A classical function of Clq is to bind immune complexes and initiate complement activation producing membrane lytic complexes, opsonins and anaphylatoxins. This classical pathway of complement activation is also elicited when Clq binds some other ligands. Besides complement activation, Clq also regulates cell differentiation, adhesion, migration, activation and survival. Clq deficiency is associated with autoimmunity as well as increased susceptibility to infections. AIM:To investigate the impact of esophagogastroduodenoscopy with conscious sedation on the subsequent 24-h catheter-based pH ***:Fifty patients with extra-esophageal symptoms of gastroesophageal reflux disease undergoing ambulatory dual-probe 24-h pH monitoring were enrolled from March 2010 to August *** of the data were collected prospectively and analyzed ***-six patients(72%,group A) underwent pH monitoring shortly after esophagogastroduodenoscopy(EGD) with conscious sedation,and 14 patients(28%,group B) underwent pH monitoring without conscious *** 24-h pH data from two time periods were analyzed:the first 4 h(Period Ⅰ) and the remaining time of the study(Period Ⅱ).RESULTS:The mean age of the patients was 49.6 ± 12.5 years;20 patients(40%) were *** baseline data,including age,sex,body mass index,reflux esophagitis,the Reflux Symptom Index,and the Reflux Findings Score,were comparable between the two *** percentage of total time with a pH < 4 and the frequency of acid reflux during Period Ⅰ were not significantly different between the two groups,as measured using both pharyngeal(0.03% ± 0.10% vs 0.07% ± 0.16%,P = 0.32;and 0.07 ± 0.23 episodes/h vs 0.18 ± 0.47 episodes/h,P = 0.33,respectively) and esophageal probes(0.96% ± 1.89% vs 0.42% ± 0.81%,P = 0.59;and 0.74 ± 1.51 episodes/h vs 0.63 ± 0.97 episodes/h,P = 0.49,respectively).The percentage of total time with a pH < 4 and the frequency of acid reflux were also not significantly different between Periods I and Ⅱ in group A patients,as measured using both pharyngeal(0.03% ± 0.10% vs 0.23% ± 0.85%,P = 0.21;and 0.07 ± 0.23 episodes/h vs 0.29 ± 0.98 episodes/h,P = 0.22,respectively) and esophageal probes(0.96% ± 1.89% vs 1.11% ± 2.57%,P = 0.55;and 0.74 ± 1.51 episodes/h vs 0.81 ± 1.76 episodes/h,P = 0.55,respectively).CONCLUSION:EGD with conscious sedation does not interfere with the results of subsequent 24-h pH monitoring in patients with extra-esophageal

Objective To study the role of JAK2/STAT3 in IL-6-induced activation of human proximal tubular epithelial cells (HK-2 cells). Methods Cell proliferation was measured by MTT assay. HK-2 cells growing in RPMI 1640 medium containing 10% fetal bovine serum were cultured for 24h in serum-free medium after 60%-80% cells pasted on the wail, and then cultured with IL-6 at designated concentration for designated periods. The expression of a-SMA was assessed by Western blot after the induction of IL-6 at 25 ng/mL concentrations for a period of 24h, 48h, 72h respectively. The expression of P-STAT3 was detected with Western blot after the induction of IL-6 at 25 ng/mL concentration for a period of 0, 15, 30, 60, 120 min respectively and at the concentrations of 0, 5, 10, 25, 50 ng/mL for 30 min. The cells were pre-incubated with 100 uM AG490 for 4h and then stimulated with 25 ng/mi IL-6 for 30 min and 48h respectively, The expression of P-STAT3, P-JAK2 andα-SMA was determined by Western blot. Results Incubation for 48 hours with 10-50 ng/mL rIL-6 caused concentration- and time-dependent stimulation of HK-2 cells proliferation, IL-6 induced the expression ofα-SMA. IL-6 rapidly induced the phosphorylation of STAT3 in a dose- and time-dependent manner with a peak expression at 30 min. 100 IxM AG490, a specific JAK2 inhibitor, partially inhibited STAT3, JAK2 activation and a-SMA production. Conelusion It is suggested that JAK2/STAT3 pathway partially mediate IL-6-induced et-SMA production in HK-2 cells. It is implicated that JAK2/STAT3 signaling pathway might play a role in the development of tubulointerstitial fibrosis.

