Fire blight QTL analysis in a multi-family apple population identifies a reduced-susceptibility allele in ‘Honeycrisp’

Breeding apple cultivars with resistance offers a potential solution to fire blight,a damaging bacterial disease caused by Erwinia *** resistance alleles at quantitative trait loci(QTLs)were previously characterized in diverse Malus germplasm with poor fruit quality,which reduces breeding *** study utilized a pedigree-based QTL analysis approach to elucidate the genetic basis of resistance/susceptibility to fire blight from multiple genetic sources in germplasm relevant to *** breeding ***-seven important breeding parents(IBPs)were represented by 314 offspring from 32 full-sib families,with‘Honeycrisp’being the most highly represented *** resistance/susceptibility data from a two-year replicated field inoculation study and previously curated genome-wide single nucleotide polymorphism data,QTLs were consistently mapped on chromosomes(Chrs.)6,7,and *** QTLs together explained~28% of phenotypic *** Chr.6 and Chr.15 QTLs colocalized with previously reported QTLs,while the Chr.7 QTL is possibly novel.‘Honeycrisp’inherited a rare reduced-susceptibility allele at the Chr.6 QTL from its grandparent‘Frostbite’.The highly resistant IBP‘Enterprise’had at least one putative reduced-susceptibility allele at all three *** general,lower susceptibility was observed for individuals with higher numbers of reduced-susceptibility alleles across *** study highlighted QTL mapping and allele characterization of resistance/susceptibility to fire blight in complex pedigree-connected apple breeding *** gained will enable more informed parental selection and development of trait-predictive DNA tests for pyramiding favorable alleles and selection of superior apple cultivars with resistance to fire blight.

Characterization of Atomization Processes in Suspension/Emulsion Sprays

The characterization of suspension/emulsion sprays plays a decisive role in many industrial processes. A good example of such a process is the drying of a milk spray to produce milk powder, where the process efficiency and product quality is typically controlled by atomization parameters like flow rate, pressure, etc.. However, these parameters influence directly the droplet size and droplet velocity distributions in a spray, so that optimizing a spray drying process often involves adjusting the spray to a desired droplet size and droplet velocity distribution. This requires a measurement technique capable of characterizing in real time, the droplets in a suspension/emulsion spray. To achieve this aim, we present developments to the well-known time-shift technique for spray measurements.

Management and prevention of acute and chronic lateral ankle instability in athletic patient populations

Acute and chronic lateral ankle instability are common in high-demand patient populations. If not managed appropriately, patients may experience recurrent instability, chronic pain, osteochondral lesions of the talus, premature osteoarthritis, and other significantlong-term disability. Certain populations, including young athletes, military personnel and those involved in frequent running, jumping, and cutting motions, are at increased risk. Proposed risk factors include prior ankle sprain, elevated body weight or body mass index, female gender, neuromuscular deficits, postural imbalance, foot/ankle malalignment, and exposure to at-risk athletic activity. Prompt, accurate diagnosis is crucial, and evidence-based, functional rehabilitation regimens have a proven track record in returning active patients to work and sport. When patients fail to improve with physical therapy and external bracing, multiple surgical techniques have been described with reliable results, including both anatomic and nonanatomic reconstructive methods. Anatomic repair of the lateral ligamentous complex remains the gold standard for recurrent ankle instability, and it effectively restores native ankle anatomy and joint kinematics while preserving physiologic ankle and subtalar motion. Further preventative measures may minimize the risk of ankle instability in athletic cohorts, including prophylactic bracing and combined neuromuscular and proprioceptive training programs. These interventions have demonstrated benefit in patients at heightened risk for lateral ankle sprain and allow active cohorts to return to full activity without adversely affecting athletic performance.

Resection margin influences the outcome of patients with bilobar colorectal liver metastases

AIMTo evaluate the outcome of patients with bilobar colorectal liver metastases (CRLM) and identify clinico-pathological variables that influenced survival. METHODSPatients with bilobar CRLM were identified from a prospectively maintained hepatobiliary database during the study period (January 2010-June 2014). Collated data included demographics, primary tumour treatment, surgical data, histopathology analysis and clinical outcome. Down-staging therapy included Oxaliplatin- or Irinotecan- based regimens, and Cetuximab was also used in patients that were K-RAS wild-type. Response to neo-adjuvant therapy was assessed at the multi-disciplinary team meeting and considered for surgery if all macroscopic CRLM were resectable with a clear margin while preserving sufficient liver parenchyma. RESULTSOf the 136 patients included, thirty-two (23.5%) patients were considered inoperable and referred for palliative chemotherapy, and thirty-four (25%) patients underwent liver resection. Seventy (51.4%) patients underwent down-staging therapy, of which 37 (52.8%) patients responded sufficiently to undergo liver resection. Patients that failed to respond to down-staging therapy (n = 33, 47.1%) were referred for palliative therapy. There was a significant difference in overall survival between the three groups (surgery vs down-staging therapy vs inoperable disease, P P P = 0.017). On multi-variate analysis, R0 resection (P = 0.030) and female (P = 0.036) gender significantly influenced overall survival. CONCLUSIONPatients undergoing liver resection with bilobar CRLM have a significantly better survival outcome. R0 resection is associated with improved disease-free and overall survival in this patient group.

miR-200 family expression is downregulated upon neoplastic progression of Barrett's esophagus

AIM: To investigate miR-200 family expression in Barrett's epithelium, gastric and duodenal epithelia, and esophageal adenocarcinoma. METHODS: Real-time reverse transcriptase-polymerase chain reaction was used to measure miR-200, ZEB1 and ZEB2 expression. Ingenuity Pathway Analysis of miR-200 targets was used to predict biological outcomes. RESULTS: Barrett's epithelium expressed lower levels of miR-141 and miR-200c than did gastric and duodenal epithelia (P < 0.001). In silico analysis indicated roles for the miR-200 family in molecular pathways that distinguish Barrett's epithelium from gastric and duodenalepithelia, and which control apoptosis and proliferation. All miR-200 members were downregulated in adenocarcinoma (P < 0.02), and miR-200c expression was also downregulated in non-invasive epithelium adjacent to adenocarcinoma (P < 0.02). The expression of all miR-200 members was lower in Barrett's epithelium derived high-grade dysplastic cell lines than in a cell line derived from benign Barrett's epithelium. We observed signif icant inverse correlations between miR-200 family expression and ZEB1 and ZEB2 expression in Barrett's epithelium and esophageal adenocarcinoma (P < 0.05). CONCLUSION: miR-200 expression might contribute to the anti-apoptotic and proliferative phenotype of Barrett's epithelium and regulate key neoplastic processes in this epithelium.

A new species of Mountain Dragon(Reptilia:Agamidae:Diploderma) from the D. dymondi complex in southern Sichuan Province, China

Despite continuous studies on the cryptic diversity of the Diploderma flaviceps complex in Southwest China for the pastdecade, little attention has been given to other widespreadcongeners in China. Combining both morphological and phylogenetic data, we describe a new species of Diplodermafrom populations identified previously as D. dymondi in thelower Yalong River Basin in southern Sichuan Province. Thenew species is morphologically most similar to D. dymondiand D. varcoae, but it can be differentiated by a considerablegenetic divergence and a suite of morphological characters,including having taller nuchal crest scales, smaller tympana,and a distinct oral coloration. Additionally, we discuss otherputative species complexes within the genus Diploderma inChina.
