Melatonin ameliorates experimental hepatic fibrosis induced by carbon tetrachloride in rats

AIM:To investigate the protective effects of melatonin on carbon tetrachloride(CCl4)-induced hepatic fibrosis in experimental ***:All rats were randomly divided into normal control group,model control group treated with CCl4 for 12 wk,CCl4+NAC group treated with CCl4+NAC(100 mg/kg,i.p.)for 12 wk,CCl4+MEL-1 group treated with CCl4+melatonin(2.5 mg/kg)for 12 wk,CCl4+MEL-2 group treated with CCl4+ melatonin(5.0 mg/kg)for 12 wk,and CCl4+MEL-3 group treated with CCl4+melatonin(10 mg/kg).Rats in the treatment groups were injected subcutaneously with sterile CCl4(3 mL/kg,body weight)in a ratio of 2:3 with olive oil twice a *** in normal control group received hypodermic injection of olive oil at the same dose and frequency as those in treatment *** the end of experiment,rats in each group were anesthetized and *** and eosin(HE)staining and Van Gieson staining were used to examine changes in liver *** activities of alanine aminotransferase(ALT),aspartate aminotransferase(AST)and protein concentration weremeasured with routine laboratory methods using an ***(HYP)content in liver and malondialdehyde(MDA)and glutathione peroxidase(GPx)levels in liver homogenates were assayed by *** hyaluronic acid(HA),laminin(LN),and procollagenⅢN-terminal peptide(PⅢNP)were determined by ***:Pathologic grading showed that the fibrogenesis was much less severe in CCl4+MEL3 group than in model control group(u=2.172,P<0.05),indicating that melatonin(10 mg/kg)can significantly ameliorate CCl4-induced hepatic fibrotic *** serum levels of ALT and AST were markedly lower in CCl4+MEL treatment groups(5,10 mg/kg)than in model control group(ALT:286.23 ±121.91 U/L vs 201.15±101.16 U/L and 178.67 ±103.14 U/L,P=0.028,P=0.007;AST:431.00 ±166.35 U/L vs 321.23±162.48 U/L and 292.42 ±126.23 U/L,P=0.043,P=0.013).Similarly,the serum laminin(LN)and hyaluronic acid(HA)levels and hydroxyproli

＜正＞ Paeonol （Pae）, a naturally occurring agent extracted from Moutan cortex, has shown great promise in the antitumor activity. The aim of this study was to investigate the effect of Pae and its combination with cisplatin （CDDP） on cell growth and apoptosis of human hepatoma cell lines HepG2 and SMMC - 7721. The cytotoxic effect of drugs on the two cell lines was measured by MTT assay. The interaction of Pae and CDDP was evaluated by coefficient of drag interaction. Morphologic changes were observed by acridine orange fluorescence staining. Cell cycle progression and apoptotic rate were detected by flow cytometry. The results indicated that Pae and CDDP had cytotoxic effect on the two cell lines in a dose -dependent manner. Pae combined with various concentrations of CDDP showed synergistically cytotoxic and apoptosis - inducing effect on the two cell lines. And the interaction between Pae and CDDP was specific to each cell line. Additionally, a combination of Pae with CDDP resulted in a stronger G2/M arrest, compared to these agents alone in the two cell lines. Pae may be effective and useful as a new biochemical modulator in chemotherapy.

Danggui-Buxue-Tang(DBT) is a famous traditional Chinese *** have reported that total glucosides of Danggui-Buxue-Tang(DBTG) can attenuate bleomycin-induced lung fibrosis through reduction of pro-inflammatory cytokines or the decrease of I collagen *** present study we further focuses on investigating mechanisms underlying the anti-fibrosis effects of DBTG on pulmonary fibrosis(PF) *** were randomly divided into six groups:(1) saline control group;(2) Bleomycin group in which rats were intratracheally instillated with bleomycin(5mg/kg);(3-5) Bleomycin and DBTG group,in which DBTG were given to rats daily(16,32 or 64 mg/kg/day,i.g.) one day after bleomycin instillation for 4 weeks until the end of the treatment;(6) Bleomycin and positive control group. Bleomycin stimulation resulted in a significant increase in the pathological scores for alveolitis and lung fibrosis and an evident increase in the contents of extracellular matrix(ECM) components in serum including hyaluronic acid(HA),laminin(LN), procollagen typeⅢ(PCⅢ) and collagenⅣ(CⅣ).Moreover,bleomycin-induced pulmonary injury and lung fibrosis was indicated by increased lung MMP-1,MMP-9 and TIMP-1 mRNA *** the other hand,DBTG treatment attenuated bleomycin-induced alveolitis and lung fibrosis,significant decreased the concentrations of HA,LN,PCⅢand CⅣin ***,administration of DBTG resulted in a significant reduction in metalloproteinase-1(MMP-1),MMP-9 and the tissue inhibitor of metalloproteinase-1(TIMP-1) mRNA levels in different stages of pulmonary fibrosis,and the balance between MMPs and ***,the findings of the present study provide evidence that DBTG inhibits ECM deposition and modulates MMPs and TIMPs expression in the lung tissue,which may be associated with the attenuation of experimental pulmonary fibrosis.

