Insights into the role of major bioactive dietary nutrients in lamb meat quality:a review

Feed supplementation withα-linolenic acid(ALA)and linoleic acid(LA)increases their content in muscle,ALA increases n-3 polyunsaturated fatty acids and decrease n-6/n-3 ratio in muscle,and LA increases rumenic acid.However,high LA supplementation may have negative effects on lambs’lipid oxidative stability of meat.When the sources of ALA and LA are fed as fresh forage,the negative effects are counterbalanced by the presence of other bioactive compounds,as vitamin E(mainlyα-tocopherol)and polyphenols,which delay the lipid oxidation in meat.There is a wide consensus on the capability of vitamin E delaying lipid oxidation on lamb meat,and its feed content should be adjusted to the length of supplementation.A high dietary inclusion of proanthocyanidins,phenolic compounds and terpenes reduce the lipid oxidation in muscle and may improve the shelf life of meat,probably as a result of a combined effect with dietary vitamin E.However,the recommended dietary inclusion levels depend on the polyphenol type and concentration and antioxidant capacity of the feedstuffs,which cannot be compared easily because no routine analytical grading methods are yet available.Unless phenolic compounds content in dietary ingredients/supplements for lambs are reported,no specific association with animal physiology responses may be established.

Recent Advances in Single-Cell Metabolomics Based on Mass Spectrometry

Cellular heterogeneity is essential for the physiological functions of organisms,and precise interpretation of the relevant biological mechanisms involved requires accurate measurement of biomolecules,including DnA,RnA,proteins,and metabolites at the single-cell level.Cellular metabolites react most rapidly to environmental and biochemical changes such that their detection is a critical step in determining the physiological state of cells.However,their limited amounts,structural diversity,and significant content variation render single-cell metabolomics analysis technically challenging.Mass spectrometry,with its high sensitivity,good selectivity,and wide dynamic range,has been widely used in the single-cell analysis in recent years.In terms of ionization techniques,there are four main types of single-cell mass spectrometry methods:nano-electrospray ionization mass spectrometry,matrix-assisted laser desorption/ionization mass spectrometry,secondary ion mass spectrometry,and dielectric barrier discharge ionization mass spectrometry.In this mini review,the progress of single-cellmass spectrometrymethodologies and data processing strategies are summarized,and the future trends in single-cell mass spectrometry are outlined.

Influence of parameters on the microstructure of a duplex stainless steel joint welded by a GMAW welding process

Duplex stainless steels(DSSs) are widely used in industry for their desirable properties. Any fusion welding technique modifies the microstructure and, consequently, the properties of the material. This paper presents a parametric study of the variables that determine the energy contribution(current, voltage and welding speed) and the structural modifications produced during gas metal arc welding. The analysis comprises metallographic study,phase balance, dimensioning of the weld beads and tensile and hardness tests. The influence of the variables studied on the thermal cycles obtained(temperatures reached and cooling rate) was determined by using a finite element model. The results showed that, in addition to the indicated parameters, the parameter power transfer per volume(W/m3) and the geometry in the bead must be controlled due to their influence on the heat flow distribution and phase balance.

The scientific community has focused the agenda of studies of climate change on lower-end warming and simple risk analyses,because more realistic complex assessments of risk are more difficult,the benchmark of the international targets is the Paris Agreement goal of limiting warming to nd the culture of climate science is to try to avoid alarmism[1].Current fires,prolonged droughts,floods and heat waves,together with the consequent food insecurity,civil unrest and migrations,however,are opening the eyes not only of most scientists but also of most people all over the world to the need for considering,at least,the potential catastrophic effects of the collapse of ecosystems and society due to the current emergency of climate change.

An optimized high quality male DnA extraction from spermatophores in open thelycum shrimp species

The crucial step of most of the current genetic studies is the extraction of DnA of sufficient quantity and quality.Several genomic DnA isolation methods have been described to successfully obtain male DnA from shrimp species.However,all current protocols require invasive handling methods with males for DnA isolation.Using Aristeus antennatus as a model we tested a reliable non-invasive differential DnA extraction method to male DnA isolation from spermatophores attached to female thelycum.The present protocol provides high quality and quantity DnA for polymerase chain reaction amplification and male genotyping.This new approach could be useful to experimental shrimp culture to select sires with relevant genetic patterns for selective breeding programs.More importantly,it can be applied to identify the mating pairs and male structure in wild populations of species as A.antennatus,where males are often difficult to capture.Our method could be also valuable for biological studies on other spermatophore-using species,such as myriapods,arachnids and insects.

Shaken not stirred: A molecular contribution to the systematics of genus Mugil (Teleostei, Mugilidae)

With this work we addressed some molecular systematic issues within the Mugil cephalus species complex.Particular attention was paid to the debated situations of:(i)Mugil liza,occurring in partial sympatry with Mugil cephalus in the northwestern Atlantic,and(ii)Mugil platanus,considered by some authors a synonymy of the former species and distributed in the southwestern Atlantic.We sequenced 79 individuals of a 465-bp portion of the mitochondrial control region(CR)from 8 western Atlantic and 2 Mediterranean localities.In addition,all CR sequences available from GenBank for the studied taxa were added to our dataset,for a total of 323 individuals.Overall,229 haplotypes corresponding to 8 divergent monophyletic lineages were detected.Results of phylogenetic analyses were consistent with the occurrence of past speciation events producing the observed lineages.Of these lineages,7 correspond to cryptic species and one is constituted by M.liza and M.platanus.As a matter of fact,these 2 taxa constitute a single lineage within the M.cephalus species complex.However,individuals of M.liza/M.platanus lineage analyzed by means of the 18 mitochondrial markers available in GenBank exhibited a degree of genetic diversity consistent with highly divergent populations.Of the 8 lineages detected,the Mediterraean one(type locality)corresponds to M.cephalus;the lineage M.liza/M.platanus should be named M.liza,under the priority principle,and the left 6 lineages need formal description.

