There is scarce information about the effects of danazol and its derivatives at cardiovascular level. in addition, to date the cellular site and mechanism of action of danazol at the cardiovascular level is very confusing. in order to clarify those phenomena in this study, a danazol derivative was synthesized with the objective of evaluating its activity on perfusion pressure and coronary resistance and comparing this phenomenon with the effect exerted by danazol. The Langendorff technique was used to measure changes on perfusion pressure and coronary resistance in an isolated rat heart model in the absence or presence of danazol and its derivative. Additionally, to characterize the molecular mechanism involved in the inotropic activity induced by danazol derivative was evaluated by measuring left ventricular pressure in the absence or presence of following compounds;flutamide, prazosin, metoprolol, indomethacin and nifedipine. The results showed that danazol derivative significantly increased the perfusion pressure and coronary resistance in comparison with the control conditions and danazol. Additionally, other data indicate that the danazol derivative increases left ventricular pressure in a dose-dependent manner;nevertheless, this phenomenon was significantly inhibited by flutamide. These data suggest that danazol derivative induces positive inotropic activity through of the activation androgen receptor.

Fly and bottom ash(FABA)produced primarily from coal combustion in a coal-fred power plant consists of fne particles of the organic and inorganic mixture and trace *** FABA samples from coal-fred power plants in Java,indonesia,had been collected for composition and rare earth element and Yttrium(REY)mode of occurrence *** geochemical composition of both major and trace elements was determined by inductively coupled plasma-mass spectrometry/atomic emission spectrometry(iCP-mS/AES).Furthermore,the composition of FABA was identifed by petrography with refected light microscopy and X-ray difraction(XRD)***,scanning electron microscope determines the mode of occurrence of REY with an energy-dispersive X-ray(SEm–EDX).The study fnds that the inorganic component of FABA consists of glass,Fe–mg spinel,mullite,quartz,Fe-oxide mineral,and *** contrast,the organic component is dominated by unburned *** is the most abundant component with cenospheres as major and pleiospheres as minor *** elements analysis indicates REY concentration with heavy REY(HREY)distribution ***,SEm–EDX analysis results show that Yttrium(Y)occurs in glass and has a low concentration in *** the mode occurrence of REY,in particular Yttrium,it can be predicted that alkaline fusion followed by acid leaching will be the most appropriate extraction method to extract REY from indonesian FABA.

Herein,the rational design micromilieus involved silk fibroin(SF)-based materials have been used to encapsulate the osteoblasts,forming an extracellular coated shell on the cells,which exhibited the high potential to shift the regulation of osteoblasts to osteocytes by encapsulation *** coating treated cells showed a change in cell morphology from osteoblasts-like to osteocytes-like shape comparedwith untreated ***,the expression of alkaline phosphatase(ALP),collagen i(Col i)and osteocalcin(ocN)further indicated a potential approach for inducingosteoblasts regulation,which typically accelerates calcium deposition and cell calcification,presenting a key role for the SF encapsulation in controlling osteoblasts *** discovery showed that SF-based cell encapsulation could be used for osteoblasts behavior regulation,which offers a great potential to modulate mammalian cells'phenotype involvingalternatingsurrounding cues.

