Self-assembly of highly ordered DNA origami lattices at solid-liquid interfaces by controlling cation binding and exchange

The surface-assisted hierarchical self-assembly of DNA origami lattices represents a versatile and straightforward method for the organization of functional nanoscale objects such as proteins and nanoparticles.Here,we demonstrate that controlling the binding and exchange of different monovalent and divalent cation species at the DNA-mica interface enables the self-assembly of highly ordered DNA origami lattices on mica surfaces.The development of lattice quality and order is quantified by a detailed topological analysis of high-speed atomic force microscopy(HS-AFM)images.We find that lattice formation and quality strongly depend on the monovalent cation species.Na^(+)is more effective than Li^(+)and K^(+)in facilitating the assembly of high-quality DNA origami lattices,because it is replacing the divalent cations at their binding sites in the DNA backbone more efficiently.With regard to divalent cations,Ca^(2+)can be displaced more easily from the backbone phosphates than Mg^(2+)and is thus superior in guiding lattice assembly.By independently adjusting incubation time,DNA origami concentration,and cation species,we thus obtain a highly ordered DNA origami lattice with an unprecedented normalized correlation length of 8.2.Beyond the correlation length,we use computer vision algorithms to compute the time course of different topological observables that,overall,demonstrate that replacing MgCl_(2) by CaCl_(2) enables the synthesis of DNA origami lattices with drastically increased lattice order.

Background:The Iberian Peninsula comprises one of the largest boundaries between Mediterranean and Eurosiberian vegetation,known as sub-Mediterranean zone.This ecotone hosts many unique plant species and communities and constitutes the low-latitude(warm)margin of numerous central European species which co-occur with Mediterranean vegetation.Two of the main species found in this region are the Eurosiberian European beech(Fagus sylvatica L.)and the Mediterranean Pyrenean oak(Quercus pyrenaica Willd.).It remains unclear how the different physiological and adaptive strategies of these two species reflect their niche partitioning within a subMediterranean community and to what extent phenotypic variation(intraspecific variability)is driving niche partitioning across Eurosiberian and Mediterranean species.Methods:We quantified functional niche partitioning,based on the n-dimensional hypervolume to nine traits related to resource acquisition strategies(leaf,stem and root)plus relative growth rate as an additional wholeplant trait,and the environmental niche similarity between Pyrenean oak and European beech.Further,we analyzed the degree of phenotypic variation of both target species and its relationship with relative growth rates(RGR)and environmental conditions.Plant recruitment was measured for both target species as a proxy for the average fitness.Results:Species’functional space was highly segregated(13.09%overlap),mainly due to differences in niche breadth(59.7%)rather than niche replacement(25.6%),and beech showed higher trait variability,i.e.,had larger functional space.However,both species shared the environmental space,i.e.,environmental niches were overlapped.Most plant traits were not related to abiotic variables or RGR,neither did RGR to plant traits.Conclusions:Both target species share similar environmental space,however,show notably different functional resource-use strategies,promoting a high complementarity that contributes to maintaining a high functionality in sub-Mediterranean ecosystems.Therefore,w

Conventional dendritic cells(cDCs)scan and integrate environmental cues in almost every tissue,including exogenous metabolic signals.While cDCs are critical in maintaining immune balance,their role in preserving energy homeostasis is unclear.Here,we showed that Batf3-deficient mice lacking conventional type 1 DCs(cDC1s)had increased body weight and adiposity during aging.This led to impaired energy expenditure and glucose tolerance,insulin resistance,dyslipidemia,and liver steatosis.cDC1 deficiency caused adipose tissue inflammation that was preceded by a paucity of NK1.1+invariant NKT(iNKT)cells.Accordingly,among antigen-presenting cells,cDC1s exhibited notable induction of IFN-γproduction by iNKT cells,which plays a metabolically protective role in lean adipose tissue.Flt3L treatment,which expands the dendritic cell(DC)compartment,mitigated diet-induced obesity and hyperlipidemia in a Batf3-dependent manner.This effect was partially mediated by NK1.1+cells.These results reveal a new critical role for the cDC1-iNKT cell axis in the regulation of adipose tissue homeostasis.

