Comprehensive evaluation of the metabolic effects of porcine CRTC3 overexpression on subcutaneous adipocytes with metabolomic and transcriptomic analyses

Background:Meat quality is largely driven by fat deposition,which is regulated by several genes and signaling *** cyclic adenosine monophosphate(cAMP)-regulated transcriptional coactivator 3(CRTC3)is a coactivator of cAMP response element binding protein(CREB)that mediates the function of protein kinase A(PKA)signaling pathway and is involved in various biological processes including lipid and energy ***,the effects of CRTC3 on the metabolome and transcriptome of porcine subcutaneous adipocytes have not been studied ***,we tested whether porcine CRTC3 expression would be related to fat deposition in Heigai pigs(a local fatty breed in China)and Duroc×Landrace×Yorkshire(DLY,a lean breed)pigs in *** effects of adenovirus-induced CRTC3 overexpression on the metabolomic and transcriptomic profiles of subcutaneous adipocytes were also determined in vitro by performing mass spectrometry-based metabolomics combined with RNA sequencing(RNA-seq).Results:Porcine CRTC3 expression is associated with fat deposition in *** addition,CRTC3 overexpression increased lipid accumulation and the expression of mature adipocyte-related genes in cultured porcine subcutaneous *** to the metabolomic analysis,CRTC3 overexpression induced significant changes in adipocyte lipid,amino acid and nucleotide metabolites in *** RNA-seq analysis suggested that CRTC3 overexpression alters the expression of genes and pathways involved in adipogenesis,fatty acid metabolism and glycerophospholipid metabolism in ***:We identified significant alterations in the metabolite composition and the expression of genes and pathways involved in lipid metabolism in CRTC3-overexpressing *** results suggest that CRTC3 might play an important regulatory role in lipid metabolism and thus affects lipid accumulation in porcine subcutaneous adipocytes.

The North China Craton （NCC） is one of the oldest continental fragments, which underwent multistage crust growth during the early Precambrian from 〉3.8 Ga to 1.8 Ga. In previous studies, many zircon ages have been obtained from the exposed basement rocks. However, we know little about the hidden basement because of Phanerozoic to Cenozoic strata coverage. The drill holes provided a rare opportunity to obtain the basement rocks beneath the Bohai Bay Basin （BBB）. In this study, we carry out SHRIMP U-Pb dating analysis on two samples from the basement of the BBB, and discuss the geological significance and its relationship with the exposed basement around the BBB.

Broadband interference of received pulse for single normal mode and solitary internal wave localization

To analyze the solitary internal wave(SIW) influence on acoustic field interference pattern and thereby monitor SIW, for broadband pulse signal propagating through SIWs, we adopt a parabolic equation model, and simulate the variation of received single normal mode(SNM) pulse sequence with SIW position, and analyze the spatiotemporal interference characteristics and mechanism of the sequence. Based on the coupled mode theory, we derive the relationship between SIW position and interference striation slope(ISS). The relationship can monitor the SIW position precisely, with greatly improved precision and robustness compared with previous work. Furthermore, based on the SNM interference pattern periodicity in frequency, we also develop another method to estimate SIW range, with better real-time property than the first method. The SIW-normal mode interference relationship can be used for improving acoustic field localization in the presence of SIWs, which has great value for oceanology,marine engineering, marine military and so on. Besides, the relationship can be expanded to other detection for local variation, e.g., detecting metal flaw and locating dramatic variation in seabed topography.

How Digital Economy Affects High-Quality Economic Development:Based on International Comparison

Selective adsorption of paraffins over olefins(mostly C2 and C3) by porous solids could offer tremendous advantages for the purification of olefins, which is, however, challenging as it has stringent requirements on the pore dimensions and pore surface chemistry of the adsorbent. We report here a metal-organic framework(MOF) with novel topology, Y_(3)(μ_(3)-OH)(TCMPB)2(denoted as HIAM-316, TCMPB^(4-)= 1,2,4,5-tetra(4-carboxy-3-methylphenyl)benzene), which shows reversed separation of C_(2)H_(6)/C_(2)H_(4) and C_(3)H_(8)/C_(3)H_(6) simultaneously. The ideal adsorbed solution theory(IAST) selectivities for equimolar binary mixtures of C_(2)H_(6)/C_(2)H_(4) and C_(3)H_(8)/C_(3)H_(6) were calculated to be 1.67 and 1.23 at 1 bar and 298 K for HIAM-316. Its capability of removing paraffines from paraffin/olefin mixtures is validated by breakthrough measurements. Molecular simulations reveal that the preferential adsorption of paraffins over olefins by HIAM-316 may be attributed to the abundant carboxylate oxygen atoms along its channel.

