The modification of amyloid fibrils cytotoxicity through exogenous nanomaterials is crucial to understand the processes controlling the role of protein aggregation in the related *** influence of nanoparticles on amyloid stability yields great interest due to the small size and high surface area-to-volume ratio of *** physico-chemical parameters play a role in the interaction of proteins and nanoparticles in solution,thus influencing the disaggregation of preformed *** have examined the influence of two kinds of metallic nanoparticles on lysozyme amyloid fibrils using a multi-technique approach and focalized their impact on cytotoxicity on human neuroblastoma cells(SH-SY5Y).In particular,fluorescence,infrared and circular dichroism spectroscopies,optical and atomic force microscopy experiments have been carried out;the results are analyzed to rationalize the effects of these complexes on neural cell *** is remarkable,that the fibrils in the presence of AuNPs,unlike fibrils alone or with AgNPs,do not generate a significant cytotoxic effect even at high concentration and an amyloid degradation effect is visible.

Processing bioactive peptides from natural sources using electrodialysis with ultrafiltration membranes(EDUF)have gained attention since it can fractionate in terms of their charge and molecular *** is a pseudo-cereal highlighted by its high protein content,amino acid profile and adapting growing *** present work aimed at the production of quinoa peptides through fractionation using EDUF and to test the fractions according to antihypertensive and antidiabetic *** data showed the production of peptides ranging between 0.4 and 1.5 k ***(CQPF)(3.01%),anionic(AQPF)(1.18%)and the electrically neutral fraction quinoa protein hydrolysate(QPH)-EDUF(~95%)were ***-vitro studies showed the highest glucose uptake modulation in L6 cell skeletal myoblasts in presence of QPH-EDUF and AQPF(17%and 11%)indicating potential antidiabetic *** antihypertensive effect studied in-vivo in spontaneously hypertensive rats(SHR),showed a decrease in systolic blood pressure in presence of the fractionated peptides,being 100 mg/kg a dose comparable to Captopril(positive control).These results contribute to the current knowledge of bioactive peptides from quinoa by reporting the relevance of EDUF as tool to produce selected peptide ***,further characterization is needed towards peptide sequencing,their respective role in the metabolism and scaling-up production using EDUF.

Sexual behavior and male volatile compounds in wild and mass-reared strains of the Mexican fruit fly Anastrepha ludens （Diptera： Tephritidae） held under different colony management regimes

We compared the calling and mating behavior and volatile release of wild males Anastrepha ludens （Loew） with males from 4 mass-reared strains：（i） a stan- dard mass-reared colony （control）, （ii） a genetic sexing strain （Tap-7）, （iii） a colony started from males selected on their survival and mating competitiveness abilities （selected）, and （iv） a hybrid colony started by crossing wild males with control females. Selected and wild males were more competitive, achieving more matings under field cage conditions. Mass-reared strains showed higher percentages of pheromone calling males under field conditions except for Tap-7 males, which showed the highest percentages of pheromone calling males under laboratory cage conditions. For mature males of all strains, field-cage calling behavior increased during the last hour before sunset, with almost a 2 fold increase exhibited by wild males during the last half hour. The highest peak mating activity of the 4 mass-reared strains occurred 30 rain earlier than for wild males. By means of solid phase microextraction （SPME） plus gas chromatography-mass spectrometry （GC- MS）, the composition of volatiles released by males was analyzed and quantified, Wild males emitted significantly less amounts of （E,E）-α-farnesene but emitted significantly more amounts of （E,E）-suspensolide as they aged than mass-reared males. Within the 4 mass-reared strains, Tap-7 released significantly more amounts of （E,E）-α-farnesene and hybrid more of （E,E）-suspensolide. Differences in chemical composition could be ex- plained by the intrinsic characteristics of the strains and the colony management regimes. Characterization of calling behavior and age changes of volatile composition between wild and mass-reared strains could explain the differences in mating competitiveness and may be useful for optimizing the sterile insect technique in A. ludens.

