Epizootic hemorrhagic disease virus(EHDV)is a member of the genus Orbivirus,family Sedoreoviridae.It was firstly recognized in 1955 to cause a highly fatal disease of wild white-tailed deer in America.So far,EHDV was detected and isolated in many wild or domestic ruminants,and widely distributed all over the world.Although the domestic cattle and sheep infected by EHDV were usually asymptomatic or subclinical,several outbreaks of epizootic hemorrhagic disease(EHD)in deer and cattle had been reported.Many EHDV strains were isolated and sequenced in last two decades in China,which promoted a general serologic investigation of EHDV in China.In this study,18,122 sera were collected from asymptomatic or subclinical domestic ruminants(cattle,cow,yaks,sheep,goats,and deer)in 116 regions belonging to 15 provinces in China.All the sera were tested by EHDV CELISA,and the results were obtained by big data analysis.EHDV infections were detected in the 14 of 15provinces,and only Tibet(average altitude≥4000 m)which was the highest province in China was free of EHDV.The numbers of seropositive collections in both bovine and goat/sheep were in an inverse proportion to the latitude.However,the seropositive rates in bovine were ranged from 0%to 100%,while the seropositive rates in goat/sheep were no more than 50%.The results suggested that bovine was obviously more susceptive for EHDV infection than goat and sheep,therefore might be a major reservoir of EHDV in China.The prevalence of EHDV was consistent with the distribution of Culicoides which were known as the sole insect vectors of EHDV.In particular,the seropositive rates of EHDV were very high in the southern provinces,which required the enhanced surveillance in the future.

AIM:To develop a sensitive assay for screening compounds against hepatitis C virus (HCV).METHODS:The proteolytic cleavage of NS3/4A on enhanced yellow fluorescent protein (eYFP)-mitochondrial antiviral signaling protein (MAVS) was examined by reporter enzyme secreted placental alkaline phosphatase (SEAP),which enabled us to perform ongoing monitoring of anti-HCV drugs through repeated chemiluminescence.Subcellular localization of eYFP-MAVS was assessed by fluorescence microscopy.Cellular localization and protein levels were examined by Western blotting.RESULTS:HCV NS3/4A protease cleaved eYFP-MAVSfrom mitochondria to block the activation of interferon (IFN)-β promoter,thus resulting in downregulation of SEAP activity.The decrease in SEAP activity was proportional to the dose of active NS3/4A protease.Also this reporter assay was used to detect anti-HCV activity of IFN-α and cyclosporine A.CONCLUSION:Our data show that this reporter system is a sensitive and quantitative reporter of anti-HCV inhibitors.This system will constitute a new tool to allow the efficient screening of HCV inhibitors.

Objective To investigate the variability of human cytomegalovirus(HCMV)UL140 open reading frame(ORF)in clinical strains,and to explore the relationship between the variability of UL140 ORF and different symptoms of HCMV infection.Methods HCMV UL140 ORF was amplified by polymerase chain reaction and sequenced selectedly in 30 clinical strains.Results UL140 ORF of all clinical strains was amplified successfully.Compared with that of Toledo strain,the nucleotide and amino acid sequence identities among all strains were 96.5%-100.0% and 95.2%-100.0%,respectively.All of the nucleotide changes were substitutions.The post-translational modification sites were conserved.The result of phylogenetic tree showed that the strains did not cluster according to different clinical symptoms.Conclusion HCMV UL140 ORF in clinical strains is highly conserved,which may play an important role in HCMV infection.

Objective To investigate the variability of human cytomegalovirus(HCMV) UL138 open reading frame(ORF) in clinical strains.Methods HCMV UL138 ORF was amplified by polymerase chain reaction(PCR) and PCR amplification products were sequenced directly,and the data were analyzed in 19 clinical strains.Results UL138 ORF in all 30 clinical strains was amplified successfully.Compared with that of Toledo strain,the nucleotide and amino acid sequence identities of UL138 ORF in all strains were 97.41% to 99.41% and 98.24% to 99.42%,respectively.All of the nucleotide mutations were substitutions.The spatial structure and post-translational modification sites of UL138 encoded proteins were conserved.The result of phylogenetic tree showed that HCMV UL138 sequence variations were not definitely related with different clinical symptoms.Conclusion HCMV UL138 ORF in clinical strains is high conservation,which might be helpful for UL138 encoded protein to play a role in latent infection of HCMV.

