Biosynthesizing Au nanoparticles(AuNPs)from gold-bearing scraps provides a sustainable method to meet the urgent demand for AuNPs.However,it remains challenging to efficiently biosynthesize AuNPs of which the diameter is less than 10 nm from a trace amount of Au^(3+)concentration at the level of tens ppm.Here,we constructed an exoelectrogenic cell(eCell)-conductive reduced-graphene-oxide aero-gel(rGA)biohybrid by assembling Shewanella sp.S1(SS1)as living biocatalyst and rGA as conductive ad-sorbent,in which Au^(3+)at trace concentrations would be enriched by the adsorption of rGA and reduced to AuNPs through the extracellular electron transfer(EET)of SS1.To regulate the size of the synthe-sized AuNPs to 10 nm,the strain SS1 was engineered to enhance its EET,resulting in strain RS2(pYYD-P tac-ribADEHC&pHG13-P_(bad)-omcC in SS1).Strain RS2 was further assembled with rGA to construct the RS2-rGA biohybrid,which could synthesize AuNPs with the size of 7.62±2.82 nm from 60 ppm Au^(3+)so-lution.The eCell-rGA biohybrid integrated Au^(3+)adsorption and reduction,which enabled AuNPs biosyn-thesis from a trace amount of Au^(3+).Thus,the required Au^(3+)ions concentration was reduced by one or two orders of magnitude compared with conventional methods of AuNPs biosynthesis.Our work devel-oped an AuNPs size regulation technology via engineering eCell’s EET with synthetic biology methods,providing a feasible approach to synthesize AuNPs with controllable size from trace level of gold ions.

Small-scale electromagnetic soft actuators are characterized by a fast response and simplecontrol,holding prospects in the field of soft and miniaturized robotics.The use of liquid metal(LM)to replace a rigid conductor inside soft actuators can reduce the rigidity and enhance the actuation performance and robustness.Despite research efforts,challenges persist in the flexible fabrication of LM soft actuators and in the improvement of actuation performance.To address these challenges,we developed a fast and robust electromagnetic soft microplate actuator based on a laser-induced selective adhesion transfer method.Equipped with unprecedentedly thin LM circuit and customized low Young’s modulus silicone rubber(1.03 kPa),our actuator exhibits an excellent deformation angle(265.25?)and actuation bending angular velocity(284.66 rad·s^(-1)).Furthermore,multiple actuators have been combined to build an artificial gripper with a wide range of functionalities.Our actuator presents new possibilities for designing small-scaleartificial machines and supports advancements in ultrafast soft and miniaturized robotics.

Dual-interlayers constructed by Ti_(3)C_(2)T_(x)/ionic-liquid for enhanced performance of solid garnet batteries

Li(6.4)La_(3)Zr_(1.4)Ta_(0.6)O_(12)(LLZTO) solid garnet-type electrolyte has been widely reported due to its outstanding safety and electrochemical stability.However,the inherent rigidity and brittleness of LLZTO lead to poor contact with anode/cathode and the operation failure of full cells.Herein,the dual-interlayers are constructed as the fast interfacial ion-migration channel by using Ti_(3)C_(2)T_(x)(MXene,Txis-O,-OH,-F) with trace ionic liquid(IL),which promote the intimate contact between LLZTO and anode/cathode and suppress Li-dendrites growth.Notably,IL can wet the cathode to promote intimate interface contact and be decomposed into some inorganic compounds(such as Li3N,LiF,and Li2Sx),resulting in reduced interfacial resistance and fast Li-ion transportation.Consequently,in the prepared Li-symmetric cell,the interfacial resistance on the anode side plunges to 33.1 Ω cm^(-2),and stably maintains over 1000 h without short circuit at 0.05 mA cm^(-2).The full cell of Li|LiFePO4delivers a high initial capacity of 158.52 mA h g^(-1)and outstanding retention of 90.18% after 100 cycles at 60℃ and 0.2 C.Our work provides an efficient strategy to design dual-interlayers between LLZTO and anode/cathode for the interfacial modification to enhance the performance of solid garnet batteries.

食品胶体是指食品中常见的多组分非均相分散体系,根据组分可划分为蛋白类胶体和多糖类胶体,通常具有增稠、乳化、悬浮、胶凝、黏合、持水、稳定、成膜等重要功能,并且广泛应用于食品加工业等众多领域。本文综述了食品胶体的概念、组成、基本分类,重点阐述了典型的多糖胶体(卡拉胶、琼脂、海藻酸钠、阿拉伯胶、瓜尔豆胶、魔芋胶、黄原胶、结冷胶以及可得然胶)的功能特性,包括流变学特性、凝胶性、稳定性、复配性等;总结了海藻类、植物类、微生物类多糖胶体在食品加工业中的应用现状并展望其发展前景,为进一步开发和利用多糖类食品胶体提供理论依据和实践应用基础。

