Background The thirteen-valent pneumococcal conjugate vaccine(PCV13)is not included in the national immu-nization program and is administered voluntarily with informed consent in China.In preparation for assessing the impact of pilot introduction in Hainan Province,we conducted a carriage study among children under 5 years of age from four locations in Hainan Province,China.Methods From March to June 2022,nasopharyngeal(NP)swabs,collected from healthy children aged younger than 59 months who lived in the 4 different locations(Haikou,Wanning,baisha and Qiongzhong)in Hainan Province,were tested for pneumococcus using conventional culture.Pneumococcal isolates were serotyped using the Quel-lung reaction.Risk factors associated with pneumococcal colonization were assessed using univariate analysis and multivariable logistic regression adjusting for age,daycare attendance and other factors.Results Pneumococcus was isolated in 710(30.4%)of the 2333 children enrolled.Of 737 pneumococci,29 serotypes were identifed;60.9%were PCV13 serotypes;the most common vaccine serotypes were 6B(20.4%),19F(13.0%),6A(11.9%)and 23F(6.1%);and the most common nonvaccine serotypes were 23A(12.9%),34(6.1%)and nontypeable(NT)pneumococci(5.6%).Children vaccinated with PCV13 had lower carriage(17.7%vs 32.5%;P=0.0001)and fewer PCV13 serotypes(41.9%vs 62.7%;P=0.0017)compared to unimmunized children.After adjustment,NP carriage Was higher among children attending daycare(aOR=2.3,95%CI:1.7-3.2),living in rural areas(aOR=1.4,95%CI:1.1-1.8),living with siblings(aOR=1.3,95%CI:1.0-1.6)and whose mothers had completed senior high/technical sec-ondary school(aOR=1.5,95%CI:1.1-2.0).In contrast,completion of 3-4 doses of PCV13 were associated with a lower carriage rate(aOR=0.6,95%CI:0.4-0.9).Conclusions We established the baseline of pneumococcal carriage,serotype distribution and PCV13 immuni-zation rates among healthy children under 5 years of age in Hainan Province,prior to the introduction of PCV13 into the national immunization program.The high proport

Connecting Calcium-Based Nanomaterials and Cancer:From Diagnosis to Therapy

As the indispensable second cellular messenger,calcium signaling is involved in the regulation of almost all physiological processes by activating specific target proteins.The importance of calcium ions(Ca^(2+))makes its“Janus nature”strictly regulated by its concentration.Abnormal regulation of calcium signals may cause some diseases;however,artificial regulation of calcium homeostasis in local lesions may also play a therapeutic role.“Calcium overload,”for example,is characterized by excessive enrichment of intracellular Ca^(2+),which irreversibly switches calcium signaling from“positive regulation”to“reverse destruction,”leading to cell death.However,this undesirable death could be defined as“calcicoptosis”to offer a novel approach for cancer treatment.Indeed,Ca^(2+)is involved in various cancer diagnostic and therapeutic events,including calcium overload-induced calcium homeostasis disorder,calcium channels dysregulation,mitochondrial dysfunction,calcium-associated immunoregulation,cell/vascular/tumor calcification,and calcification-mediated CT imaging.In paral-lel,the development of multifunctional calcium-based nanomaterials(e.g.,calcium phosphate,calcium carbonate,calcium peroxide,and hydroxyapatite)is becoming abundantly available.This review will highlight the latest insights of the calcium-based nanomaterials,explain their application,and provide novel perspective.Identifying and characterizing new patterns of calcium-dependent signaling and exploiting the disease element linkage offer additional translational opportunities for cancer theranostics.

Two central circadian oscillators OsPRR59 and OsPRR95 modulate magnesium homeostasis and carbon fixation in rice

Photosynthesis,which provides oxygen and energy for all living organisms,is circadian regulated.Photosynthesis-associated metabolism must tightly coordinate with the circadian clock to maximize the efficiency of the light-energy capture and carbon fixation.However,the molecular basis for the interplay of photosynthesis and the circadian clock is not fully understood,particularly in crop plants.Here,we report two central oscillator genes of circadian clock,OsPRR95 and OsPRR59 in rice,which function as transcriptional repressors to negatively regulate the rhythmic expression of OsMGT3 encoding a chloroplast-localized Mg2+transporter.OsMGT3-dependent rhythmic Mg fluctuations modulate carbon fixation and consequent sugar output in rice chloroplasts.Furthermore,sugar triggers the increase of superoxide,which may act as a feedback signal to positively regulate the expression of OsPRR95 and OsPRR59.Taken together,our results reveal a negative-feedback loop that strengthens the crosstalk between photosynthetic carbon fixation and the circadian clock,which may improve plan adaptation and performance in fluctuating environments.

