Fast,simple,efficient Agrobacterium rhizogenes-mediated transformation system to non-heading Chinese cabbage with transgenic roots

Non-heading Chinese cabbage, a variety of Brassica campestris, is an important vegetable crop in the Yangtze River Basin of China. However,the immaturity of its stable transformation system and its low transformation efficiency limit gene function research on non-heading Chinese cabbage. Agrobacterium rhizogenes-mediated(ARM) transgenic technology is a rapid and effective transformation method that has not yet been established for non-heading Chinese cabbage plants. Here, we optimized conventional ARM approaches(one-step and two-step transformation methods) suitable for living non-heading Chinese cabbage plants in nonsterile environments. Transgenic roots in composite non-heading Chinese cabbage plants were identified using phenotypic detection, fluorescence observation, and PCR analysis. The transformation efficiency of a two-step method on four five-day-old non-heading Chinese cabbage seedlings(Suzhouqing, Huangmeigui, Wuyueman, and Sijiu Caixin) was 43.33%-51.09%, whereas using the stout hypocotyl resulted in a transformation efficiency of 54.88% for the 30-day-old Sijiu *** one-step method outperformed the two-step method;the transformation efficiency of different varieties was above 60%, and both methods can be used to obtain transgenic roots for functional studies within one month. Finally, optimized ARM transformation methods can easily,quickly, and effectively produce composite non-heading Chinese cabbage plants with transgenic roots, providing a reliable foundation for gene function research and non-heading Chinese cabbage genetic improvement breeding.

Due to its simplicity, high efficiency, and chemo-selectivity, bioorthogonal chemistry has shown a great application potential in ***, four bioorthogonal pairs as targeting tools, including (strept)avidin/biotin, antibody/antigen, oligonucleotide hybridization and IEDDA tools, have been developed and applied in targeted ***, all of these tools still suffer from some limitations, such as difficult modification, biochemical fragility and larger molecular weight for biological association tools, as well as chemical instability for IEDDA *** host-guest pairs with relatively small molecular sizes not only possess strong chemical stability, but also have the features of fast conjugation rate, tunable binding affinity , easy modification, and high ***, they can be used as a novel non-covalent bioorthogonal tool for *** order to further promote the development of host-guest pairs as novel bioorthogonal tools for pre-targeted delivery, we firstly calculate their conversion rate to make researcher aware of their unique advantages;next, we summarize the recent research progress in this *** future perspectives and limitations of these unique tools will be *** review will provide a systemic overview of the development of synthetic host-guest pairs as novel bioorthogonal tools for pre-targeting, and may serve as a “go for” resort for researchers who are interested in searching for new synthetic tools to improve pre-targeting.