Seismic hazard analysis for central-western Argentina

In this study, we present a PSHA（Probabilistic Seismic Hazard Analysis） for the city of San Juan, which is located in the central-western region of Argentina（30°S-35.5°S; 66.5°W-71°W）. In addition to crustal earthquakes provided by catalogues, recent paleoseismological and neotectonic investigations have permitted to consider events which occurred during the last 400 years.Four seismogenic sources that could cause damages to the studied site corresponding to Precordillera,Western Sierras Pampeanas, Basement of the Cuyana Basin and Cordillera Principal were identified.Based on the evaluation of the contribution of these sources,maximum moment magnitudes above 7.5（Mw） are expected.High values of SA（spectral acceleration）（0.2 and 1 s periods） and PGA（peak ground acceleration） were found in the city of San Juan, which suggests that it is located in a zone of high seismic hazard.Finally, the obtained SA spectra were compared with the seismic-resistant construction standards of Argentina INPRES-CIRSOC 103 [1]. Results suggest that for the city of San Juan and for a return period of475 years, it covers the seismic requirements of the structures.

Subpicosecond pulses in a self-starting mode-locked semiconductor-based figure-of-eight fiber laser

We have experimentally studied the mode-locking dynamics of a polarization-maintaining figure-of-eight laser which has a semiconductor optical amplifier as gain medium. Self-starting mode-locking at the fundamental repetition rate of 18 MHz is obtained at the lasing threshold, and further increasing the bias current leads to the progressive emission of additional optical pulses in each round trip and eventually to mode-locked emission at increasingly high harmonics of the fundamental repetition rate, up to 2.45 GHz. The intensity autocorrelation of the amplified mode-locked pulses has a full width at half-maximum duration of 382 fs, which corresponds to a pulse duration of 247 fs.

Kinetic and morphology study of alginate-vineyard pruning waste biocomposite vs. non modified vineyard pruning waste for dye removal

In this work a comparative bioadsorption study between a biocomposite consisting of hydrolysed vineyard pruning waste entrapped in calcium alginate spheres and non entrapped vineyard residue was carried out.Results have demonstrated that the biocomposite based on lignocellulose-calcium alginate spheres removed 77.3% of dyes,while non entrapped lignocellulose eliminated only removed 27.8% of colour compounds.The experimental data were fitted to several kinetic models（pseudo-first order,pseudo-second order,Chien-Clayton model,intraparticle diffusion model and Bangham model）; being pseudo-second order the kinetic model that better described the adsorption of dyes onto both bioadsorbents.In addition,a morphological study（roughness and shape） of alginate-vineyard biocomposite was established under extreme conditions,observing significant differences between hydrated and dehydrated alginate-vineyard biocomposite.The techniques used to carry out this morphological study consisted of scanning electron microscopy（SEM）,perfilometry and 3D surface analysis.

Querétaro-Celaya highway is the obligatory road to the center of Mexico;the establishment of new companies in the area has generated the need to study the dynamics of the vehicular flow that transits this route.Given that the flow of traffic on a highway is a stochastic phenomenon,it is necessary to apply tools that take into account the randomness of the system to measure performance.Queueing theory models capture the random nature of the phenomenon and provide direct information about the relationship between the variables of the system.In this work,the flow of vehicles on the Querétaro-Celaya highway is analyzed with a macroscopic approach and using analytical models of queueing theory;it is concluded that currently the flow on this road is non-congested.The method is an alternative for institutions that do not have specialized packages to carry out studies of this nature.