Homocysteine impedes neurite outgrowth recovery after intracerebral haemorrhage by downregulating pCAMK2A

Hyperhomocysteinemia(HHcy)is independently associated with poorer long-term prognosis in patients with intracerebral haemorrhage(ICH);however,the effect and mechanisms of HHcy on ICH are still unclear.Here,we evaluated neurite outgrowth and neurological functional recovery using simulated models of ICH with HHcy in vitro and in vivo.We found that the neurite outgrowth velocity and motor functional recovery in the ICH plus HHcy group were significantly slower than that in the control group,indicating that homocysteine(Hcy)significantly impedes the neurite outgrowth recovery after ICH.Furthermore,phosphoproteomic data and signalome analysis of perihematomal brain tissues suggested that calmodulin-dependent protein kinases 2(CAMK2A)kinase substrate pairs were significantly downregulated in ICH with HHcy compared with autologous blood injection only,both western blot and immunofluorescence staining confirmed this finding.Additionally,upregulation of pCAMK2A significantly increased neurite outgrowth recovery in ICH with HHcy.Collectively,we clarify the mechanism of HHcy-hindered neurite outgrowth recovery,and pCAMK2A may serve as a therapeutic strategy for promoting neurological recovery after ICH.

Minimally invasive interventional embolization has tremendous advantages over conventional surgery in the treatment of vascular diseases and malignant tumors.Particularly,liquid embolic agents have garnered much attention owing to their properties of deep vascular penetration and full occlusion of vasculature independent of thrombus formation,which relies on the patient’s coagulation system.Despite their drawbacks,traditional liquid embolic agents,such as lipiodol,N-butyl-2-cyanoacrylate,and Onyx,have benefited numerous patients.The original liquid embolic agents have been modified and optimized,and novel liquid embolic agents have also been developed using different strategies and mechanisms.This minireview provides a brief overview of liquid embolic agents,including approved and potential ones.Challenges,prospects,and rational design guidelines are also discussed.This review may spark interest in liquid embolic agents and open new avenues for the smart design of improved embolic agents in the development of personalized and precision medicine.

Distribution of functional microorganisms and its significance for iron, sulphur, and nitrogen cycles in reservoir sediments

The biogeochemical cycles of sulphur(S),iron(Fe)and nitrogen(N)elements play a key role in the reservoir ecosystem.However,the spatial positioning and interrelationship of S,Fe and N cycles in the reservoir sediment profile have not been explored to a greater extent.Here,we measure the gradients of Fe^(2+),SO_(4)^(2-),NO_(3)^(-),NH_(4)^(+),DOC,TC and TN in the pore water of the sediment,and combining the vertical distribution of the functional microorganisms involved in S,Fe and N cyclings in the sediments to determine the redox stratification in the sediment.It is found that the geochemical gradient of S,Fe and N of the reservoir sedimentary column is mainly defined by the redox process involved in the related functional microorganisms.According to the type of electron acceptor,the sediment profile is divided into 3 redox intervals,namely aerobic respiration(0–10 cm),denitrification/iron reduction(10–28 cm)and sulfate reduction(28–32 cm).In the aerobic respiration zone,NH_(4)^(+)is oxidized by aerobic AOB to NO_(3)^(-)(0–5 cm),and Fe^(2+)is oxidized by microaerobic FeRB to Fe^(3+)(3–10 cm).In the denitrification/iron reduction zone,Acinetobacter and Pseudomonas,as the dominant NRB genera,may use nitrate as an electron acceptor to oxidize Fe^(2+)(11–16 cm).The dominant genera in SOB,such as Sulfururvum,Thiobacillus and Thioalkalispira,may use nitrate as an electron acceptor to oxidize sulfide,leading to SO_(4)^(2-)accumulation(14–24 cm).In the sulfate reduction zone,SO_(4)^(2-)is reduced by SRB.This study found that functional microorganisms forming comprehensive local ecological structures to adapt to changing geochemical conditions,and which would be potentially important for the degradation and preservation of C and the fate of many nutrients and contaminants in reservoirs.

