Lymnaeid snails are key intermediate hosts for the development and survival of Fasciola spp.,the causative agent of Fascioliasis which are economically important parasites infecting humans and livestock *** current control method for treating Fascioliasis is heavily reliant on anthelmintic drugs,particularly Triclabendazole(TCBZ)which has resulted in drug-resistant parasites and poses significant risk as there are no long-term efficacious alternatives *** control measures at the farm level could include both parasite and snail control will play an important role in Fasciola *** and reduce the reliance on anthelmintic *** of such sustainable control measures requires effective identification of snails on the property however Lymnaeid snails are small and difficult to physically *** identification using an environmental DNA approach is a recent approach in which physically locating snails are not *** tomentosa,is the primary intermediate snail host for *** transmission in South-East Australia and we present an in-field loop-mediated isothermal amplification and water filtering method for the detection of *** eDNA from water samples to improve current surveillance *** methodology is highly sensitive with a detection limit of 5×10^(−6)ng/μL,detected in<20 minutes,with cumulative sample preparation and amplification time under 1 *** proposed workflow could assist in monitoring areas to determine the risk of Fascioliasis infection and implement strategies to manage snail populations to ultimately reduce the risk of infection for humans and livestock.

Effects of aspartame on hsp70,bcl-2 and bax expression in immune organs of Wistar albino rats

Aspartame, a ＂first generation sweetener＂, is widely used in a variety of foods, beverages, and medicine. The FDA has determined the acceptable daily intake （ADI） value of aspartame to be 50 mg/kg, day, while the JECFA （Joint FAO/WHO Expert Committee on Food Additives） has set this value at 40 mg/kg of body weight/day. Safety issues have been raised about aspartame due to its metabolites, specifically toxicity from methanol and/or its systemic metabolites formaldehyde and formic acid. The immune system is now recognized as a target organ for many xenobiotics, such as drugs and chemicals, which are able to trigger unwanted apoptosis or to alter the regulation of apoptosis. Our previous studies has shown that oral administration of aspartame [40 mg/（kg, day）] or its metabolites for 90 days increased oxidative stress in immune organs of Wistar albino rats. In this present study, we aimed to clarify whether aspartame consumption over a longer period （90-days） has any effect on the expression ofhsp70, bcl- 2 and bax at both mRNA transcript and protein expression levels in immune organs. We observed that oral administration of aspartame for 90 days did not cause any apparent DNA fragmentation in immune organs of aspartame treated animals; however, there was a significant increase in hsp70 expression, apart from significant alteration in bcl-2 and bax at both mRNA transcript and protein expression level in the immune organs of aspartame treated animals compared to controls. Hence, the results indicated that hsp70 levels increased in response to oxidative injury induced by aspartame metabolites; however, these metabolites did not induce apoptosis in the immune organs. Furthermore, detailed analyses are needed to elucidate the precise molecular mechanisms involved in these changes.

Free radical scavenging potential and HPTLC analysis of Indigofera tinctoria linn(Fabaceae)

The objective of this study was to evaluate the free radical scavenging potential and high performance thin layer chromatography （HPTLC） fingerprinting of Indigofem tinctoria （I. tinctoria）, Phytochemical analysis was carried out using standard methods, and free radical scavenging activity of the plant was determined using 2,2-diphenyl-l-picrylhydrazy （DPPH）, nitric oxide （NO） and superoxide anion （O2-） radical scavenging capacities. HPTLC plate was kept in CAMAG TLC Scanner 3 and the Rf values at fin- gerprint data were recorded by WlNCATS software, Aqueous extract of I. tinctoria reliably showed the total phenolics （267.2 ± 2.42 mg/g）, flavonoids （75.43 ± 3.36 mg/g） and antioxidants （349.11 ±8.04 mg/g）. The extract was found to have DPPH （52.08%）, NO （23.12%） and 02 （26.79%） scavenging activities at the concentration of 250 pg/mL and the results were statistically significant compared with ascorbic acid standard （p 〈 0.05）. HPTLC results confirmed that the extract contained several potential active com- ponents such as phenols, flavonoids, saponins and terpenoids as the slides revealed multi-colored bands of varying intensities. This study confirmed that the plant had multipotential antioxidant and free ra- dicals scavenging activities.