柯乐猪CRISP 3基因SNP鉴定及其对繁殖性状的影响

为分析富半胱氨酸的分泌蛋白3(cysteine rich secretory protein 3,CRISP3)基因单核苷酸多态性(single nucleotide polymorphism,SNP)位点对柯乐猪繁殖性状的影响,采用Sanger测序法检测250头健康经产柯乐猪母猪CRISP 3基因的SNP突变位点,利用SPSS 22.0软件分析SNP位点与柯乐猪产活仔猪数、初生窝重、初生均重、断奶窝重、断奶仔猪数和断奶均重6个指标的关联性。结果显示,在柯乐猪CRISP 3基因第4内含子共发现3个SNP突变位点,即g.43796318T>C、g.43796424T>A和g.43796425G>T,均存在3种基因型。g.43796318T>C和g.43796424T>A位点的优势基因型为TT,优势等位基因为T;g.43796425G>T位点的优势基因型为GG,优势等位基因为G。χ^(2)检验显示,g.43796318T>C位点基因型分布符合Hardy-Weinberg平衡状态(P>0.05);g.43796424T>A和g.43796425G>T位点基因型分布均极显著(PA与g.43796425G>T之间存在强连锁不平衡。关联分析显示,g.43796318T>C位点TT和CC基因型个体的初生窝重和初生均重均显著(PA位点TA基因型个体的产活仔猪数、初生窝重和断奶仔猪数均显著(PT位点GT基因型个体的产活仔猪数、初生窝重和断奶仔猪数均显著(P<0.05)高于GG和TT基因型。双倍型关联分析显示,双倍型H2H4(CCTAGT)个体的产活仔猪数、初生窝重、断奶仔猪数和断奶窝重显著(P<0.05)高于H1H2(TCTAGT)个体和H2H2(CCAATT)个体;双倍型H2H2(CCAATT)个体的初生均重显著(P<0.05)高于除H4H4(CCTTGG)个体外的其他个体,且其断奶均重显著(P<0.05)高于其他双倍型个体。CRISP 3基因第4内含子上的3个突变位点与柯乐猪的6个繁殖性状均存在显著关联,可作为柯乐猪繁殖性状选择的候选分子标记。

Zebrafish ppp1r21 mutant as a model for the study of primary biliary cholangitis

Primary biliary cholangitis (PBC) is an autoimmune cholestatic liver disease that progresses to fibrosis and cirrhosis, resulting from the gradual destruction of intrahepatic bile ducts. Exploring genetic variants associated with PBC is essential to understand the pathogenesis of PBC. Here we identify a zebrafish balloon dog (blg) mutant with intrahepatic bile duct branching defects, exhibiting several key pathological PBC-like features, including immunodominant autoantigen PDC-E2 production, cholangiocyte apoptosis, immune cell infiltration, inflammatory activation, and liver fibrosis. blg encodes the protein phosphatase 1 regulatory subunit 21 (Ppp1r21), which is enriched in the liver and its peripheral tissues and plays a vital role in the early intrahepatic bile duct formation stage. Further studies show an excessive activation of the PI3K/AKT/mTOR pathway in the hepatic tissues in the mutant, while treatment with the pathway inhibitor LY294002 and rapamycin partially rescues intrahepatic bile duct branching defects and alleviates the PBC-like symptoms. These findings implicate the potential role of the Ppp1r21-mediated PI3K/AKT/mTOR pathway in the pathophysiology of PBC.

【目的】研究角蛋白10(keratin 10,KRT10)基因多态性对柯乐猪繁殖性状的影响,以提高柯乐猪的繁殖力。【方法】以255头柯乐猪为研究对象,采用PCR产物测序、DANMAN序列比对软件及人工校对相结合的方法鉴定KRT10基因单核苷酸多态性(single nucleotide polymorphism, SNP)位点,利用SHEsis在线软件分析SNP位点的群体遗传特性,通过RNAfold在线工具对SNP位点进行生物信息学分析,使用SPSS 22.0软件中的一般线性模型分析KRT10基因SNP位点与柯乐猪繁殖性状的关联性。【结果】在柯乐猪KRT10基因中共鉴定到3个SNPs位点:第4内含子的g.21643703 C>T和g.21643714 G>A;第5外显子的g.21643741 G>A。g.21643741 G>A突变导致密码子由AAG突变为AAA,编码氨基酸均为赖氨酸(K),为同义突变,引起编码的mRNA二级结构发生改变。3个SNPs位点均检测到3种基因型,g.21643703 C>T和g.21643741 G>A属于中度多态位点(0.25A)属于低度多态位点(PICT和g.21643741 G>A位点均处于Hardy-Weinberg平衡状态(P>0.05),g.21643714 G>A位点显著偏离Hardy-Weinberg平衡状态(PT位点对柯乐猪初生窝重、断奶仔数和断奶窝重的影响均达到显著水平(PA位点对柯乐猪总产仔数和断奶窝重的影响均达到显著水平(PA位点对柯乐猪总产仔数、产活仔数、断奶仔数和断奶窝重的影响均达到显著水平(P<0.05)。双倍型H3H3对柯乐猪总产仔数、产活仔数、断奶仔数和断奶窝重的影响最明显。【结论】KRT10基因中存在的3个SNPs位点对柯乐猪繁殖性状有显著影响,其中H3H3为有利双倍型,可作为柯乐猪繁殖性状选择的遗传标记。