Genetic predisposition to colorectal cancer:Where we stand and future perspectives

The development of colorectal cancer(CRC)can be influenced by genetic factors in both familial cases and sporadic *** CRC has been associated with genetic changes in high-,moderate-and low-penetrance susceptibility ***,despite the availability of current gene-identification techniques,the genetic causes of a considerable proportion of hereditary cases remain ***-wide association studies of CRC have identified a number of common lowpenetrance alleles associated with a slightly increased or decreased risk of *** accumulation of low-risk variants may partly explain the familial risk of CRC,and some of these variants may modify the risk of cancer in patients with mutations in high-penetrance *** the predisposition to develop CRC will require investigators to address the following challenges:the identification of genes that cause uncharacterized hereditary cases of CRC such as familial CRC type X and serrated polyposis;the classification of variants of unknown significance in known CRC-predisposing genes;and the identification of additional cancer risk modifiers that can be used to perform risk assessments for individual mutation *** performed a comprehensive review of the genetically characterized and uncharacterized hereditary CRC syndromes and of lowand moderate-penetrance loci and variants identified through genome-wide association studies and candidate-gene *** challenges and future perspectives in the field of CRC predisposition are also discussed.

Pancreatic ductal adenocarcinoma(PDAC)is a major cause of cancer-related death *** recent advances in treatment options,prognosis remains poor,and data estimates from the United States(US)show that PDAC will surpass colorectal cancer to become the second most common cause of cancer-related death by 2040(1).Surgery followed by adjuvant chemotherapy is the only potentially curative treatment option for patients with PDAC;however,the majority present with advanced stage,non-resectable *** diagnosis could improve the dismal prognosis for this patient group,but this remains challenging due to numerous factors including a lack of approved biomarkers and screening programmes,and non-specificity or late presentation of symptoms.

Polymorphisms and resistance mutations of hepatitis C virus on sequences in the European hepatitis C virus database

AIM To evaluate the occurrence of resistant mutations in treatment-na?ve hepatitis C virus(HCV) sequences deposited in the European hepatitis C virus database(euH CVdb). METHODS The sequences were downloaded from the eu HCVdb(https://***/eu HCVdb/). The search was performed for full-length NS3 protease, NS5 A and NS5 B polymerase sequences of HCV, separated by genotypes 1a, 1b, 2a, 2b and 3a, and resulted in 798 NS3, 708 NS5 A and 535 NS5 B sequences from HCV genotypes1a, 1b, 2a, 2b and 3a, after the exclusion of sequences containing errors and/or gaps or incomplete sequences, and sequences from patients previously treated with direct antiviral agents(DAA). The sequence alignment was performed with MEGA 6.06 MAC and the resulting protein sequences were then analyzed using the BioE dit 7.2.5. for mutations associated with resistance. Only positions that have been described as being associated with failure in treatment in in vivo studies, and/or as conferring a more than 2-fold change in replication in comparison to the wildtype reference strain in in vitro phenotypic assays were included in the *** The Q80 K variant in the NS3 gene was the most prevalent mutation, being found in 44.66% of subtype 1a and 0.25% of subtype 1b. Other frequent mutations observed in more than 2% of the NS3 sequences were: I170V(3.21%) in genotype 1a, and Y56F(15.93%), V132I(23.28%) and I170V(65.20%) in genotype 1b. For the NS5 A, 2.21% of the genotype 1a sequences have the P58 S mutation, 5.95% of genotype 1b sequences have the R30 Q mutation, 15.79% of subtypes 2a sequences have the Q30 R mutation, 23.08% of subtype 2b sequences have a L31 M mutation, and in subtype 3a sequences, 23.08% have the M31 L resistant variants. For the NS5 B, the V321 L RAV was identified in 0.60% of genotype 1a and in 0.32% of genotype 1b sequences, and the N142 T variant was observed in 0.32% of subtype 1b sequences. Experimental Assessment of the Battery Lifetime in WSN Based on the Duty-CycleCurrentAverageMethod

A great amount of work addressed methods for predicting the battery lifetime in wireless sensor systems. In spite of these efforts, the reported experimental results demonstrate that the duty-cycle current average method, which is widely used to this aim, fails in accurately estimating the battery life time of most of the presented wireless sensor system applications. The aim of this paper is to experimentally assess the duty-cycle current average method in order to give more effective insight on the effectiveness of the method. An electronic metering system, based on a dedicated PCB, has been designed and developed to experimentally measure node current consumption profiles and charge extracted from the battery in two selected case studies. A battery lifetime measurement (during 30 days) has been carried out. Experimental results have been assessed and compared with estimations given by using the duty-cycle current average method. Based on the measurement results, we show that the assumptions on which the method is based do not hold in real operating cases. The rationality of the duty-cycle current average method needs reconsidering.
