Inhibitory neurotransmission ensures normal brain function by counteracting and integrating excitatory activity.-Aminobutyric acid (GABA) is the main inhibitory neurotransmitter in the mammalian central nervous system,and mediates its effects via two classes of receptors:the GABA A and GABA B *** A receptors are heteropentameric GABA-gated chloride channels and responsible for fast inhibitory *** B receptors are heterodimeric G protein coupled receptors (GPCR) that mediate slow and prolonged inhibitory *** extent of inhibitory neurotransmission is determined by a variety of factors,such as the degree of transmitter release and changes in receptor activity by posttranslational modifications (e.g.,phosphorylation),as well as by the number of receptors present in the plasma membrane available for signal *** level of GABA B receptors at the cell surface critically depends on the residence time at the cell surface and finally the rates of endocytosis and *** this review we focus primarily on recent advances in the understanding of trafficking mechanisms that determine the expression level of GABA B receptors in the plasma membrane,and thereby signaling strength.

Natural products have been extensively used to treat diseases throughout human history. These are mainly because natural products normally target biological macromolecules selectively. Target identification could help us to develop new therapeutic agents and discover new biological pathways underlying human diseases. Herein, we highlight some recent examples of using natural products and their derivatives as chemical probes to identify the molecular targets and elucidate mode of action.

The paper presents a multi-scale modelling approach for simulating macromolecules in fluid flows. Macromolecule transport at low number densities is frequently encountered in biomedical devices, such as separators, detection and analysis systems. Accurate modelling of this process is challenging due to the wide range of physical scales involved. The continuum approach is not valid for low solute concentrations, but the large timescales of the fluid flow make purely molecular simulations prohibitively expensive. A promising multi-scale modelling strategy is provided by the meta-modelling approach considered in this paper. Meta-models are based on the coupled solution of fluid flow equations and equations of motion for a simplified mechanical model of macromolecules. The approach enables simulation of individual macromolecules at macroscopic time scales. Meta-models often rely on particle-corrector algorithms, which impose length constraints on the mechanical model. Lack of robustness of the particle-corrector algorithm employed can lead to slow convergence and numerical instability. A new FAst Linear COrrector （FALCO） algorithm is introduced in this paper, which significantly improves computational efficiency in comparison with the widely used SHAKE algorithm. Validation of the new particle corrector against a simple analytic solution is performed and improved convergence is demonstrated for ssDNA motion in a lid-driven micro-cavity.

The identification of design pattern instances is important for program understanding and software maintenance. Aiming at the mining of design patterns in existing systems, this paper proposes a subgraph isomorphism approach to discover several design patterns in a legacy system at a time. The attributed relational graph is used to describe design patterns and legacy systems. The sub-graph isomorphism approach consists of decomposition and composition process. During the decomposition process, graphs corresponding to the design patterns are decomposed into sub-graphs, some of which are graphs corresponding to the elemental design patterns. The composition process tries to get sub-graph isomorphism of the matched graph if sub-graph isomorphism of each subgraph is obtained. Due to the common structures between design patterns, the proposed approach can reduce the matching times of entities and relations. Compared with the existing methods, the proposed algorithm is not linearly dependent on the number of design pattern graphs. Large-scale chlorophyll （Chl） degradation, along with concomi- tant anthocyanin biosynthesis, creates the magnificent canopy scenery in the autumn. Dynamic Chl degradation greatly facili- tates plant growth and development by remobilizing nutrients from senescing organs to rapidly developing ones, seeds and storage organs in particular. In the last decade or so, the major Chl degradation pathway, termed the pheophorbide a oxygenase （PAO） pathway, has been extensively characterized by identifying Chl catabofic genes （CCGs） or enzymes （CCEs）. Chl a is the degradable form of Chls; while Chl b needs to be converted to Chl a before being channeled for degradation （Figure 1A）.

A numerical scheme for the Reissner-Mindlin plate model is *** method is based on a discrete Helmholtz decomposition and can be viewed as a generalization of the nonconforming finite element scheme of Arnold and Falk[SIAM ***.,26(6):1276-1290,1989].The two unknowns in the discrete formulation are the in-plane rotations and the gradient of the vertical *** decomposition of the discrete shear variable leads to equivalence with the usual Stokes system with penalty term plus two Poisson equations and the proposed method is equivalent to a stabilized discretization of the Stokes system that generalizes the Mini *** method is proved to satisfy a best-approximation result which is robust with respect to the thickness parameter t.