Objective Although robotic surgery adoption and its indications are growing worldwide,for multiple factors,including costs,there is a lack of training and experience.Our aim was to study the impact of a robotic introduction training program on gesture performance,such as suturing,in robot-naive individuals.Methods Using the DaVinci robot,a 2-hour program was based on virtual reality and anatomical model exercises.All participants performed 3 repetitions of virtual reality exercises on the virtual simulator,and then performed and were assessed on 2 tests,ie robot and laparoscopic training box.After the course,the participants were surveyed for this training program.Results Twenty-seven residents and surgeons were enrolled in the training program.With only 2 hours of training,all of the participants were able to complete the training program,thus learning generic and specific skills in robotic surgery.In virtual reality exercise,the scores of the 3 exercises increased significantly with every repetition(p<0.001)and the size of the increase was large.The completion time on the robot platform was 2.6 times faster(169.33±28.28 s vs.447.96±156.55 s,p<0.001)than that in the laparoscopic box,and the difference between both types of tests was large(pη2=0.797).The centralization and passage of the needle were significantly better on the robot platform(5 vs.3,p<0.001,r=0.47;5 vs.4,p<0.001,r=0.59)than in the laparoscopic box.For the intracorporeal stitch+knot test,every participant was able to perform the exercise on the robot but only 85.2%(23/27)in the laparoscopic box.Twenty-one participants answered the survey,and 13(61.9%)of them considered robotic performance independent of laparoscopic experience.Conclusions Surgeons are interested and seek training in robotic surgery.We implemented the first hands-on robotic surgery training program in Portugal and participants considered it was important and adequate for its purpose.All participants,even without robotic experience,learned quicker,performed better,faster and more pre

The connection of resources, data, and knowledge through communication technology plays a vital role in current collaborative design methodologies and Product Lifecycle Management （PLM） systems, as these elements act as channels for information and meaning. Despite significant advances in the area of PLM, most communication tools are used as separate services that are disconnected from existing development environments. Consequently, during a communication session, the specific elements being discussed are usually not linked to the context of the discussion, which may result in important information getting lost or becoming difficult to access. In this paper, we present a method to add synchronous communication functionality to a PLM system based on annotated information embedded in the CAD model. This approach provides users a communication channel that is built directly into the CAD interface and is valuable when individuals need to be contacted regarding the annotated aspects of a CAD model. We present the architecture of a new system and its integration with existing PLM systems, and describe the implementation details of an annotation-based video conferencing module for a commercial CAD application.

Lithic soils in the semi-arid region of Brazil:edaphic characterization and susceptibility to erosion

Soils(Leptosols or Epileptic Regosols)with lithic contact at a depth of 50 cm occupy almost 20%of the Brazilian semi-arid region.These lithic soils are susceptible to erosion due to faster saturation of water-holding capacity during rainfall,which accelerates the beginning of runoff.However,erosion traits of lithic soils in the semi-arid region of Brazil are less studied.The aim of this study was to characterize the soil and landscape attributes in areas with Neossolos Litólicos(Entisols)in the Caatinga biome to identify region of high susceptibility to erosion.Results showed that the soils were characterized by a sandy texture,soil structure with poor development and low content of organic carbon.These attributes increase susceptibility to erosion and reduce water storage capacity,especially in the states of Cearáand Sergipe.In these states,the content of rock fragments in the soil reaches 790 g/kg.High contents of silt and fine sand,high silt/clay ratio,predominance of Leptosols and strong rainfall erosivity were observed in Piauíand northwestern Ceará.A very high degree of water erosion was observed in the states of Pernambuco and Paraíba.Despite the low degree of erosion observed in the state of Bahia,it is highly susceptible to erosion due to the predominance of very shallow soils,rugged relief and high values of rainfall erosivity.Lower vulnerability was observed in the state of Alagoas because of its more smoothed relief,greater effective soil depth,thicker A horizon of soil and lower rainfall erosivity.In general,the characteristics that intensify the susceptibility to erosion in the Caatinga biome are those soil structures with poor development or without aggregation,low contents of organic carbon,high contents of silt and fine sand,high values of silt/clay ratio and rugged relief in some regions.This study collected information contributing to a better characterization of soils with lithic contact in the semi-arid region of Brazil.In addition,regions with a higher susceptibility to erosion were identifi