Objective: Rac1 and epithelial-mesenchymal transition（EMT） is an important target in new cancer treatment strategies. We investigated the clinical significance of Rac1 and markers of EMT expression in colon cancer, and their possible correlation with EMT phenotype. Methods: Immunohistochemistry was used to assess the expressions of Rac1, Snail1, Twist1, N-cadherin（Ncad）, Vimentin（Vim）, and E-cadherin（E-cad） in 126 colon cancer paraffin-embedded specimens and 50 tumor-adjacent normal specimens. The association of Rac1 and EMT markers with clinicopathological characteristics and relationships between the protein levels and progression free survival（PFS） and overall survival（OS） were analyzed. Results: Compared with non-tumorous tissues, Rac1, Snail1, Twist1, N-cad, Vim levels were markedly increased in colon cancer tissues, whereas decreased E-cad levels were observed（P ＜ 0.05）. Based on immunohistochemical evaluation, the statistical analysis suggested that Rac1 and EMT markers aberrant expression were significantly associated with TNM stage and metastasis（P ＜ 0.05）. Patients who displayed high expression of Rac1 may achieve a poorer OS and PFS, compared to those with low expression of Rac1（P＜0.001）. Furthermore, significant correlations were observed between every EMT markers’ expression levels and OS or PFS（P＜0.01）. In addition, among the clinicopathologic characteristics, multivariateanalysis indicated Rac1, Snail1, Twist1, N-cad, Vim, and E-cad expression respectively could be as an independent prognostic factor in colon cancer. Rac1 expression was positively correlated with Snail1, Twist1, N-cad, Vim levels（P＜0.01） and negatively correlated with E-cadherin amounts（P＜0.001） in colon cancer tissues. Conclusion: Rac1 may promote colon cancer progression and metastasis by involving EMT program, which may be considered a potential therapeutic target in colon cancer.

Objective: PHB1 has been proved tumor-associated and functions as tumor suppressor in multiple cancer cells. This study aimed to investigate the effects of PHB1 on the proliferation in human nasopharyngeal carcinoma（NPC） cell lines and its mechanisms. Method: PHB1 was stably overexpressed in CNE1 and HNE1 cells. Cell proliferation was assessed by colony formation and CCK8 assay. Cell-cycle distribution and apoptosis were analyzed using flow cytometry. Luciferase reporter assays, immunofluorescent assay, western blot and quantitative RT-PCR were used to detect the NF-κB activity and downstream signaling pathway expression. Results: PHB1 significantly inhibited the proliferation of NPC cell lines CNE1 and HNE1 in vitro. PHB1 induced an apparent cell cycle arrest at the G1/S phase and apoptosis, which was accompanied by an obvious inactivation of the NF-κB signaling pathway. PHB1 led to a remarkable decrease the expression of NF-κB p65, IKKβ, cyclin D1, cyclin-dependent kinase（CDK） 4 and Bcl-2, obviously increased the expression of IκBα, p21, Bax at both mRNA and protein levels in NPC cells. In addition, PHB1 restrained the LPS-induced activation of NF-κB signaling via inhibiting p65 translocation into nuclear. Conclusion: PHB1 could inhibit cell proliferation in vitro via inactivation of the NF-κB signaling pathway. PHB1 maybe serve as a tumor suppressor gene and a potential therapeutic target against NPC.

目的分析630 nm发光二极管(light emitting diode,LED)产生红光照射对人单核细胞白血病细胞(THP-1)来源M0巨噬细胞的作用与机制。方法用佛波酯(phorbol-12-myristate-13-acetate,PMA)处理THP-1细胞,制备THP-1来源M0巨噬细胞,并使用强度为28.8 J/cm^(2)的630 nm LED红光照射M0巨噬细胞,随后采用非标定量蛋白质组学分析方法筛选差异表达蛋白并进行基因本体论(gene ontology,GO)注释和富集;通过string在线网站(https://***/)和Cytoscape软件分析蛋白质互作情况,根据关键蛋白的作用,分析630 nm LED红光照射与THP-1来源M0巨噬细胞的关系。结果蛋白质组学结果显示,630 nm LED红光照射THP-1来源M0巨噬细胞后,62个差异表达蛋白能够显著富集在细胞对刺激反应、线粒体膜电势负性调控和平滑肌细胞增殖的调控等相关生物学进程。在显著性排名前10的生物学进程中,细胞对刺激反应相关生物学进程占70%,主要为对糖刺激反应(4个进程)、对神经生长因子反应(2个进程)等;进一步分析细胞应对刺激相关生物学进程中涉及的8个蛋白间蛋白互作情况结果显示,转录因子叉头箱蛋白O3亚基(forkhead box O3,FOXO3)、炎症相关蛋白前列腺素内过氧化物合酶2(prostaglandin-endoperoxide synthase 2,PTGS2)或称环加氧酶2(cyclooxygenase-2,COX-2)和炎症因子白介素-18(interleukin-18,IL-18)为关键互作蛋白,并且在630 nm LED红光照射后的THP-1来源M0巨噬细胞中,FOXO3和IL-18的蛋白水平显著升高(P<0.05),PTGS2(COX-2)蛋白水平显著降低(P<0.05)。结论630 nm LED红光照射对THP-1来源M0巨噬细胞的作用,类似于糖和神经生长因子对细胞的刺激反应;通过上调THP-1来源M0巨噬细胞中FOXO3蛋白的表达抑制M0巨噬细胞中PTGS2(COX-2)介导的炎症状态。