Protective mechanism of Paeoniae Radix Alba against chemical liver injury based on network pharmacology,molecular docking,and in vitro experiments

Objective:To explore and validate the potential targets of Paeoniae Radix Alba(***,Bai Shao)in protecting against chemical liver injury through network pharmacology,molecular docking technology,and in vitro cell ***:Network pharmacology was used to identify the common potential targets of *** and chemical liver *** docking was used to fit the components,which were subsequently verified in vitro.A cell model of hepatic fibrosis was established by activating hepatic stellate cell(HSC)-LX2 cells with 10 ng/mL transforming growth factor-β*** cells were exposed to different concentrations of total glucosides of paeony(TGP),the active substance of ***,and then evaluated using the cell counting kit-8 assay,enzyme-linked immunosorbent assay,and western ***:Analysis through network pharmacology revealed 13 key compounds of ***,and the potential targets for preventing chemical liver injury were IL-6,AKT serine/threonine kinase 1,jun protooncogene,heat shock protein 90 alpha family class A member 1(HSP90AA1),peroxisome proliferator activated receptor gamma(PPARG),PTGS2,and *** Ontology(GO)enrichment analysis indicated the involvement of response to drugs,membrane rafts,and peptide *** Encyclopedia of Genes and Genomes(KEGG)enrichment analysis revealed that the main pathways involved lipid and atherosclerosis and chemical carcinogenesis-receptor *** and albiflorin exhibited strong affinity for HSP90AA1,PTGS2,PPARG,and *** concentrations of TGP can inhibit the expression of COL-I,COL-III,IL-6,TNF-a,IL-1β,HSP-90a,and PTGS2 while increasing the expression of PPAR-γand CASP3 in activated HSC-LX2 ***:*** primarily can regulate targets such as HSP90AA1,PTGS2,PPARG,***,the main active compound of ***,protects against chemical liver injury by reducing the inflammatory response,activating apoptotic proteins,and promoting the apoptosis of activated HSCs.

Steroids and dihydroisocoumarin glycosides from Xylaria *** the one strain many compounds strategy and their bioactivities

The fungus Xylaria ***-15 was isolated from Illigera *** on the one strain many compounds(OSMAC)strategy,the strain was fermented on potato and rice solid media,*** a result,two novel steroids,xylarsteroids A(1)and B(2),which are the first examples of C28-steroid with an unusualβ-andγ-lactone ring,respectively,along with two new dihydroisocoumarin glycosides,xylarglycosides A(3)and B(4),were *** structures were elucidated by spectroscopic methods,X-ray diffraction and electronic circular dichroism(ECD)*** isolated compounds were evaluated for cytotoxicity,DPPH radical scavenging activity,acetylcholinesterase inhibitory and antimicrobial *** 1 exhibited potent AChE inhibitory activity with an IC50 value of 2.61±0.05μmol·L^(−1).Theβ-lactone ring unit of 1 is critical for its AChE inhibitory *** finding was further confirmed through exploring the interaction of 1 with AChE by molecular *** addition,both compounds 1 and 2 exhibited obvious antibacterial activity against Bacillus subtilis with a minimum inhibitory concentration(MIC)of 2μg·mL^(−1).Compounds 3 and 4 exhibited antibacterial activities against Staphylococcus aureus with MICs of 4 and 2μg·mL^(−1),respectively,which also exhibited DPPH radical scavenging activity comparable to the positive control with IC50 values of 9.2±0.03 and 13.3±0.01μmol·L^(−1),respectively.
