FIB counts. Under the present laboratory conditions, no infectious viruses were detected from the samples PCR-positive for enteric viruses. These data emphasize both the need for additional indicators for improved monitoring of water quality, and the feasibility of using enteric viruses as these indicators.

An integrative multivariate approach for predicting functional recovery using magnetic resonance imaging parameters in a translational pig ischemic stroke model

Magnetic resonance imaging(MRI)is a clinically relevant,real-time imaging modality that is frequently utilized to assess stroke type and ***,specific MRI biomarkers that can be used to predict long-term functional recovery are still a critical ***,the present study sought to examine the prognostic value of commonly utilized MRI parameters to predict functional outcomes in a porcine model of ischemic *** was induced via permanent middle cerebral artery *** 24 hours post-stroke,MRI analysis revealed focal ischemic lesions,decreased diffusivity,hemispheric swelling,and white matter *** deficits including behavioral abnormalities in open field and novel object exploration as well as spatiotemporal gait impairments were observed at 4 weeks *** graphical models identified specific MRI outputs and functional recovery variables,including white matter integrity and gait performance,that exhibited strong conditional *** correlation analysis revealed a prognostic relationship between lesion volume and white matter integrity and novel object exploration and gait ***,these analyses may also have the potential of predicting patient recovery at chronic time points as pigs and humans share many anatomical similarities(e.g.,white matter composition)that have proven to be critical in ischemic stroke *** study was approved by the University of Georgia(UGA)Institutional Animal Care and Use Committee(IACUC;Protocol Number:A2014-07-021-Y3-A11 and 2018-01-029-Y1-A5)on November 22,2017.

Androgen and estrogen receptors in placental physiology and dysfunction

BACKGROUNd： The placenta is recognized as an endocrine organ, largely due to its secretions of steroid hormones, including progesterone, androgens, and estrogens. Steroid hormones play an essential role in the progression of pregnancy, fetal development, and growth. Furthermore, steroids are necessary for establishment and maintenance of a normal pregnancy, preparing the endometrium for implantation, stimulating endometrial secretions, and regulating uterine blood flow, however the exact mechanism of sex steroid signaling through their receptors in placental function is unknown. OBJECTIVE： In this review, we will provide an overview of the current knowledge on sex steroid receptors in normal placental development, as well as evidence of abnormal signaling associated with placental dysfunction. METHOdS： A systematic literature search was performed using the NCBI PubMed search engine, including the following key works： estrogen receptor, androgen receptor, placenta, placental development, cytotrophoblast, and differentiation. RESULTS： Of the over 700 articles that were returned, 125 studies focused on estrogen and androgen receptors in human placenta development and function during normal and abnormal pregnancy, as well as in rodents and ruminants placentae. CONCLUSION： Receptors for both estrogens and androgens have been localized within the mammalian placenta, but surprisingly little is known about their signaling in trophoblast cell differentiation and function. An emerging picture is developing in which estrogen receptors possibly play role in cytotrophoblast proliferation and extravillous trophoblast invasion, whereas androgen receptors are involved in syncytiotrophoblast differentiation and function.

Mining of plant-derived antimicrobials is the major focus at current to counter antibiotic resistance. This study was conducted to characterize the antimicrobial activity and mode of action of linalyl anthranilate(LNA) against carbapenemase-producing Klebsiella pneumoniae(KPC-KP). LNA alone exhibited bactericidal activity at 2.5%(V/V), and in combination with meropenem(MPM) at 1.25%(V/V). Comparative proteomic analysis showed a significant reduction in the number of cytoplasmic and membrane proteins,indicating membrane damage in LNA-treated KPC-KP cells. Up-regulation of oxidative stress regulator proteins and down-regulation of oxidative stress-sensitive proteins indicated oxidative stress. Zeta potential measurement and outer membrane permeability assay revealed that LNA increases both bacterial surface charge and membrane permeability. Ethidium bromide influx/efflux assay showed increased uptake of ethidium bromide in LNA-treated cells, inferring membrane damage. Furthermore, intracellular leakage of nucleic acid and proteins was detected upon LNA treatment. Scanning and transmission electron microscopies again revealed the breakage of bacterial membrane and loss of intracellular materials. LNA was found to induce oxidative stress by generating reactive oxygen species(ROS) that initiate lipid peroxidation and damage the bacterial membrane. In conclusion, LNA generates ROS, initiates lipid peroxidation, and damages the bacterial membrane, resulting in intracellular leakage and eventually killing the KPC-KP cells.

Parental social support, perceived competence and enjoyment in school physical activity

Background:Beginning in the elementary school years,there are differences among children on how they perceive their competence in physical activity(PA).Children's competence perceptions may influence their affective reactions to PA.A crucial question is how to motivate children who hold low competence perceptions to enhance their enjoyment and PA *** parents play critical roles in children's development and socialization,social support from parents can be an important factor to complement teachers'effort to enhance children's enjoyment and PA *** this research we identified the associations among children's beliefs about parental social support,perceived competence,and enjoyment in school ***:Three hundred and twenty children(9-11 years old)participated in a two-wave *** the first wave,children completed questionnaires measuring their beliefs about parental social support,perceived competence,and enjoyment in school PA;they reported their enjoyment again 8 months later at the second ***:Both concurrent and longitudinal analyses revealed that beliefs about parental social support were important factors associated with children's enjoyment in school PA,especially among girls with low competence ***:Family socialization factors should be taken into *** efficacy of individual and community-level strategies should be included and evaluated when designing effective intervention strategies that enhance children's PA in school.

Today’s rapid proliferation of information and technological advancements has led to complex and uncertain modern systems *** problems resulting from this increased complexity may surpass engineers’current capacity to perform effectively within the domain of complex *** response to this situation,the concept of Systems Thinking(ST)has been advanced as an aid to building a mental map that offers a robust conceptual understanding to offset the challenges of modern system of systems(SoS)*** there has been some research regarding the effect of age and gender on ST preferences,there is still a lack of studies investigating how an individual’s ST skills preferences in system of systems(SoS)domain vary across educational *** addition,most of the extant literature focuses on one or two measures to assess the individual ST;thus,there is a need to include the full spectrum of ST measures to assess the ST skills preferences of an individual in the domain of complex *** address these gaps,this research uses an established ST skills preferences instrument to gauge an individual’s ST skills preferences in the SoS domain based on the educational *** hundred and fifty-eight participants with educational qualifications ranging from non-degree to graduate degree participated in the *** analysis of the responses was performed by a post-hoc test to show which groups differ *** the results obtained through aggregate individual responses,we conclude that each group(i.e bachelor,masters and phd),possesses a different ST skills preference profile on average,and the educational qualifications in the SoS environment has a moderation impact on individuals’system skills preferences.
