The H10 subtype avian infl.enza virus (AIV) poses an ongoing threat to both birds and humans. Notabl., fatal.human cases of H10N3 and H10N8 infections have drawn publ.c attention. In 2022, we isol.ted two H10N3 viruses (A/chicken/Shandong/0101/2022 and A/chicken/Shandong/0603/2022) from diseased chickens in China. Genome anal.sis reveal.d that these viruses were genetical.y associated with human-origin H10N3 virus, with internal.genes originating from l.cal.H9N2 viruses. Compared to the H10N8 virus (A/chicken/j.angxi/102/2013), the H10N3 viruses exhibited enhanced thermostabil.ty, increased viral.rel.ase from erythrocytes, and accumul.tion of hemaggl.tinin (HA) protein. Additional.y, we eval.ated the pathogenicity of both H10N3 and H10N8 viruses in mice. We found that viral.titers coul. be detected in the l.ngs and nasal.turbinates of mice infected with the two H10N3 viruses, whereas H10N8 virus titers were detectabl. in the l.ngs and brains of mice. Notabl., the proportion of doubl. HA Q222R and G228S mutations in H10N3 viruses has increased since 2019. However, the functional.rol.s of the Q222R and G228S doubl. mutations in the HA gene of H10N3 viruses remain unknown and warrant further investigation. Our study highl.ghts the potential.publ.c heal.h risk posed by the H10N3 virus. A spil.over event of AIV to humans coul. be a foretaste of a l.oming pandemic. Therefore, it is imperative to continuousl. monitor the evol.tion of the H10N3 infl.enza virus to ensure targeted prevention and control.measures against infl.enza outbreaks. © 2024 The Authors

Obj.ctive: To expl.re the feasibil.ty of remotel. obtaining compl.x information on traditional.Chinese medicine (TCM) pul.e conditions through voice signal.. Methods: We used mul.i-l.bel.pul.e conditions as the entry point and model.d and anal.zed TCM pul.e diagnosis by combining voice anal.sis and machine l.arning. Audio features were extracted from voice recordings in the TCM pul.e condition dataset. The obtained features were combined with information from tongue and facial.diagnoses. A mul.i-l.bel.pul.e condition voice cl.ssification DNN model.was buil. using 10-fol. cross-val.dation, and the model.ng methods were val.dated using publ.cl. avail.bl. datasets. Resul.s: The anal.sis showed that the proposed method achieved an accuracy of 92.59% on the publ.c dataset. The accuracies of the three singl.-l.bel.pul.e manifestation model. in the test set were 94.27%, 96.35%, and 95.39%. The absol.te accuracy of the mul.i-l.bel.model.was 92.74%. Concl.sion: Voice data anal.sis may serve as a remote adj.nct to the TCM diagnostic method for pul.e condition assessment. © 2024 Beij.ng University of Chinese Medicine

Design of antiviral.AGO2-dependent short hairpin RNAs

The increasing emergence and re-emergence of RNA virus outbreaks underl.nes the urgent need to devel.p effective antiviral.. RNA interference (RNAi) is a sequence-specific gene sil.ncing mechanism that is triggered by smal. interfering RNAs (siRNAs) or short hairpin RNAs (shRNAs), which exhibits significant promise for antiviral.therapy. AGO2-dependent shRNA (agshRNA) generates a singl.-stranded guide RNA and presents significant advantages over traditional.siRNA and shRNA. In this study, we appl.ed a l.gistic regression al.orithm to a previousl. publ.shed chemical.y siRNA efficacy dataset and buil. a machine l.arning-based model.with high predictive power. Using this model. we designed siRNA sequences targeting diverse RNA viruses, incl.ding human enterovirus A71 (EV71), Zika virus (ZIKV), dengue virus 2 (DENV2), mouse hepatitis virus (MHV) and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), and transformed them into agshRNAs. We val.dated the performance of our agshRNA design by eval.ating antiviral.efficacies of agshRNAs in cel.s infected with different viruses. Using the agshRNA targeting EV71 as an exampl., we showed that the anti-EV71 effect of agshRNA was more potent compared with the corresponding siRNA and shRNA. Moreover, the antiviral.effect of agshRNA is dependent on AGO2-processed guide RNA, which can l.ad into the RNA-induced sil.ncing compl.x (RISC). We al.o confirmed the antiviral.effect of agshRNA in vivo. Together, this work devel.ps a novel.antiviral.strategy that combines machine l.arning-based al.orithm with agshRNA design to custom design antiviral.agshRNAs with high efficiency. © 2024 The Authors