Monkeypox virus:a re-emergent threat to humans

Human monkeypox(MPX)is a rare zoonotic infection characterized by smallpox-like signs and symptoms.It is caused by monkeypox virus(MPXV),a double stranded DnA virus belonging to the genus Orthopoxvirus.MPX was first identified in 1970 and mostly prevailed in the rural rainforests of Central and West Africa in the past.Outside Africa,MPX was reported in the United Kingdom,the USA,Israel,and Singapore.In 2022,the resurgence of MPX in Europe and elsewhere posed a potential threat to humans.MPXV was transmitted by the animals-human or human-human pathway,and the symptoms of MPXV infection are similar to that of smallpox,but in a milder form and with lower mortality(1%–10%).Although the smallpox vaccination has been shown to provide 85%protection against MPXV infection,and two anti-smallpox virus drugs have been approved to treat MPXV,there are still no specific vaccines and drugs against MPXV infection.Therefore it is urgent to take active measures including the adoption of novel anti-MPXV strategies to control the spread of MPXV and prevent MPX epidemic.In this review,we summarize the biological features,epidemiology,pathogenicity,laboratory diagnosis,and prevention and treatment strategies on MPXV.This review provides the basic knowledge for prevention and control of future outbreaks of this emerging infection.

Measurements of Laminar Burning Velocities of Alternative Fuels:Application in Transport and Aerospace

The laminar burning velocity belongs to the fundamental combustion properties of fuels being a measure for their heat release,flame length,as well as reactivity and combustion stability,and thus,may impact the design of burners and combustion chambers.Also,these experimental data are needed for the validation and optimization within the construction and development of detailed chemical kinetic reaction mechanisms.Within this study,an overview of the different applications of fuel characterization regarding the specific area of interest(road transport,aviation,and aerospace)will be given.Depending on the application,effects of different molecular characteristics on the laminar burning velocity are evaluated:the presence of oxygen atoms and the grade of branching in a specific fuel molecule as well as the difference in the type of a chemical bond,here,single and double bonds.Examples of alternative fuels being discussed in the present study are:(Ⅰ)oxymethylene ether(OMEn)in the field of road transport;(Ⅱ)a paraffinic Alcohol-to-Jet fuel as sustainable aviation fuel;and(Ⅲ)mixtures of ethane or ethene with nitrous oxide as green propellants for rocket propulsion applications.

Sarcomas constitute a rare heterogeneous group of tumors,including a wide variety of histological subtypes.Despite advances in our understanding of the pathophysiology of the disease,first-line sarcoma treatment options are still limited and new treatment approaches are needed.Histone H2AX phosphorylation is a sensitive marker for double strand breaks and has recently emerged as biomarker of DnA damage for new drug development.In this study,we explored the role of H2AX phosphorylation at Ser139 alone or in combination with MAP17 protein,an inducer of DnA damage through ROS increase,as prognostic biomarkers in sarcoma tumors.next,we proposed doxorubicin and olaparib combination as potential therapeutic strategies against sarcomas displaying high level of both markers.We evaluate retrospectively the levels of pH2AX(Ser139)and MAP17 in a cohort of 69 patients with different sarcoma types and its relationship with clinical and pathological features.We found that the levels of pH2AX and MAP17 were related to clinical features and poor survival.next,we pursued PARP1 inhibition with olaparib to potentiate the antitumor effect of DnA damaging effect of the DnA damaging agent doxorubicin to achieve an optimal synergy in sarcoma.We demonstrated that the combination of olaparib and doxorubicin was synergistic in vitro,inhibiting cell proliferation and enhancing pH2AX intranuclear accumulation,as a result of DnA damage.The synergism was corroborated in patient-derived xenografts(PDX)where the combination was effective in tumors with high levels of pH2AX and MAP17,suggesting that both biomarkers might potentially identify patients who better benefit from this combined therapy.

MYC plays a central role in tumorigenesis by orchestrating cell proliferation,growth and survival,among other transformation mechanisms.In particular,MYC has often been associated with lymphomagenesis.In fact,MYC overexpressing lymphomas such as high-grade B-cell lymphoma(HGBL)and double expressor diffuse large B-cell lymphomas(DLBCL),are considered addicted to MYC.In such a context,MYC targeting therapies are of special interest,as MYC withdrawal is expected to result in tumor regression.However,whether high MYC levels are always predictive of increased sensitivity to these approaches is not clear yet.Even though no MYC inhibitor has received regulatory approval to date,substantial efforts have been made to investigate avenues to render MYC a druggable target.Here,we summarize the different classes of molecules currently under development,which mostly target MYC indirectly in aggressive B-cell lymphomas,paying special attention to subtypes with MYC/BCL2 or BCL6 translocations or overexpression.