目的:观察低氧促进大鼠骨骼肌成肌细胞增殖的作用,并分析CoCl2对成肌细胞增殖的影响。方法:实验于2005-05/2006-07在解放军军事医学科学院基础医学研究所神经肌肉发育研究室完成。①低氧Co2温箱(Forma Scientific,美国);CoCl2(北京化工厂)。②选取4～5周龄Wistar雄性大鼠5只,脱颈处死无菌切取后腿肌群,剪除脂肪和筋膜,制备单细胞悬液。以连续贴壁法筛选大鼠骨骼肌成肌细胞,接种于96孔板进行单克隆培养。采用成肌细胞特异性标志抗原desmin免疫化学染色,鉴定成肌细胞标志蛋白——结蛋白的表达,弃去desmin阴性的细胞克隆,继续培养desmin阳性的细胞克隆,隔天换液1次,7d进行酶消化传代,获得大量扩增的细胞,并可冻存复苏,用于实验。③以1×107L-1接种于20个35mm培养皿中,接种细胞3h后,将培养皿随机数字表法分为5组:常氧对照组、轻度低氧组、中度低氧组、CoCl2组、轻度低氧+CoCl2组,4皿/组。常氧对照组置于体积分数为0.2的o2常规氧气环境中;轻、中度低氧组分别于低氧温箱中维持体积分数为0.1与0.03的o2低氧环境中;CoCl2组向培养皿中加入终浓度为15μmol/L的CoCl2;轻度低氧+CoCl2组向细胞培养皿中加入终浓度为15μmol/L的CoCl2后,放入体积分数为0.1的o2低氧温箱。低氧培养24,48,72h时,各组取出培养皿进行细胞消化离心,倒置相差显微镜下观察细胞生长情况,血球计数板计数法进行细胞计数。④以1×108L-1接种于8个60mm培养皿中,接种细胞3h后,将培养皿随机数字表法分为2组:常氧对照组、轻度低氧组,4皿/组。两组干预措施同细胞计数过程。低氧培养48h时,两组取出培养皿进行细胞消化离心,流式细胞仪检测细胞周期。结果:①骨骼肌成肌细胞的单克隆培养结果:成肌细胞可在体外存活6个月,增殖旺盛期约为3个月,可传代15次以上。单克隆培养2周后,可以由单个细胞长满96孔板中的1个孔,并不断扩增,最终可以得到细胞类型专一单克隆化的成肌细胞。②成肌细胞特异性标志抗原desmin鉴定结果:镜下不同视野desmin阳性率达100%,即培养的成肌细胞单克隆纯度达100%。③细胞计数结果:与常氧对照组比较,低氧培养24,48,72h时轻、中度低氧组均可明显促进大鼠骨骼肌成肌细胞体外增殖,且轻度低氧组作用尤为明显(t=4.98,PoCl2组无明显变化,但轻度低氧+CoCl2组细胞数量显著增加(t=4.62,PoCl2对骨骼肌成肌细胞没有促增殖作用。

Achieving intravital optical imaging with diffraction-limited spatial resolution of deep-brain structures represents an important step toward the goal of understanding the mammalian central nervous system1–*** in wavefrontshaping methods and computational power have recently allowed for a novel approach to high-resolution imaging,utilizing deterministic light propagation through optically complex media and,of particular importance for this work,multimode optical fibers(mmFs)5–*** report a compact and highly optimized approach for minimally invasive in vivo brain imaging *** volume of tissue lesion was reduced by more than 100-fold,while preserving diffraction-limited imaging performance utilizing wavefront control of light propagation through a single 50-μm-core ***,we demonstrated high-resolution fluorescence imaging of subcellular neuronal structures,dendrites and synaptic specializations,in deep-brain regions of living mice,as well as monitored stimulus-driven functional Ca2+*** results represent a major breakthrough in the compromise between high-resolution imaging and tissue damage,heralding new possibilities for deep-brain imaging in vivo.

Exploring the potential for an evolutionarily conserved role of the taste 1 receptor gene family in gut sensing mechanisms of fish