目的探讨MEK/ERK通路在高脂饮食诱导NAFLD大鼠固醇调控元件结合蛋白-1(SREBP-1)表达中的作用。方法应用高脂饮食复制大鼠非酒精性脂肪性肝病NAFLD模型。32只雄性Wister大鼠随机分成正常对照组(N组)、PD98059+普通饲料组(P组)、高脂模型组(M组)和PD98059+高脂组(PM组),每组8只。于分组喂养的第4、6、8周末分别给予P组和PM组大鼠鼠尾静脉注射MEK抑制剂PD98059干预。第10周末结束实验,检测血清丙氨酸转移酶(ALT)、门冬氨酸转移酶(AST)、甘油三酯(TG)、总胆固醇(TC),肝组织TG、TC;光镜观察大鼠肝脏的病理改变,免疫组化法检测肝脏磷酸化细胞外信号调节激酶1/2(p-ERK1/2)和SREBP-1表达。结果与N组比较,M组所有大鼠肝细胞脂肪变性范围均超过30%并伴有不同程度的炎性细胞浸润和坏死,p-ERK1/2和SREBP-1表达增强,ALT、AST、TC、TG、肝组织TG、TC显著升高(P<0.01或0.05)。与M组比较,PM组p-ERK1/2和SREBP-1表达减少(P<0.01),ALT、TC、TG、肝组织TG、TC显著降低(P<0.01或0.05),同时大鼠的肝脏病理学得到了改善。结论 MEK/ERK通路在高脂饮食诱导NAFLD大鼠固醇调控元件结合蛋白-1(SREBP-1)表达中起重要作用。

Non-muscle invasive bladder cancer(NMIBC)is a major type of bladder cancer with a high incidence worldwide,resulting in a great disease *** and surveillance are the most important part of NIMBC *** 2018,we issued“Treatment and surveillance for non-muscle-invasive bladder cancer in china:an evidencebased clinical practice guideline”.Since then,various studies on the treatment and surveillance of NMIBC have been *** is a need to incorporate these materials and also to take into account the relatively limited medical resources in primary medical institutions in *** a version of guideline which takes these two issues into account to promote the management of NMIBC is therefore *** formed a working group of clinical experts and *** questionnaire investigation of clinicians including primary medical institutions,24 clinically concerned issues,involving transurethral resection of bladder tumor(TURBT),intravesical chemotherapy and intravesical immunotherapy of NMIBC,and follow-up and surveillance of the NMIBC patients,were determined for this *** and recommendations on the management of NMIBC in databases,guideline development professional societies and monographs were referred to,and the European Association of Urology was used to assess the certainty of generated ***,we issued 29 statements,among which 22 were strong recommendations,and 7 were weak *** recommendations cover the topics of TURBT,postoperative chemotherapy after TURBT,Bacillus Calmette–Guérin(BCG)immunotherapy after TURBT,combination treatment of BCG and chemotherapy after TURBT,treatment of carcinoma in situ,radical cystectomy,treatment of NMIBC recurrence,and follow-up and *** hope these recommendations can help promote the treatment and surveillance of NMIBC in china,especially for the primary medical institutions.

Benign prostatic hyperplasia (BPH) is highly prevalent among older men, impacting on their quality of life, sexual function, and genitourinary health, and has become an important global burden of disease. Transurethral plasmakinetic resection of prostate (TUPKP) is one of the foremost surgical procedures for the treatment of BPH. It has become well established in clinical practice with good efficacy and safety. In 2018, we issued the guideline “2018 Standard Edition”. However much new direct evidence has now emerged and this may change some of previous recommendations. The time is ripe to develop new evidence-based guidelines, so we formed a working group of clinical experts and methodologists. The steering group members posed 31 questions relevant to the management of TUPKP for BPH covering the following areas: questions relevant to the perioperative period (preoperative, intraoperative, and postoperative) of TUPKP in the treatment of BPH, postoperative complications and the level of surgeons’ surgical skill. We searched the literature for direct evidence on the management of TUPKP for BPH, and assessed its certainty generated recommendations using the grade criteria by the European Association of Urology. Recommendations were either strong or weak, or in the form of an ungraded consensus-based statement. Finally, we issued 36 statements. Among them, 23 carried strong recommendations, and 13 carried weak recommendations for the stated procedure. They covered questions relevant to the aforementioned three areas. The preoperative period for TUPKP in the treatment of BPH included indications and contraindications for TUPKP, precautions for preoperative preparation in patients with renal impairment and urinary tract infection due to urinary retention, and preoperative prophylactic use of antibiotics.
