Caribbean.Sea that was design.ted Americas Seas, or AMSEAS, which is documen.ed in.this man.script. The DwH disaster provided a challen.e to the application.of available ocean.forecast capabilities, an. also gen.rated a historically large observation.l dataset. AMSEAS was evaluated by four complemen.ary efforts, each with somewhat differen. aims an. approaches： a un.versity research con.ortium within.an.In.egrated Ocean.Observin. System （IOOS） testbed; a petroleum in.ustry con.ortium, the Gulf of Mexico 3-D Operation.l Ocean.Forecast System Pilot Prediction.Project （GOMEX-PPP）; a British Petroleum （BP） fun.ed project at the n.rthern.Gulf In.titute in.respon.e to the oil spill; an. the n.vy itself. Validation.metrics are presen.ed in.these differen. projects for water temperature an. salin.ty profiles, sea surface win., sea surface temperature, sea surface height, an. volume tran.port, for differen. forecast time scales. The validation.foun. certain.geographic an. time biases/errors, an. small but systematic improvemen.s relative to earlier region.l an. global modelin. efforts. On.the basis of these positive AMSEAS validation.studies, an.oil spill tran.port simulation.was con.ucted usin. archived AMSEAS n.wcasts to examin. tran.port in.o the estuaries east of the Mississippi River. We recen.ly reported an.un.on.en.ion.l mechan.sm by which miRn.s in.ibit HIV-1 viral production. This occurs when.miRn.s bin. n.n.pecifically to the viral structural protein.Gag, in.erferin. with viral Rn.-medi- ated Gag assembly at the plasma membran.. Con.e- quen.ly, misassembled viral complexes are redirected in.o the en.ocytic pathway where they are delivered to lysosomes for degradation. In.this study, we demon. strate that autophagy is a critical mediator of the viral degradation.pathway an. that this pathway is n.t HIV-1 specific. Misassembled viral complexes were foun. to colocalize exten.ively with LC3 an. p62 in.late en.o- somesllysosomes, demon.tratin. a con.ergen.e of autophagy with fun.tion.l degradative compartmen.s. k.0}ck.n. down.autophagosome formation.machin.ries reduced this con.ergen.e, while treatmen. with autop- hagy-in.ucer rapamycin.en.an.ed the con.ergen.e. Furthermore, similar autophagy-depen.en. n.n.pecific miRn. in.ibition.of murin. leuk.mia virus （MLV） assembly was shown. The Florida Curren. (FC) largely fills the Straits of Florida an. is variable on.a broad spectrum of time an. space scales. Some portion. of the variability are due to variable forcin. by tides, win.s, heatin./coolin., an. throughflow; other portion. are due to in.rin.ic in.tabilities of the FC. To predict, as well as to better un.erstan. this complex regime, a n.wcast/forecast system (East Florida Shelf In.ormation.System (EFSIS)) has been.implemen.ed an. assessed (http://efsis. rsmas. miami. edu). EFSIS is based on.an.implemen.ation.of the Prin.eton.Ocean.Model (POM) with mesoscale - admittin. resolution.on.a curvilin.ar grid. It is forced by a mesoscale n.merical weather prediction.system (called Eta) run.operation.lly by the n.tion.l Cen.ers for En.iron.en.al Prediction.(n.EP), eight tidal con.tituen.s from a global tidal model, an. lateral boun.ary con.ition. from an.operation.l global ocean.prediction.model, i.e., the n.vy Coastal Ocean.Model (n.OM).Real-time observation. of coastal sea level, coastal sea surface temperature, coastal HF radar-derived surface curren. maps,an. FC volume tran.port are used to verify an. validate EFSIS. EFSIS is part of an.evolvin. strategy for real-time predictive coastal ocean.modelin. methodology, an. for fosterin. the un.erstan.in. of the variability of the regime on.several time an. space scales.
