Friend or foe for obesity: How hepatokines remodel adipose tissues and translational perspective

Due to excess energy intake and a sedentary lifestyle, the prevalence of obesity is rising steadily and has emerged as a global public health problem. Adipose tissue undergoes structural remodeling and dysfunction in the obese state. Secreted proteins derived from the liver, also termed as hepatokines, exert multiple effects on adipose tissue remodeling and the development of obesity, and has drawn extensive attention for their therapeutic potential in the treatment of obesity and related diseases. Several novel hepatokines and their functions on systemic metabolism have been interrogated recently as well. The drug development programs targeting hepatokines also have shown inspiring benefits in obesity treatment. In this review, we outline how adipose tissue changes during obesity. Then, we summarize and critically analyze the novel findings on the effects of metabolic “beneficial” and metabolic “harmful” hepatokines to adipose tissue. We also discuss the in-depth molecular mechanism that hepatokines may mediate the liver-adipose tissue crosstalk, the novel technologies targeting hepatokines and their receptors in vivo to explore their functions, and the potential application of these interventions in clinical practice.

Background Anthocyanins(AC)showed positive effects on improving the intestinal health and alleviating intestinal pathogen infections,therefore,an experiment was conducted to explore the protective effects of supplemented AC on Salmonella-infected chickens.Methods A total of 240 hatchling chickens were randomly allocated to 4 treatments,each with 6 replicates.Birds were fed a basal diet supplemented with 0(CON,and ST),100(ACL)and 400(ACH)mg/kg of AC for d 60,and orally challenged with PBS(CON)or 10^(9) CFU/bird(ST,ACL,ACH)Salmonella Typhimurium at d 14 and 16.Results(1)Compared with birds in ST,AC supplementation increased the body weight(BW)at d 18 and the average daily gain(ADG)from d 1 to 18 of the Salmonella-infected chickens(P<0.05);(2)AC decreased the number of Salmonella cells in the liver and spleen,the contents of NO in plasma and inflammatory cytokines in ileal mucosa of Salmonella-infected chickens(P<0.05);(3)Salmonella infection decreased the ileal villi height,villi height to crypt depth(V/C),and the expression of zonulaoccludins-1(ZO-1),claudin-1,occludin,and mucin 2(MUC2)in ileal mucosa.AC supplementation relieved these adverse effects,and decreased ileal crypt depth(P<0.05);(4)In cecal microbiota of Salmonella-infected chickens,AC increased(P<0.05)the alpha-diversity(Chao1,Pd,Shannon and Sobs indexes)and the relative abundance of Firmicutes,and decreased(P<0.05)the relative abundance of Proteobacteria and Bacteroidota and the enrichment of drug antimicrobial resistance,infectious bacterial disease,and immune disease pathways.Conclusions Dietary AC protected chicken against Salmonella infection via inhibiting the Salmonella colonization in liver and spleen,suppressing secretion of inflammatory cytokines,up-regulating the expression of ileal barrier-related genes,and ameliorating the composition and function of cecal microbes.Under conditions here used,100 mg/kg bilberry anthocyanin was recommended.

Effect of powdery mildew on interleaf microbial communities and leaf antioxidant enzyme systems

Chinese peony(Paeonia lactiflora Pall.)is both medicinally and aesthetically beneficial.Powdery mildew is a common fungal disease that seriously jeopardizes the value of numerous species,including peonies as a crop.In order to provide a basis for the prevention and treatment of peony powdery mildew,we examined the microbial diversity,the malondialdehyde(MDA)concentrations and antioxidant enzyme activities of peony leaves infected with three levels of powdery mildew to determine any modifications to the leaf's antioxidant enzyme systems and microbial community structure following the onset of disease.The results show that the MDA content rose as the degree of infection became worse.Antioxidant enzyme activity rose and then declined.Following the initiation of powdery mildew,fungal community diversity decreased,whereas there was not any appreciable change in bacterial communities according to microbial diversity sequencing.The relative abundance of more than half of fungal species decreased,with the bacterial genera displaying both abundant and diminished communities with less pronounced alterations in their community structure after the disease spread.Significant different taxa that were critical to the organization of each microbiome were found.Correlation analysis showed that the relative abundance of powdery mildew pathogenic fungal genus Erysiphe was correlated with those of 11 fungal genera and one bacterial genus.Among them,Aureobasidium,Neosetophoma and Sclerostagonospora showed significant positive correlations with Erysiphe and MDA.

