Objective:To investigate the effect of Benner Pury on Endothelial progenitor cells(EPCs)proliferation,migration,adhesion,apoptosis and oxidative stress in vitro from patients with hypertension and its time ***:The patients who were diagnosed as hypertension grade 1 according to the standard“Guidelines for Prevention and Treatment of Hypertension”were *** cells were isolated by density gradient centrifugation and stained with fluorescence chemical acetylated low density lipoprotein marked with DiI(acLDL-DiI)and fluorescein isothiocyanate(FITC)-*** staining positive cells were considered as the differentiation of *** control group included healthy subjects matched with study group in age,*** cultivated for 5 days were used for *** harvested at different set times under the stimulation of Benner Pury,and the EPCs proliferation,migration,adhesion ability,apoptosis and oxidative stress indicators were detected ***:(1)Compared with the control group,peripheral blood EPCs proliferation,migration and adhesion ability were ob-viously decreased in the hypertension group(p<.01),EPCs apoptosis rate and oxidative stress response were significantly increased(p<.01).(2)Benner Pury significantly increased the EPCs proliferation,migration,adhesion ability and improved apoptosis and oxidative stress reaction in a time-dependent ***:Benner Pury can improve the EPCs proliferation,migration,adhesion,apoptosis and oxidative stress in patients with hypertension in a time-dependent manner.

Background: Lysophosphatidic acid(LPA),one of the active lysophospholipid(LP), is an important extracellular signal molecule. As a bioactive lipid,LPA can regulates several cellular functions critical for proliferation, survival, cytoskeletal reorganization, migration, invasion and cytokine production via G-protein coupled receptors EDG family(LPA1, LPA2, and LPA3) and perhaps non-EDG family(LPA4–6).This experiment found that LPAR1 and LPAR3 was highly expressed in the tumor cells in the human ovarian cancer cell on the research of tumor proliferation and metastasis, while the LPA had obvious promotes effect on the proliferation and metastasis of ovarian cancer cells. Therefore, LPA has been suggested as a biomarker for ovarian cancer. It is worthwhile to conduct deep exploration on whether the LPA can be developed into a drug to treat the ovarian cancer. Methods: Culture human ovarian cancer cells. Real Time PCR detect the LPAR expression in the human ovarian cancer cell. Detect the proliferation by MTT method. At the same time using Micro chemotaxis chamber method to test the effects of LPA on cell migration. Then interference LPAR1/3 detected A2780/HO-8910 cell proliferation and migration. Protein expression was detected by Western Blot. Results: The Real Time PCR result discovered that LPAR1/LPAR3 is higher than other receptors expression in the human ovarian cancer cell A2780/HO-8910. LPA can stimulate A2780/HO-8910 cells to proliferation and found a dose-dependent. At the same time we found that LPA can induce cell migration.. The Western Blot results display that LPA stimulate A2780 cell proliferation and migration induced mainly Gi proteins play a role in the process, and activate the downstream MAKA signaling pathways. Conclusions: LPA induces migration and proliferation of ovarian cancer cells A2780/HO-8910 through LPAR1/LPAR3/Gi//P38 and LPAR1/LPAR3/Gi/ERK respectively.

The migration and proliferation of vascular smooth muscle cells(SMCs) is a hallmark of the pathogenesis of atherosclerosis. We previously showed that Plasma low-density lipoprotein(LDL) induced the migration of human coronary artery SMCs(CASMCs). Among bioactive lipids postulated to be present in LDL, lysophosphatidic acid(LPA),amount of which is higher than others, mimicked the LDL action. In this work, we tested the effects of LPA on human artery smooth muscle cells(HASMCs)and found that LPA induced migrat ion and proliferat ion of HASMCs. This effect was markedly inhibited by Ki-16425, an antagonist of LPA1/3 receptor, and a small interfering RNA(si RNA) targeted for LPAR1 receptors. LPA can also induce extracellular signal-regulated kinase(ERK1/2) activation. Both of si RNA targeting LPAR1 receptor and PTX,an inhibitor of Gi protein,inhibit ed LPA induced ERK activat ion and HASMCs proliferat ion. Due to these results, we suppose that LPA can promote proliferation and migrat ion of SMCs and final induced the atherosclerosis through LPAR1. To confirm this hypothesis,the atherosclerosis disease animal model was studied. As the experimental group, ten rabbits were feed with high-lipid forage for 70 days and the another ten were feed with normal forage for a contrast. The atherosclerotic plaques were occurred in aorta of the experimental group of which atherosclerosis index(AI) was higher than control group. Our data indicate that the lpr1 gene expression was obviously increased in atherosclerotic plaque. Taken together, these data indicate that the G protein coupled receptor LPAR1 is an atherosclerosis related receptor.