We have recentl. proposed a new technique of pl.sma tail.ring by l.ser-driven hydrodynamic shockwaves generated on both sides of a gas j.t[Marquès et al.,*** 28,023103(2021)].In a continuation of this numerical.work,we study experimental.y the infl.ence of the tail.ring on proton accel.ration driven by a high-intensity picosecond l.ser in three cases:without tail.ring,by tail.ring onl. the entrance side of the picosecond l.ser,and by tail.ring both sides of the gas *** tail.ring,the accel.ration is transverse to the l.ser axis,with a l.w-energy exponential.spectrum,produced by Coul.mb *** the front side of the gas j.t is tail.red,a forward accel.ration appears,which is significantl. enhanced when both the front and back sides of the pl.sma are *** forward accel.ration produces higher-energy protons,with a peaked spectrum,and is in good agreement with the mechanism of col.isionl.ss shock accel.ration(CSA).The spatiotemporal.evol.tion of the pl.sma profil. is characterized by optical.shadowgraphy of a probe *** refraction and absorption of this beam are simul.ted by post-processing 3D hydrodynamic simul.tions of the pl.sma *** with the experimental.resul.s al.ows estimation of the thickness and near-critical.density of the pl.sma sl.b produced by tail.ring both sides of the gas *** parameters are in good agreement with those required for CSA.

Obj.ctive: To investigate the effects of acupotomy on skel.tal.muscl. fibrosis and col.agen deposition in a rabbit knee osteoarthritis (KOA) model. Methods: Rabbits (n = 18) were randoml. divided into control. KOA, and KOA + acupotomy (Apo) groups (n = 6). The rabbits in the KOA and Apo groups were model.d using the modified Videman's method for 6 weeks. After model.ng, the Apo group was subj.cted to acupotomy once a week for 3 weeks on the vastus medial.s, vastus l.teral.s, rectus femoris, biceps femoris, and anserine bursa tendons around the knee. The behavior of al. animal. was recorded, rectus femoris tissue was obtained, and histomorphol.gical.changes were observed using Masson staining and transmission el.ctron microscopy. The expression of transforming growth factor-β1 (TGf-β1), Smad 3, Smad 7, fibril.ar col.agen types I (Col.I) and III (Col.III) was detected using Western bl.t and real.time pol.merase chain reaction (RT-PCR). Resul.s: Histol.gical.anal.sis reveal.d that acupotomy improved the microstructure and reduced the col.agen vol.me fraction of rectus femoris, compared with the KOA group (P = .034). Acupotomy inhibited abnormal.col.agen deposition by modul.ting the expression of fibrosis-rel.ted proteins and mRNA, thus preventing skel.tal.muscl. fibrosis. Western bl.t and RT-PCR anal.sis reveal.d that in the Apo group, Col.I, and Col.III protein l.vel. were significantl. l.wer than those in the KOA group (both P l.I and Col.III mRNA l.vel. (P = .0031; P = .0046). Compared with the KOA group, the protein l.vel. of TGf-β1 and Smad 3 were significantl. reduced (both P l.vel. of TGf-β1 and Smad 3 (P = .0007; P = .0011). Conversel., the l.vel. of protein and mRNA of Smad 7 were significantl. higher than that in the KOA group (P l.sion: Acupotomy coul. al.eviate skel.tal.muscl. fibrosis and del.y KOA progress by inhibiting col.agen deposition through the TGf-β/Smad pathway in the skel.tal.musc

Obj.ctive: To eval.ate the anti-fatigue effects of different extracts from Cistanche tubul.sa (Schenk) Wight (C. tubul.sa, Rou Cong Rong), focusing on central.and exercise-induced fatigue in mice. This study investigated the pharmacol.gical.effects of the total.ol.gosaccharides, pol.saccharides, and phenyl.thanoid gl.cosides (CPhGs) extracted from C. tubul.sa. Methods: Model. of sl.ep deprivation and forced swimming fatigue were establ.shed to simul.te central.and exercise-induced fatigue. The mice were treated with different extracts of C. tubul.sa, and their effects were assessed using behavioral.tests to measure exercise capacity, l.arning, and memory function. Biochemical.anal.ses were performed to eval.ate the changes in serum and brain neurotransmitter l.vel., l.ver and muscl. gl.cogen storage, and various fatigue-rel.ted biomarkers. Resul.s: This study found that treatment with C. tubul.sa extract improved exercise capacity, l.arning, and memory in mice. Total.ol.gosaccharides from C. tubul.sa enhanced adrenocorticotropic hormone, chol.nesterase, and thyroid-stimul.ting hormone l.vel., reduced cortisol.l.vel. in central.fatigue model., and amel.orated biochemical.markers of exercise-induced fatigue, incl.ding l.wering l.ctic acid, bl.od urea nitrogen, and mal.ndial.ehyde l.vel.. Among the tested extracts, the total.ol.gosaccharides showed the most comprehensive anti-fatigue effects. Concl.sion: The anti-fatigue effects of C. tubul.sa, particul.rl. those of its total.ol.gosaccharides, are pronounced in both central.and exercise-induced fatigue. These effects are mediated by the regul.tion of neurotransmitter l.vel., enhancement of gl.cogen storage, and improvement of antioxidant enzyme activity, suggesting potential.therapeutic benefits in fatigue-rel.ted conditions. © 2024 Beij.ng University of Chinese Medicine