in this study,we investigated the transcriptional spatio-temporal dynamics of the taste 1 receptor(T1R)gene family repertoire in seabream(Sparus aurata[sa]),during larval ontogeny and in adult *** early larval development,sa T1R expression arises heterochronously,*** extraoral taste-related perception in the gastrointestinal tract(GiT)anticipates first exogenous feeding(at 9 days post hatching[dph]),followed by the buccal/intraoral perception from 14 dph onwards,supporting the hypothesis that the early onset of the molecular machinery underlying sa T1R expression in the GiT is not induced by food but rather genetically *** adulthood,we characterized the expression patterns of sa T1R within specific tissues(n=4)distributed in oropharingeal,GiT and brain regions substantiating their functional versatility as chemosensory signaling players to a variety of biological functions beyond oral taste ***,we provided for the first time direct evidences in fish for m RNA coexpression of a subset of sa T1R genes(mostly sa T1R3,*** common subunit of the heterodimeric T1R complexes for the detection of“sweet”and“umami”substances),with the selected gut peptides ghrelin(ghr),cholecystokinin(cck),hormone peptide yy(pyy)and proglucagon(pg).Each peptide defines the enteroendocrine cells(ECCs)identity,and establishes on morphological basis,a direct link for T1R chemosensing in the regulation of fish digestive ***,we analyzed the spatial gene expression patterns of 2 taste signaling components functionally homologous to the mammalian G(i)a subunit gustducin,namely sa G(i)a1 and sa G(i)a2,and demonstrated their co-localization with the sa T1R3in EECs,thus validating their direct involvement in taste-like transduction mechanisms of the fish *** conclusion,data provide new insights in the evolutionary conservation of gut sensing in fish suggesting a conserved role for nutrient sensors modulating entero-endocrine secretion.

Rooftop photovoltaic （PV） halls： Achieving economic energy demand systems for industrial benefit via lowering

industrial hails are characterized with their retatively high roof-to-floor ratio, which facilitates ready deployment of renewable energy generation, such as photovoltaic （PV） systems, on the rooftop. To promote deployment of renewable energy generation, feed-in tariff （FiT） higher than the electricity rate is available in many countries to subsidize the capital investment. FiT comes in different forms. For net FiT, in order to maximize the economic benefit, surplus electridty generation at each hour is desirable. one way to achieve surplus electricity generation is by increasing generation capacity, which is synonymous to higher capital investment. in fact, surplus electricity generation can also be achieved by lowering the energy demand of the building. This particularly the case for industrial hatls, which are usually subject to high energy demand for space conditioning in order to remove the excess heat gain due to the many power-intensive processes. Building energy performance simulation toots can be used to explore the different building design options that could lower the energy demand. in this paper, single-objective optimization on investment return will be deployed to study the cost effectiveness among different options in lowering energv demand. it Will-be demonstrated with a case study of a warehouse.

Factors that contribute to the mycoparasitism stimulus in Trichoderma atroviride. strain P1

Trichoderma atroviride strain P1 has been used extensively to study the mycoparasitic mechanisms in the interaction between plant pathogenic host and beneficial antagonistic fungi. mutants of P1 containing the green fluorescent protein (gfp) or glucose oxidase (gox) reporter systems and different inducible promoters (from the exochitinase nag1 gene, or the endochitinase ech42 gene of P1) were used to determine the factors that activate the biocontrol gene expression cascade in the antagonist. The following compounds were tested singly and in various combinations: purified Trichoderma P1 enzymes (endochitinase, exochitinase, chitobiosidase, glucanase); antagonist culture filtrates (T. atroviride P1 wild-type and relative knock-out mutants, T. harzianum, T. reesei); pathogen culture filtrates (Botrytis, Pythium, Rhizoctonia); purified fungal cell walls (CWs) from Trichoderma, Botrytis, Pythium, Rhizoctonia; colloidal crab shell chitin; and plant extracts from cucumber leaves, stems or roots. Strong induction of mycoparasitism was found with the various digestion products produced by treating fungal CWs and colloidal chitin with purified enzymes or fungal culture filtrates. Filtrates from chitinase knock-out mutants, as well as CWs from oomycetes fungi, were less active in producing the stimulus for mycoparasitism. The host CW digestion products were separated by molecular weight (mW) to determine which compounds were able to activate Trichoderma. micromolecules of mW less than 3 kDa were found to trigger mycoparasitism gene expression before physical contact with the host pathogen. These compounds stimulated mycelial growth and spore germination of the antagonist. Purification of these host-derived compounds was conducted by HPLC and in vivo assay. The obtained inducers were able to stimulate both the production of endochitinase and exochitinase enzymes, even under repressing conditions in the presence of glucose. inducers stimulated the biocontrol effect of P1 in the