Improved salt tolerance of Populus davidiana × P. bolleana overexpressed LEA from Tamarix androssowii

Development of transgenic plants with tolerance to environ- mental stress is an important goal of plant biotechnology. （LEA） proteins accumulate in seeds dur- ing late embryogenesis, where they protect cellular membranes and macromolecules against drought. In this work, we transferred the Tamarix androssowii LEA gene into hybrids of Populus davidiana xp. bolleana. We compared relative rates of height growth, chlorophyll fluo- rescence kinetic parameters, and leaf Na＋ levels of six TaLEA-containing lines with non-transferred plants （NT）, all grown under 0.8% NaC1 stress condition. Survival percentages of transgenic lines were all higher than for NT controls after rehydration and the sur- vival percentage of SL2 was five-fold higher than for NT controls. Seed- ling height increased 48.7% in SL2 （from the onset of induced stress to the end of the growing season）, 31% more than for the NT controls. Chlorophyll fluorescence kinetic parameters showed a marked increase in photosynthetic capacity in SL2 and SL5. Na＋ levels in young leaves of transgenic lines were lower than in control NT leaves, but higher in yel- low and withered leaves, indicating improved salt tolerance in transgenic lines.

Clinical and pathological features and risk factors for primary breast cancer patients

BACKGROUND Breast cancer is the most common malignancy in women all around the world.According to the latest statistics in 2018,there were more than 2.08 million new breast cancer cases all around the world and more than 620000 deaths;the proportion of breast cancer deaths in women with cancer is 15%.By studying age,clinicopathological characteristics and molecular classification,age at menarche,age at birth,number of births,number of miscarriages,lactation time,surgical history of benign breast lesions,history of gynecological diseases,and other factors,we retrospectively summarized and compared the disease history of patients with primary breast cancer and patients with benign thyroid tumors admitted to our hospital in the past 10 years to explore the clinicopathological characteristics and risk factors for primary breast cancer.AIM To investigate the clinical and pathological features and risk factors for primary breast cancer treated at our center in order to provide a reference for the prevention and treatment of breast cancer in the Zhuhai-Macao region.METHODS Through a retrospective case-control study,149 patients with primary breast cancer diagnosed and treated at Zhuhai Hospital of Guangdong Provincial Hospital of Traditional Chinese Medicine from January 2013 to March 2020 were included as a case group,and 165 patients with benign breast tumors diagnosed and treated from January 2019 to March 2020 were included as a control group.The data collected included age,age at menarche,age at first birth,number of births,number of miscarriages,lactation time,history of surgery for benign breast lesions,history of familial malignant tumors,history of gynecological diseases,history of thyroid diseases,and the tumor characteristics of the patients in the case group including pathological diagnosis,pathological type,tumor size,lymph node metastasis,distant metastasis,stage,and molecular classification,among others.In the case group,the chi-square test was used to analyze the clinical and pathological features of pat

Cylindrical polymer brushes-anisotropic unimolecular micelle drug delivery system for enhancing the effectiveness of chemotherapy

Polymer systems can be designed into different structures and morphologies according to their physical and chemical performance requirements,and are considered as one of the most promising controlled delivery systems that can effectively improve the cancer therapeutic index.However,the majority of the polymer delivery systems are designed to be simple spherical nanostructures.To explore morphology/size-oriented delivery performance optimization,here,we synthesized three novel cylindrical polymer brushes(CPBs)by atom transfer radical polymerization(ATRP),which were cellulose-g-(CPT-b-OEGMA)(CCO)with different lengths(~86,~40,and~21 nm).The CPBs are composed of bio-degradable cellulose as the carrier,poly(ethylene glycol)methyl ether methacrylate(OEGMA)as hydrophily block,and glutathione(GSH)-responsive hydrophobic camptothecin(CPT)monomer as loaded anticancer drug.By controlling the chain length of the initiator,three kinds of polymeric prodrugs with different lengths(CCO-1,CCO-2,and CCO-3)could be self-organized into unimolecular micelles in water.We carried out comparative studies of three polymers,whose results verified that the shorter CPBs exhibited higher drug release efficiency,more cellular uptake,and enhanced tumor permeability,accompanied by shortened blood circulation time and lower tumor accumulation.As evidenced by in vivo experiments,the shorter CPBs exhibited higher anti-tumor efficiency,revealing that the size advantage has a higher priority than the anisotropic structure advantage.This provided vital information as to design an anisotropic polymer-based drug delivery system for cancer therapy.

为了解高邮湖优势沉水植物菹草剧烈变化的原因及其时空演变对区域水环境的影响,利用2019—2023年长时序多光谱卫星遥感影像,结合现场和水质自动站监测,对高邮湖菹草与水质开展长期观测。结果显示,菹草的生长与水温、浊度等环境因子密切相关,其在生长期对水质具有净化作用,而在衰退期则会产生大量污染物;高邮湖的菹草盖度由2020年的65.5%下降至2023年的5.9%,对区域水质、生物多样性和生态质量指数产生不利影响;菹草的消失与水位变化、藻类影响、鱼类数量及其优势种变化密切相关,高邮湖存在由草型湖泊向藻型湖泊转化的趋势。
