Mesuaferrea stem bark extract induces apoptosis and inhibits metastasis in human colorectal carcinoma HCT 116 cells, through modulation of multiple cell signalling pathways

Considering the great potential of natural products as anticancer agents, the present study was designed to explore the molecular mechanisms responsible for anticancer activities of Mesua ferrea stem bark extract against human colorectal carcinoma. Based on MTT assay results, bioactive sub-fraction(SF-3) was selected for further studies using HCT 116 cells. Repeated column chromatography resulted in isolation of less active α-amyrin from SF-3, which was identified and characterized by GC-MS and HPLC methods. α-amyrin and betulinic acid contents of SF-3 were measured by HPLC methods. Fluorescent assays revealed characteristic apoptotic features, including cell shrinkage, nuclear condensation, and marked decrease in mitochondrial membrane potential in SF-3 treated cells. In addition, increased levels of caspases-9 and-3/7 levels were also observed in SF-3 treated cells. SF-3 showed promising antimetastatic properties in multiple in vitro assays. Multi-pathway analysis revealed significant down-regulation of WNT, HIF-1α, and EGFR with simultaneous up-regulation of p53, Myc/Max, and TGF-β signalling pathways in SF-3 treated cells. In addition, promising growth inhibitory effects were observed in SF-3 treated HCT 116 tumour spheroids, which give a hint about in vivo antitumor efficacy of SF-3 phytoconstituents. In conclusion, these results demonstrated that anticancer effects of SF-3 towards colon cancer are through modulation of multiple molecular pathways.

Syzygium campanulatum Korth is a plant, which is a rich source of secondary metabolites （especially flavanones, chalcone, and triterpenoids）. In our present study, three conventional solvent extraction （CSE） techniques and supercritical fluid extraction （SFE） techniques were performed to achieve a maximum recovery of two flavanones, chalcone, and two triterpenoids from S. campanulatum leaves. Furthermore, a Box-Behnken design was constructed for the SFE technique using pressure, temperature, and particle size as independent variables, and yields of crude extract, individual and total secondary metabolites as the dependent variables. In the CSE procedure, twenty extracts were produced using ten different solvents and three techniques （maceration, soxhletion, and reflux）. An enriched extract of five secondary metabolites was collected using n-hexane：methanol （1：1） soxhletion. Using food-grade ethanol as a modifier, the SFE methods produced a higher recovery （25.5%-84.9%） of selected secondary metabo- lites as compared to the CSE techniques （0.92%-66.00%）.

Antioxidant and antiangiogenic activities of the essential oils of Myristica fragrans and Morinda citrifolia

Objective:Toinvestigate the anti-angiogenic activity and antioxidant properties of Myristica fragrans（***）（nutmeg） and Morinda citrifolia（***）（mengkudu） oils. Methods:The nutmeg and megkudu essential oils were obtained by steam distillation. The antioxidant activities of both essenlial oils were delermined by beta-carotene/ linoleic acid bleaching assay and reducing power while the anti-angiogenic activity was investigated using rat aortic ring assay using various ***:The results showed that nutmeg oil has higher antioxidant activity than mengkudu *** nutmeg oil effectively inhibited the oxidation of linoleic acid with（88.68±0.1）%while the inhibition percentage of oxidation of linoleic acid of the mengkudu oil is（69.44±0.4）%.The nutmeg oil and mengkudu oil showed reducing power with an EC50 value of 181.4μg/mL and 3 043.0μg/mL,*** anliangiogenic activity of nutmeg oil showed significant antiangiogenic activity with IC50 of 77.64μg/mL comparing to mengkudu oil which exhibits IC50 of 109.30μg/***:Bioactive compound（s） will be isolated from the nutmeg essential oil to be developed as antiangiogenic drugs.