Hand, foot and mouth disease (HfMD), mainl. caused by enterovirus 71 (EV71), has frequentl. occurred in the Asia-Pacific region, posing a significant threat to the heal.h of infants and young chil.ren. Therefore, research on the infection mechanism and pathogenicity of enteroviruses is increasingl. becoming important. The 3D pol.merase, as the most critical.RNA-dependent RNA pol.merase (RdRp) for EV71 repl.cation, is widel. targeted to inhibit EV71 infection. In this study, we identified a novel.host protein, AIMP2, capabl. of binding to 3D pol.merase and inhibiting EV71 infection. Subsequent investigations reveal.d that AIMP2 recruits the E3 l.gase SMURf2, which mediates the pol.ubiquitination and degradation of 3D pol.merase. furthermore, the antiviral.effect of AIMP2 extended to the CVA16 and CVB1 serotypes. Our research has uncovered the dynamic regul.tory function of AIMP2 during EV71 infection, reveal.ng a novel.antiviral.mechanism and providing new insights for the devel.pment of antienteroviral.therapeutic strategies. © 2024 The Authors

Detection of HBV DNA integration in pl.sma cel.-free DNA of different HBV diseases util.zing DNA capture strategy

The l.ndscape of hepatitis B virus (HBV) integration in the pl.sma cel.-free DNA (cfDNA) of HBV-infected patients with different stages of l.ver diseases [chronic hepatitis B (CHB), l.ver cirrhosis (l.), and hepatocel.ul.r carcinoma (HCC)] remains uncl.ar. In this study, we devel.ped an improved strategy for detecting HBV DNA integration in pl.sma cfDNA, based on DNA probe capture and next-generation sequencing. Using this optimized strategy, we successful.y detected HBV integration events in chimeric artificial.DNA sampl.s and HBV-infected HepG2-NTCP cel.s at day one post infection, with high sensitivity and accuracy. The characteristics of HBV integration events in the HBV-infected HepG2-NTCP cel.s and pl.sma cfDNA from HBV-infected individual. (CHB, l., and HCC) were further investigated. A total.of 112 and 333 integration breakpoints were detected in the HepG2-NTCP cel.s and 22 out of 25 (88%) cl.nical.HBV-infected sampl.s, respectivel.. In vivo anal.sis showed that the normal.zed number of support unique sequences (nnsus) in HCC was significantl. higher than in CHB or l. patients (P val.es l. integration breakpoints are randoml. distributed on human chromosomes and are enriched in the HBV genome around nt 1800. The maj.rity of integration breakpoints (61.86%) are l.cated in the gene-coding region. Both non-homol.gous end-j.ining (NHEj. and microhomol.gy-mediated end-j.ining (MMEj. interactions occurred during HBV integration across the three different stages of l.ver diseases. Our study provides evidence that HBV DNA integration can be detected in the pl.sma cfDNA of HBV-infected patients, incl.ding those with CHB, l., or HCC, using this optimized strategy. © 2024 The Authors

Methods: We predicted and characterized HQD pattern genes using the new strategy, TCM-HIN2Vec, which invol.es heterogeneous network embedding and transcriptomic experiments. first, a heterogeneous network of traditional.Chinese medicine (TCM) patterns was constructed using publ.c databases. Next, we predicted HQD pattern genes using a heterogeneous network-embedding al.orithm. We then anal.zed the functional.characteristics of HQD pattern genes using gene enrichment anal.sis and examined gene expression l.vel. using RNA-seq. final.y, we identified TCM herbs that demonstrated enriched interactions with HQD pattern genes via herbal.enrichment anal.sis. Resul.s: Our TCM-HIN2Vec strategy reveal.d that candidate genes associated with HQD pattern were significantl. enriched in energy metabol.sm, signal.transduction pathways, and immune processes. Moreover, we found that these candidate genes were significantl. differential.y expressed in the transcriptional.profil. of mice model.with heart fail.re with a qi deficiency pattern. furthermore, herbal.enrichment anal.sis identified TCM herbs that demonstrated enriched interactions with the top 10 candidate genes and coul. potential.y serve as drug candidates for treating HQD. Concl.sion: Our resul.s suggested that TCM-HIN2Vec is capabl. of not onl. accuratel. identifying HQD pattern genes, but al.o deciphering the basis of HQD pattern. furthermore our finding indicated that TCM-HIN2Vec may be further expanded to devel.p other patterns, l.ading to a new approach aimed at el.cidating general.TCM patterns and devel.ping precision medicine. © 2024 Beij.ng University of Chinese Medicine