Acute effect of aspartame-induced oxidative stress in Wistar albino rat brain

The present study was carried out to investigate the acute effect of aspartame on oxidative stress in the Wistar albino rat brain. We sought to investigate whether acute administration of aspartame （75 mg/kg） could release methanol and induce oxidative stress in the rat brain 24 hours after administration. To mimic human methanol metabolism, methotrexate treated rats were used to study aspartame effects. Wistar strain male albino rats were administered with aspartame orally as a single dose and studied along with controls and methotrexate treated controls. Blood methanol and formate level were estimated after 24 hours and rats were sacrificed and free radical changes were observed in discrete regions by assessing the scavenging enzymes, reduce dglutathione （GSH）, lipid peroxidation and protein thiol levels. There was a significant increase in lipid peroxidation levels, superoxide dismutase activity （SOD）, glutathione peroxidase levels （GPx）, and catalase activity （CAT） with a significant decrease in GSH and protein thiol. Aspartame exposure resulted in detectable methanol even after 24 hours. Methanol and its metabolites may be responsible for the generation of oxidative stress in brain regions. The observed alteration in aspartame fed animals may be due to its metabolite methanol and elevated formate. The elevated free radicals due to methanol induced oxidative stress.

Self-reported sleep quality and depression in post myocardial infarction patients attending cardiology outpatient clinics in Oman

Ethanolic extract of Puhuang(Pollen Typhae) modulates lipopolysaccharide-induced inflammatory response through inducible nitric oxide synthase/cyclooxygenase-2 signaling in RAW 264.7 macrophages

OBJECTIVE:To evaluate the immune modulatory response of Puhuang(Pollen Typhae),ethanolic extract of dried pollens(TP-E)and charcoal activated pollens(CTP-E)were used for their phytochemical evaluation and their modulatory response against lipopolysaccharide(LPS)induced inflammatory activity on RAW264.7 macrophage ***:Biochemical assays were carried out to quantify the 1,1-Diphenyl-2-picrylhydrazyl Radical Scavenging Activity,Reducing Power,Ferrous ion chelating ability and total polyphenol content and ***-toxic dose of the extract(TP-E and CTP-E)was chosen based on 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide *** of TP-E and CTP-E on lipopolysaccharides-induced inducible nitric oxide synthase(iNOS)and cyclooxygenase-2(COX-2)expression was measured by Western blot and quantitative PCR(q RT-PCR).Expression of inflammatory cytokines,such as interleukins(IL-1βand IL-6)and tumor necrosis factorα(TNF-α),was quantified using q ***-activated protein kinase pathway was analyzed using Western ***:Phytochemical analysis revealed that both TP-E and CTP-E have strong antioxidant activities and high flavonoid and phenolic ***-E and CTP-E effectively inhibit the expression of i NOS and COX-2,thereby inhibiting its downstream proinflammatory regulators,the extracellular signal-related kinase-1/2,that decreases the expression of IL-1β,IL-6 and TNF-α.CONCLUSION:Phytochemical constituents present in Typha angustifolia Linn could be used for treating inflammation-related diseases.

Antioxidant mediated response of Scoparia dulcis in noise-induced redox imbalance and immunohistochemical changes in rat brain

Noise has been regarded as an environmental/occupational stressor that causes damages to both auditory and non- auditory organs. Prolonged exposure to these mediators of stress has often resulted in detrimental effect, where oxidative/nitrosative stress plays a major role. Hence, it would be appropriate to examine the possible role of free radicals in brain discrete regions and the ＂antioxidants＂ mediated response of S. dulcis. Animals were subjected to noise stress for 15 days （100 dB/4 hours/day） and estimation of endogenous free radical and antioxidant activity were carried out on brain discrete regions （the cerebral cortex, cerebellum, brainstem, striatum, hippocampus and hypothalamus）. The result showed that exposure to noise could alleviate endogenous free radical generation and altered anfioxidant status in brain discrete regions when compared to that of the control groups. This alleviated free radical generation （H202 and NO） is well supported by an upregulated protein expression on immunohistochemistry of both iNOS and nNOS in the cerebral cortex on exposure to noise stress. These findings suggest that increased free radical generation and altered anti-oxidative status can cause redox imbalance in the brain discrete regions~ However, free radical scavenging activity of the plant was evident as the noise exposed group treated with S. dulcis[200 mg/（kg · b. w）] displayed a therapeutic effect by decreasing the free radical level and regulate the anti-oxidative status to that of control animals. Hence, it can be concluded that the efficacy of S. dulcis could be attributed to its free radical scavenging activity and anti-oxidative property.

Effect of quercetin against lindane induced alterations in the serum and hepatic tissue lipids in wistar rats