Tomato(Solanum lycopersicum)is a perishable fruit because of its fast water loss and susceptibility to pathogens in the post-harvest stage,which leads to huge economic losses every *** this study,firstly from 19 tomato cultivars,we screened out two cultivars,Riogrand and SalarF1,having long and short shelf-life spans,***,shelf-life analysis was carried out for both cultivars at room *** exhibited that Riogrand showed higher firmness and less weight loss than *** ethylene production was higher in SalarF1,compared with Riogrand during post-harvest *** performed transcriptomic analysis of both cultivars in different storage *** discovered 2913,2188,and 11,119 differentially expressed genes(DEGs)for three post-harvest stages(0,20,and 40 Days Post-Harvest(DPH)),*** genes are enriched in ethylene biosynthesis and response,as well as cell wall-related *** response factor(ERF)ERF2 and ERF4 were highly expressed in SalarF1 with a short shelf life in 40 DPH,and the ethylene biosynthetic genes ACO1,ACO4,ACS6,and ACS2 were significantly upregulated in *** cell wall loosening and cell wall-related genes XTH3,XTH7,XTH23,1,3;1,4-β-D-Gluc-like,pGlcT1,Cellulase,PGH1,PL5,PL-like 1,PL-like 2 exhibited the highest levels of significance,being notably upregulated in the last stage of *** quantitative real-time polymerase chain reaction(qRT-PCR)analysis validated these gene expressions,which is in line with the transcriptome *** findings suggested that the extension of tomato fruit shelf life is mostly dependent on ethylene biosynthesis,signaling pathway genes,cell wall loosening,and cell wall-associated genes.

Response of Rice Cultivars to Elevated Air Temperature and Soil Amendments: Implications towards Climate Change Adaptations and Mitigating Global Warming Potentials

Global mean surface air temperature is expected to increase 1.1˚C - 6.4˚C by the end of 21st century which may affect rice productivity and methane emissions in the future climate. This experiment was conducted to investigate the response of rice cultivars to elevated air temperature (+1.5˚C higher than ambient) and soil amendments in regards to rice yield, yield scaled methane emissions and global warming potentials. The experimental findings revealed that replacement of inorganic fertilizers (20% - 40% of recommended NPKS) with Vermicompost, Azolla biofertilizer, enriched sugarcane pressmud, rice husk biochar and silicate fertilization increased rice yield 13.0% - 23.0%, and 11.0% - 19.0% during wet aman and dry boro season, respectively. However, seasonal cumulative CH4 fluxes were decreased by 9.0% - 25.0% and 5.0% - 19.0% during rainfed wet aman and irrigated dry boro rice cultivation, respectively with selected soil amendments. The maximum reduction in seasonal cumulative CH4 flux (19.0% - 25.0%) was recorded with silicate fertilization and azolla biofertilizer amendments (9.0% - 13.0%), whereas maximum grain yield increment 10.0 % - 14.0% was found with Vermicompost and Sugarcane pressmud amendments compared to chemical fertilization (100% NPKS) treated soils at ambient air temperature. However, rice grain yield decreased drastically 43.0% - 50.0% at elevated air temperature (3˚C higher than ambient air temperature), eventhough accelerated the total cumulative CH4 flux as well as GWPs in all treatments. Maximum seasonal mean GWPs were calculated at 391.0 kg CO2 eq·ha−1 in rice husk biochar followed by sugarcane pressmud (mean GWP 387.0 kg CO2 eq·ha−1), while least GWPs were calculated at 285 - 305 kg CO2 eq·ha−1 with silicate fertilizer and Azolla biofertilizer amendments. A consensus meeting of national experts from all major national hepatobiliary centres in the country was held on May 26,2023,at the Pakistan Kidney and Liver Institute&Research Centre(PKLI&RC)after initial consultations with the *** Pakistan Society for the Study of Liver Diseases(PSSLD)and PKLI&RC jointly organised this *** effort was based on a comprehensive literature review to establish national practice guidelines for hilar cholangiocarcinoma(hCCA).The consensus was that hCCA is a complex disease and requires a multidisciplinary team approach to best manage these *** coordinated effort can minimise delays and give patients a chance for curative treatment and effective *** diagnostic and staging workup includes high-quality computed tomography,magnetic resonance imaging,and magnetic resonance *** cytology or biopsy utilizing endoscopic retrograde cholangiopancreatography is a mainstay for ***,histopathologic confirmation is not always required before *** ultrasound with fine needle aspiration of regional lymph nodes and positron emission tomography scan are valuable adjuncts for *** only curative treatment is the surgical resection of the biliary tree based on the Bismuth-Corlette *** patients with unresectable hCCA can be considered for liver *** chemotherapy should be offered to patients with a high risk of *** use of preoperative biliary drainage and the need for portal vein embolisation should be based on local multidisciplinary *** with acute cholangitis can be drained with endoscopic or percutaneous biliary *** chemotherapy with cisplatin and gemcitabine has shown improved survival in patients with irresectable and recurrent hCCA.

