The autotetraploid Carassius auratus(4nRR,4n=200,RRRR)is derived from whole-genome duplication of Carassius auratus red var.(RCC,2n=100,RR).In the current study,we demonstrated that chromatophores and pigment changes directly caused the coloration and variation of 4nRR skin(red in RCC,brownish-yellow in4nRR).To further explore the molecular mechanisms underlying coloration formation and variation in 4nRR,we performed transcriptome profiling and molecular functional verification in RCC and *** revealed that scarb1,associated with carotenoid metabolism,underwent significant down-regulation in *** editing of this candidate pigment gene provided clear evidence of its significant role in RCC ***,we identified four divergent scarb1 homeologs in 4nRR:two original scarb1 homeologs from RCC and two duplicated ***,three of these homeologs possessed two highly conserved alleles,exhibiting biased and allelespecific expression in the ***,after precise editing of both the original and duplicated scarb1homeologs and/or alleles,4nRR individuals,whether singly or multiply mutated,displayed a transition from brownishyellow skin to a cyan-gray ***,the proportional areas of the cyan-gray regions displayed a gene-dose *** findings illustrate the subfunctionalization of duplicated scarb1,with all scarb1genes synergistically and equally contributing to the pigmentation of *** is the first report concerning the functional differentiation of duplicated homeologs in an autopolyploidfish,substantiallyenrichingour understanding of coloration formation and change within this group of organisms.

Quantitative structure-activity relationship study on the biodegradation of acid dyestuffs

Quantitative structure-biodegradability relationships （QSBRs） were established to develop predictive models and mechanistic explanations for acid dyestuffs as well as biological activities. With a total of four descriptors, molecular weight （MW）, energies of the highest occupied molecular orbital （EHOMO）, the lowest unoccupied molecular orbital （ELUMO）, and the excited state （EES）, calculated using quantum chemical semi-empirical methodology, a series of models were analyzed between the dye biodegradability and each descriptor. Results showed that EHOMO and Mw were the dominant parameters controlling the biodegradability of acid dyes. A statistically robust QSBR model was developed for all studied dyes, with the combined application of EHOMO and Mw. The calculated biodegradations fitted well with the experimental data monitored in a facultative-aerobic process, indicative of the reliable prediction and mechanistic character of the developed model.

目的探讨粪便Syndecan-2(SDC2)甲基化与肠道菌群失调在结直肠癌(CRC)中的意义。方法将70例CRC患者(CRC组)、65例结直肠腺瘤性息肉(CAP)患者(CAP组)及50例健康志愿者(对照组)作为研究对象,检测其粪便DNA SDC2甲基化与肠道菌群失调情况,实时荧光定量PCR法定量分析粪便菌群,Spearman相关系数分析CRC患者粪便SDC2甲基化与其定量分析结果之间的相关性。结果实时荧光定量PCR法在分析粪便菌群中的稳定性及特异性良好,CRC组粪便SDC2甲基化阳性率及肠道菌群失调率均高于对照组与CAP组(P<0.05);与对照组相比,CRC组及CAP组有益菌定量分析水平降低,CRC组肠道菌群中有害菌水平上升(P<0.05)。CRC患者粪便SDC2甲基化与肠道乳酸杆菌、双歧杆菌定量水平呈负相关(P<0.05),与大肠杆菌、粪肠球菌以及脆弱拟杆菌定量分析水平呈正相关(P<0.05)。结论CRC患者存在明显的肠道菌群紊乱表现及高水平SDC2甲基化阳性率,肠道菌群可能通过影响SDC2甲基化参与CRC发生与发展。

The Gengma-Lancang-Menghai seismic belt,the southernmost part of the North-South seismic belt,is controlled by four active faults:the Nantinghe fault(NTHF),the Sanjianshan fault(SJSF),the Hanmuba-Lancang fault(HMB-LCF),and the Heihe fault(HHF),from northwest to ***,the tectonic activity of the faults in the Gengma-Lancang-Menghai seismic belt has not been fully studied *** the present work,we conducted tectonic geomorphic indices to analyze the relative tectonic activity along the faults in the seismic belt based on the digital elevation *** interpreted asymmetric factor,index of drainage basin shape,hypsometric integral,normalized streamlength gradient,valley floor-to-width to height ratio,and longitudinal profiles to show that the relative tectonic activity is higher in the north and lower in the south,and is strong in the region from Shanjia to Huimin in the center of the seismic belt.

Use of calcein and alizarin red S for immersion marking of black rockfish Sebastes schlegelii juveniles

Black rockfi sh Sebastes schlegelii juveniles(30–40 mm total length) were immersed in a range of calcein(CAL) solutions at concentrations ranging from 50 to 250 mg/L and alizarin red S(ARS) solutions at concentrations ranging from 100 to 500 mg/L in fi ltered seawater(salinity 30) for 24 h. Fluorescent marks were detected in otoliths(sagittae, asteriscus), scales, fi n rays(dorsal, pectoral, ventral, anal, and caudal fi n rays), and fi n spines(dorsal, ventral, and anal fi n spines) after a 60-d growth experiment. With the exception of 50–100 mg/L CAL, acceptable marks were produced in the otoliths and fi n spines by all concentrations of CAL and ARS. In particular, marks were clearly visible under normal light in the sagittae, asteriscus, and fi n spines of fi sh immersed in 200–500 mg/L, 300–500 mg/L, and 200–500 mg/L ARS, respectively. Scales and fi n rays had acceptable marks at much higher concentrations(≥50 mg/L CAL, ≥300 mg/L ARS for scales and ≥50 mg/L CAL, ≥200 mg/L ARS for fi n rays). The mark quality was highest(i.e., acceptable marks were observed in all sampled structures after immersion marking) in fi sh immersed in 150–250 mg/L CAL or 300–500 mg/L ARS. In addition, there was no signifi cant difference in survival and growth of marked fi sh compared with controls 60 d post-marking( P >0.05).

采用热裂解-气相色谱-质谱联用法对生物基降解塑料制品中生物基降解材料聚己二酸/对苯二甲酸丁二酯(PBAT)定性定量分析;选用二氯甲烷为生物降解塑料材料制品的溶剂,通过对生物降解材料PBAT的热裂解温度,气质试验参数,标准物质筛选,特征碎片的确定及多组分检测干扰因素排查,确定了PBAT裂解特征产物1,6-二氧杂环十二烷-7,12-二酮和苯甲酸,采用二者峰面积之和进行定量。在10~500 mg·L^(-1)内具有良好的线性关系,且在10、50、250 mg·L^(-1)3种添加水平下的回收率在90.7%~97.9%,相对标准偏差在10%以内,稳健度符合要求。