【目的】研究孕激素相关子宫内膜蛋白(progestogen associated endometrial protein,PAEP)基因多态性对柯乐猪繁殖性状的影响,以挖掘柯乐猪的高繁殖力基因,用于提升柯乐猪的繁殖力。【方法】以189头经产柯乐猪为试验动物,采用PCR产物测序和序列比对鉴定单核苷酸多态性(SNP)位点,利用SHEsis在线软件分析SNP位点的群体遗传特性,通过RNAfold、SOPMA、SWISS-MODEL和SWISS-PdbViewer程序对SNP位点进行生物信息学分析,利用SPSS 22.0软件分析PAEP基因SNP位点与柯乐猪繁殖性状的相关性。【结果】在柯乐猪PAEP基因中共鉴定到3个SNPs位点:外显子4的g.1884992 T>C位点,为错义突变,导致第135位缬氨酸(V)变成丙氨酸(A);内含子4的g.1885152 G>C和g.1887834 G>A位点。3个突变位点均检测到3种基因型;g.1884992 T>C位点属于低度多态(PICC、g.1887834 G>A位点属于中度多态(0.25C位点产生的基因型分布未偏离Hardy-Weinberg平衡(P>0.05);g.1885152 G>C和g.1887834 G>A位点产生的基因型分布均极显著偏离Hardy-Weinberg平衡(PC和g.1887834 G>A位点完全连锁,共产生3种单倍型和6种双倍型。g.1884992 T>C位点引起PAEP基因mRNA及蛋白的二级结构发生改变。关联分析结果表明,g.1884992 T>C位点对柯乐猪总产仔数和产活仔数的影响均达到显著水平(PC、g.1887834 G>A位点对柯乐猪总产仔数、产活仔数、断奶仔数和断奶窝重的影响均达到显著水平(P<0.05);双倍型H3H3个体总产仔数、产活仔数、断奶仔数和断奶窝重均显著高于其他5种双倍型(P<0.05),初生窝重显著高于双倍型H1H1、H2H2和H2H3(P<0.05);双倍型H1H3个体的总产仔数和断奶仔数显著高于双倍型H1H1、H1H2、H2H2和H2H3(P<0.05),产活仔数显著高于双倍型H2H3(P<0.05)。【结论】PAEP基因中存在的3个SNPs对柯乐猪繁殖性状有显著影响,其中H3H3为有利双倍型,可作为柯乐猪繁殖性状选择的遗传标记。

Major historical events often trigger the rapid flourishing of a few lineages,which in turn shape established biodiversity patterns.How did this process occur and develop?This study provides a window into this issue.The endemic East Asian carps(EEAC)dominated the ichthyofauna of East Asia and exhibited a high degree of adaptation to monsoonal river-lake ecosystems.A series of evidence,including ecogeography,phylogenetics,and macroevolution,suggests that the EEAC is a lineage that arose with the East Asian monsoon and thrived intimately with subsequent monsoon activities.We further deduce the evolution of the EEAC and find that a range of historical events in the monsoon setting(e.g.,marine transgression and regression and glacial-interglacial cycle)have further reshaped the distribution patterns of EEAC’s members.Comparative genomics analyses reveal that introgressions during the initial period of EEAC radiation and innovations in the regulation of the brain and nervous system may have aided their adaptation to river-lake ecosystems in a monsoon setting,which boosted radiation.Overall,this study strengthens knowledge of the evolutionary patterns of freshwater fishes in East Asia and provides a model case for understanding the impact of major historical events on the evolution of biota.

目的总结医务人员对丧亲家属丧亲支持的最佳证据,为临床医务人员对丧亲家属开展丧亲支持服务提供参考。方法按照“6S”证据金字塔模型,系统检索国内外相关数据库中关于医务人员对丧亲家属丧亲支持的临床决策、指南、证据总结、系统评价、专家共识、随机对照试验,检索时限为建库至2023年6月30日,由2名研究人员选取符合纳入标准的文献,对文献质量进行评价,根据主题提取和汇总证据。结果共纳入13篇文献,包括1篇指南、6篇系统评价、3篇专家共识、3篇随机对照试验。从丧亲支持团队要求、丧亲支持原则、丧亲支持评估、通用丧亲支持策略、专业丧亲支持策略5个方面总结形成25条最佳证据。结论总结的医务人员对丧亲家属提供丧亲支持的最佳证据内容较全面,可为临床开展高质量的丧亲支持提供循证依据。