Interactions of ciprofloxacin hydrochloride(CPFH)with sodium dodecyl sulfate(SDS)were investigated by conductivity measurement in H2O/electrolyte solutions(NaCl,Na2SO4&Na3PO4)over 298.15–318.15 K temperature range(with 5 K interval)considering the human body *** all cases,two critical micelle concentrations(c*)were observed which are increased in the presence of drug and decreased in the presence of salts enunciating the presence of interaction amongst the studied ***(CPFH+SDS)system in the presence of salt,the c*values at 303.15 K and I=0.50 mmol·kg^-1 followed the order:CNaCl>CNa2SO4>***ΔG1,m0 andΔG2,m0values are found to be negative for all systems that show that the micellization process is thermodynamically ***(CPFH+SDS)system in water,theΔHm0&ΔSm0 values reveal that the micellization processes is both entropy dominated in almost all *** the occurrence of electrolytes,ΔHm0 andΔSm0 values indicate that micellization processes are both entropy&enthalpy restricted at upper temperature but it becomes totally entropy dependent at higher *** higher positiveΔSm0 values indicate the enhanced hydrophobic interaction in presence of *** enthalpy-entropy compensation was determined from the linear relationship betweenΔHm0 andΔSm0 values in every *** transfer energies as well as compensation temperature and intrinsic enthalpy were also evaluated and the behaviors were comparable to other biological system.

Background: Ageing of the population is one of the most important demographic facts that come to the foreground in the 21st century. Objective: To assess the relationship between psychological factors (depression, loneliness, using anti anxiety medication, fear from falls and internal displacement of the population) and falls among elderly people in Baghdad city, Iraq. Methods: A matched 1:1 community based case-control study involving 716 elderly respondents, recruited randomly from six Non Governmental Organization (NGO) in different areas of Baghdad. Interviews to each respondent were done accordingly. Geriatric depression scale (GDS) was used to assess the depression among the elderly people. Results: The minimum age for cases and controls was 60 years old while the maximum age for cases was 87 and for controls was 85 years old. The female was predominant than male, 53.6% to 46.4% respectively. The relation between depression, fear from fall, using anti anxiety medication, internal displacement and falls was statistically significant

Thyroglossal Duct Anomalies in Pediatric and Adult Population: A 10-Year Experience in a Tertiary Care Hospital

Introduction: Thyroglossal duct cyst is a well-recognized congenital midline neck swelling observed in early childhood and rarely in adults. It may reveal itself as a painless cyst, abscess, or as fistula. Several studies have been conducted across the world, but the literature is scarce on its presentation and complications in the Middle East. This indexing study aims to report a 10-year experience with thyroglossal duct cyst (TGDC) presentation, excision, and recurrence at a teaching hospital in Saudi Arabia (KSA). Methods: A retrospective chart review was conducted at the Armed Forces Hospital Southern Region, KSA from December 2008 to December 2018. Data were retrieved from the electronic medical record system of the hospital and validated with the histopathology records. A total of 48 patients diagnosed as TGDC, sinus or fistula were identified. Stata ver. 16.1 was used to analyze the data and results formulated using regression model and Pearson’s chi-square test. Results: The majority of our patients were female (60.4%), and the most common presentation was a midline neck swelling which moved with tongue protrusion. Surgical excision via Complete Sistrunk procedure was the operation of choice in 60.5% followed by excision of the cyst with tract and sinus tract. Previous infection and surgical history contributed to complications in our patients, but no association of age or gender was observed. Patients presenting with infected and discharging cysts were found to be much more likely to develop a recurrence (9 patients). Cysts deep to the hyoid showed more recurrence [n = 7 (78%)] compared to superficial cysts [n = 2 (22%)] and posterior and multi-tract lesions also showed significant recurrence. Conclusion: Despite being a common cause of neck swelling, thyroglossal duct cyst is often missed on physical exams. To reduce complications, timely diagnosis and treatment are necessary.