Breathing is an inherent human activity;however,the composition of the air we inhale and gas exhale remains unknown to us.To address this,wearable vapor sensors can help people monitor air composition in real time to avoid underlying risks,and for the early detection and treatment of diseases for home healthcare.Hydrogels with three-dimensional polymer networks and large amounts of water molecules are naturally flexible and stretchable.Functionalized hydrogels are intrinsically conductive,self-healing,self-adhesive,biocompatible,and room-temperature sensitive.Compared with traditional rigid vapor sensors,hydrogel-based gas and humidity sensors can directly fit human skin or clothing,and are more suitable for real-time monitoring of personal health and safety.In this review,current studies on hydrogel-based vapor sensors are investigated.The required properties and optimization methods of wearable hydrogel-based sensors are introduced.Subsequently,existing reports on the response mechanisms of hydrogel-based gas and humidity sensors are summarized.Related works on hydrogel-based vapor sensors for their application in personal health and safety monitoring are presented.Moreover,the potential of hydrogels in the field of vapor sensing is elucidated.Finally,the current research status,challenges,and future trends of hydrogel gas/humidity sensing are discussed.

Graphitic carbon nitride(g-C_(3)N_(4)) is a fascinating photocatalyst for solar energy utilization in photo-catalysis.Nevertheless,it often suffers from moderate photo-catalytic activity due to its low specific surface area and fast recombination rate of photogenerated electrons upon photo-excitation.Herein,we overcome the bottlenecks by constructing a porous g-C_(3)N_(4) nanosheet(PCNS)through a simple thermal oxidation etching method.Benefited from its porous layer structure,the obtained PCNS exhibits large specific surface area,efficient separation of photogenerated charge carriers,as well as high exposure of active sites.As a result,it is robust and universal in visible light-driven dehydrogenation of alcohols in water under oxidant-free condition.Almost quantitative yields(>99%)of various valuable carbonyl compounds were obtained over PCNS,while bulk g-C_(3)N_(4) was far less efficient.Moreover,the photo-catalyst was highly stable and could be facilely recovered from the aqueous system for efficient reuse.The easy preparation and excellent performance made PCNS a promising and competitive photocatalyst for the solar applications.

The development of a single analytical platform with different functions is highly desirable but remains a challenge at present.Here,a paper-based device based on fluorescent carbon dots(CDs)functionalized paper/MnO_(2)nanosheets(MnO_(2)NS)hybrid devices(PCD/NS)was proposed for single-device multi-function applications.MnO_(2)NS functioned as a fluorescence quencher of CDs and recognizer of H_(2)O_(2)released from the oxidase catalyzed system.Fluorescence recovery would occur after the decomposition of MnO_(2)NS induced by H_(2)O_(2),by which a simple and effective strategy could be developed for fluorescence monitoring multiplex biological events.Xanthine(XA)sensing,xanthine oxidase(XOD)inhibitors screening analysis and chiral recognition of glucose enantiomers were performed on PCD/NS to investigate the multifunctional application of the paper-based device.By means of PCD/NS,XA could be determined in the range of 0.1–40μmol/L with a low detection of limit of 0.06μmol/L.The IC_(50)value of allopurinol,the model inhibitor of XOD,was sensitively detected to be 7.4μmol/L.Glucose enantiomers were also recognized in terms of the specific fluorescence response to d-glucose.This work firstly presented a paper-based device capable of biomarkers detection,inhibitors screening and chiral recognition,which enlightened a promising strategy for the construction of multifunctional devices and hold the great potential application in clinical diagnosis and drug discovery.

Kinetic mechanism of copper extraction from methylchlorosilane slurry residue using hydrogen peroxide as oxidant

Copper was extracted from methylchlorosilane slurry residue by a direct hydrogen peroxide leaching method.A number of experimental parameters were analyzed to determine the extraction efficiency of copper.The extraction efficiency of copper reached 98.5%under the optimal leaching conditions,such as the hydrogen peroxide concentration of 1.875 mol·L^(-1),the leaching temperature of 323 K,the liquid–solid ratio of 20 ml·g^(-1),and the stirring speed of 300 r·min^(-1).The leaching kinetics of the copper extraction process was then described by the shrinking core model.There were two stages.The first stage was controlled by chemical reactions,while the second stage was controlled by interface transfer and product layer diffusion.The activation energy and kinetic control equations were determined,as well as an explanation of the leaching mechanism of copper extraction based on kinetic analysis and materials characterization.Copper resources can be recovered from the methylchlorosilane slurry residue efficiently and inexpensively with the methods used in this study.

This study aimed to explore the role of mechanical tension in hypertrophic scars and the change in nerve density using hematoxylin-eosin staining and S100 immunohistochemistry, and to observe the expression of nerve growth factor by western blot analysis. The results demonstrated that mechanical tension contributed to the formation of a hyperplastic scar in the back skin of rats, in conjunction with increases in both nerve density and nerve growth factor expression in the scar tissue. These experimental findings indicate that the cutaneous nervous system plays a role in hypertrophic scar formation caused by mechanical tension.