The printed circuit heat exchanger(PCHE) is receiving wide attention as a new kind of compact heat exchanger and is considered as a promising vaporizer in the LNG process. In this paper, a PCHE straight channel in the length of 500 mm is established, with a semicircular cross section in a diameter of 1.2 *** simulation is employed to investigate the flow and heat transfer performance of supercritical methane in the channel. The pseudo-boiling theory is adopted and the liquid-like, two-phase-like, and vapor-like regimes are divided for supercritical methane to analyze the heat transfer and flow *** results are presented in micro segment to show the local convective heat transfer coefficient and pressure drop. It shows that the convective heat transfer coefficient in segments along the channel has a significant peak feature near the pseudo-critical point and a heat transfer deterioration when the average fluid temperature in the segment is higher than the pseudo-critical point. The reason is explained with the generation of vapor-like film near the channel wall that the peak feature related to a nucleateboiling-like state and heat transfer deterioration related to a film-boiling-like state. The effects of parameters, including mass flow rate, pressure, and wall heat flux on flow and heat transfer were *** calculating of the averaged heat transfer coefficient of the whole channel, the traditional method shows significant deviation and the micro segment weighted average method is adopted. The pressure drop can mainly be affected by the mass flux and pressure and little affected by the wall heat flux. The peak of the convective heat transfer coefficient can only form at high mass flux, low wall heat flux, and near critical pressure, in which condition the nucleate-boiling-like state is easier to appear. This study investigates the activity of tropical cyclones(TCs)in the Bay of Bengal(BOB)from 1979 to 2018 to discover the mechanism affecting the contribution rate to the meridional moisture budget anomaly(MMBA)over the southern boundary of the Tibetan Plateau(SBTP).May and October–December are the bimodal phases of BOB TC frequency,which decreases month by month from October to December and is relatively low in ***,the contribution rate to the MMBA is the highest in *** seasonal variation in the meridional position of the westerlies is the key factor affecting the contribution *** relatively southern(northern)position of the westerlies in November and December(May)results in a lower(higher)contribution rate to the *** mechanism is confirmed by the momentum *** water vapor enters the westerlies near the trough line,the resultant meridional acceleration is directed *** follows that the farther north the trough is,and the farther north the water vapor can be *** water vapor enters the westerlies from the area near the ridge line,for Type-T(Type-R)TCs,water vapor enters the westerlies downstream of the trough(ridge).Consequently,the direction of the resultant meridional acceleration is directed south and the resultant zonal acceleration is directed east(west),which is not conducive to the northward transport of water *** is especially the case if the trough or ridge is relatively south,as the water vapor may not cross the SBTP.

The biosecurity hazards caused by pathogenic fungus have been widely *** the long-term coexistence of eukaryotic pathogens and quorum sensing bacteria in different habitats in environments,we hypothesized that they have social interactions via signal *** this work,we firstly discovered the well-known bacterial signal molecules play an adverse role in the cell morphology and metabolism in a model pathogen Trichosporon asahii.N-Tetradecanoyl-L-homoserine lactone(C14-HSL)was discovered to increase pathogen hazards of ***,which limited mycelium by 52%,but enhanced cell aggregation by 93%.Higher fluorescence intensity of tryptophan(59%)and aromatic protein(2-fold)contents after the treatment of C14-HSL,indicating that aromatic proteins helped aggregate Trichosporon and showed *** analysis revealed that C14-HSL upregulated the shikimate pathway(above1-fold)located in downstream of tricarboxylic acid cycle,which contributed to the synthesis of more aromatic proteins and the formation of larger *** limited mycelial growth of *** attributed to the up-regulated expressions of cell cycle *** fungal transboundary response to bacterial C14-HSL was controlled by signal transduction *** study provides new insights into the co-evolution of bacterial and pathogenic fungi in microecosystems.

Jiaojiang sag in the East China Sea Basin is at the earlier exploration stage,where characterizing hydrocarbon generation of source rocks is important to understand oil-gas exploration *** geochemical and basin modeling analysis,hydrocarbon generation capacity and process of the Paleocene E_(1)y,E_(1)l and E_(1)m formations were *** show that E_(1)y and E_(1)l mudstones are high-quality source rocks with Type Ⅱ kerogen,which is dominated by both aquatic organisms and terrestrial higher plants deposited in sub-reduced environment.E_(1)m mudstone interbedded with thin carbonaceous mudstone and coal is poor-quality source rock with Type Ⅲ kerogen,whose organic matter was originated from terrestrial higher plants under oxidized *** by burial and maturity histories,E_(1)y and E_(1)l source rocks experienced two hydrocarbon generation stages,which took place in the Late Paleocene and in the Middle to Late Eocene,respectively,and had high hydrocarbon generation capacity with cumulative hydrocarbon volume of 363 and 328 mg/g,respectively.E_(1)m source rock only had one hydrocarbon generation process in the Late Eocene,which had low hydrocarbon generation capacity with cumulative hydrocarbon volume of only 24 mg/*** future oil-gas exploration in the Jiaojiang sag should focus on hydrocarbon generation center and select targets in the central uplift formed before the Miocene with high-quality traps.
