Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)-induced cytokine storms constitute the primary cause of coronavirus disease 19(COVID-19)progression,severity,criticality,and *** and anti-cytokine therapies are frequently administered to treat COVID-19,but have limited clinical efficacy in severe and critical ***,the weaknesses of these treatment modalities have prompted the development of anti-inflammatory therapy against this *** found that the broad-spectrum anti-inflammatory agent inosine downregulated proinflammatory interleukin(IL)-6,upregulated anti-inflammatory IL-10,and ameliorated acute inflammatory lung injury caused by multiple infectious *** significantly improved survival in mice infected with *** indirectly impeded TANK-binding kinase 1(TBK1)phosphorylation by binding stimulator of interferon genes(STING)and glycogen synthase kinase-3β(GSK3β),inhibited the activation and nuclear translocation of the downstream transcription factors interferon regulatory factor(IRF3)and nuclear factor kappa B(NF-κB),and downregulated IL-6 in the sera and lung tissues of mice infected with lipopolysaccharide(LPS),H1N1,or ***,inosine administration is feasible for clinical anti-inflammatory therapy against severe and critical ***,targeting TBK1 is a promising strategy for inhibiting cytokine storms and mitigating acute inflammatory lung injury induced by SARS-CoV-2 and other infectious agents.

Respiratory syncytial virus(RSV)infection is the major cause of severe lower respiratory illness in infants and young children,but the underlying mechanisms responsible for the pathogenesis are not fully *** date,no drugs or vaccines have been employed to improve the clinical outcome of RSV-infected *** this study,we report that angiotensin-converting enzyme-2(ACE2)protects against the severe lung injury induced by RSV infection in an experimental mouse model and in pediatric ***,ACE2 deficiency aggravates the disease pathogenesis,mainly by targeting the angiotensin II type 1 receptor(AT1 R).Furthermore,recombinant ACE2 protein alleviates the severity of RSV-induced lung *** findings demonstrate that ACE2 plays a critical role in RSV-induced lung injury,and suggest that ACE2 is a promising potential therapeutic target for RSV-induced lung disease.

Hepatocellular carcinoma(HCC) is a leading cause of cancer mortality worldwide. Despite progress in the diagnosis and treatment of HCC, its prognosis still remains unfavorable. In accumulating studies, long non-coding RNAs(lncRNAs) are emerging as vital players in tumorigenesis and cancer progression. However, the molecular mechanisms and clinical significance of lncRNAs in HCC remain largely elusive. In this study, we analyzed lncRNA microarray expressionprofiles of tumor samples and its matched adjacent tissue samples from 10 HCC patients and found that the expression of RP11J20.1 in HCC tissues is makedly lower than adjacent tissues. Expression of RP11J20.1 was also remarkably downregulated in 112 HCC tissues and cell lines by qRT-PCR. Low RP11J20.1 expression was correlated with alphafetoprotein(AFP) level, tumor size and microvascular invasion. Kaplan-Meier survival analysis demonstrated that low RP11J20.1 expression predicts poor prognosis of HCC patients. Gain-of-function and loss-of-function experiments showed that RP11J20.1 promotes HCC cells proliferation, cell cycle progression, and inhibits HCC cells apoptosis. Further experiments revealed that RP11J20.1 promotes HCC cells proliferation through inhibiting STAT1 genes expression. Collectively, our results demonstrated the clinical prognostic significance and roles of RP11J20.1 in HCC, and suggested that RP11J20.1 may be considered as a prognostic biomarker and therapeutic target for HCC.

Influenza virus(IFV) infection is a major cause of severe respiratory illness that affects almost every country in the world. IFV infections result in respiratory illness and even acute lung injury and death, but the underlying mechanisms responsible for IFV pathogenesis remain to be fully elucidated. In this study, we found that the basic fibroblast growth factor 2(FGF2) level was markedly increased in H1N1 virus-infected humans and mice. FGF2, which is predominately derived from epithelial cells, recruits and activates neutrophils via the FGFR2-PI3K-AKT-NFκB signaling pathway. FGF2 depletion or knockoutexacerbated influenza-associated disease by impairing neutrophil recruitment and activation. More importantly, administration of recombinant FGF2 protein significantly alleviated the severity of IFV-induced lunginjury and promotedthe survival of IFV-infected animals. These findings demonstrate that FGF2 plays a critical role in preventing IFV-induced lung injury and suggest FGF2 as a promising potential therapeutic target during IFV infection.

Objective: Respiratory syncytial virus(RSV) is leading cause of lower respiratory infection in infants and children, but there is still no licensed vaccine availble. Virus-like partice(VLP)is a novel approach for RSV vaccine which form a virus-like structure but lack the viral ***: We designed VLP vaccines consist of influenza virus matrix(M1)protein and RSV fusion protein(F) or glycoprotein(G).These VLPs were develped based on the Bac-to-Bac baculovirus expression systerm and identified by western blot and electron microscoy. Finally,protective efficacy of the VLPs were evaluated using BALB/c ***:We successfully developed RSV-F and RSV-G VLPs. Morphology and sizes of VLPs were consistent with that of the wild-type *** BALB/c mice immunized intranaslly(i.n.) with RSV-F VLPs,RSV-G VLPs, or both showed viral-specific antibody responses against RSV. Total IgG, IgG1,IgG2 a,and mucosal IgA were detected in mice with RSV-F plus RSV-G VLPs, revealing potent cellular and mucosal immune responses. Moreover, we found that those mixed RSV VLPs conferred enhanced protection against live RSV challenges,showing significant decreases in lung viral replication and obvirous attenuation of histopathological changes associated with viral ***: RSV-F plus RSV-G VLPs by intransal vaccination as a promisingvaccine candidate, which warrants furhter evaluation using cotton rat and primate models.

An avian-origin influenza A(H7N9) virus has raised concern in China in the spring of 2013. Most H7N9 infections cases resulted in acute respiratory distress syndrome(ARDS), which is a severe form of acute lung injury(ALI) that further contributes to morbidity and mortality. In this study,we report a close association of CCL2 expression and acute lung injury induced by lethal H7N9 virus strain(A/Hebei/01/2013) in a mouse model. Elevated CCL2 levels were detected inconfirmed H7N9 patients and the bronchoalveolarlavage fluid(BALF) of H7N9 infected mice. Moreover, the overexpressions of CCL2 were observed in lung tissue of infected mice post infection. More importantly, CCL2 deficiency ameliorated acute lung injury induced by H7N9 virus in mice as determined by weight loss, survival rates, lung wet dry ratio and *** together, our findings demonstrated that CCL2 were essential for H7N9 virus infection andinterfering CCL2 might be promising as a potential therapeutic target for future influenza pandemic.