Sequence variability of human cytomegalovirus UL143 in low-passage clinical isolates

Background Human cytomegalovirus （HCMV） infects a number of organs and tissues in vivo. The different symptoms and tissue tropisms of HCMV infection perhaps result from genetic polymorphism. A new region of DNA containing at least 19 open reading frames （ORFs）（denoted UL133 to 151） was found in the low-passage HCMV clinical strain, Toledo, and several other low-passage clinical isolates, but not present in the HCMV laboratory strain, AD169. One of these genes, UL143, was studied to explore the sequence variability of UL143 ORF in HCMV clinical isolates and examine the possible association between gcne variability and the outcome of HCMV infection. Methods The UL143 gone of the strains obtained from suspected congenitally HCMV-infectcd infants was amplified by polymerase chain reaction （PCR） and sequenced. Results Nineteen sequences of the strains were divided into 2 major groups, G1（n=16） and G2（n=3）. All of the sequences had frame-shift mutation compared to Toledo. Nucleotide polymorphisms conferred substantial amino acid substitutions when compared with Toledo. All 16 UL143 putative proteins of the strains in G1 had a new myristylation site and loss of two PKC sites owing to missense mutations. No convincing relationships were observed between the presence of HCMV disease and the UL143 sequence group. Conclusions HCMV-UL143 existed in low passage isolates. Sequence variability caused by frame -shift mutation was found in all HCMV clinical strains. No obvious linkage was observed between UL143 polymorphisms and the outcome of suspected congenital HCMV infection.

Plants and viruses coexist in the natural ecosystem for extended periods of time,interacting with each other and even coevolving,maintaining a dynamic balance between plant disease resistance and virus pathogenicity.During virus–host interactions,plants often exhibit abnormal growth and development.However,plants do not passively withstand virus attacks but have evolved sophisticated and effective defense mechanisms to resist,limit,or undermine virus infections.It is widely believed that the initial stage of infection features the most intense interactions between the virus and the host and the greatest variety of activated signal transduction pathways.This review describes the most recent findings in rice antiviral research and discusses a variety of rice antiviral molecular mechanisms,including those based on R genes and recessive resistance,RNA silencing,phytohormone signaling,autophagy and WUSmediated antiviral immunity.Finally,we discuss the challenges and future prospects of breeding rice for enhanced virus resistance.

Introduction:Seasonal influenza is a prevalent and highly contagious acute respiratory disease that causes a major global disease burden.New strains of influenza viruses also have the potential to cause an influenza pandemic.Due to the high variability and the consequent uncertainty about virus strains,the prevention and control of influenza are faced with many challenges.Surveillance is considered a key strategy to lead to the prevention and control of influenza,and influenza is one of first infectious diseases to be monitored globally.Methods:The Chinese National Influenza Surveillance Network conducts routine influenza surveillance and includes 410 network laboratories and 554 sentinel hospitals based on the requirements of National Influenza Surveillance Guidelines.China and other countries submit epidemiological and virological data to the WHO FluMart database,which can be viewed online.Results:Seasonal influenza H3N2 virus was the dominant subtype in China when entering the 2019–2020 winter influenza season,while neighboring countries such as Japan and Republic of Korea reported the dominance of H1N1pdm09 and Mongolia and Russia reported a higher proportion of type B virus than type A virus.Conclusions and Implications for Public Health Practice:Asian countries entered the 2019–2020 winter influenza season with different type/subtypes of influenza virus dominant in different areas.Influenza surveillance needs to be strengthened to closely monitor changes in the antigenicity and genetic characteristics of emerging viruses.

Transfusion-transmitted infections(TTIs)impose meticulous screening protocols in blood banks.Emerging or re-emerging viruses with altered pathogenicity remains debatable whether or not to include those in TTI screening panel.Dengue infection is known to transmit via Aedes aegypti mosquito;however,the route of infection through blood transfusion is recently of greater importance.India with hyper-endemic and endemic dengue regions must guarantee the safety in transfusion-related products.The donor with asymptomatic dengue infection during viremic phase can act as a reservoir for transmission.The acute infection in dengue is ascertained by detecting either nonstructural-1 protein(NS1)or IgM antibody whereas the prevalence of dengue can be monitored by the presence of IgG.

Background:Natural anti-sense transcripts(NATs),which are transcribed from the complementary DNA strand of annotated genes,exert regulatory function of gene expression.Increasing studies recognized anti-sense transcription widespread throughout human cytomegalovirus(HCMV)genome,whereas the anti-sense transcription of RNA1.2 gene locus has never been investigated.In this study,the transcription of the RNA1.2 anti-sense strand was investigated in clinically isolated HCMV strain.Methods:Strand-specific high-through RNA-sequencing(RNA-seq)was performed to find possible anti-sense transcripts(ASTs).For analyzing and visualization of RNA-seq data sets,Integrative Genomics Viewer software was applied.To confirm these possibilities,Northern blotting and rapid amplification of cDNA ends(RACE)were used.Results:Transcription of the opposite strand of RNA1.2 gene locus was detected by RNA-sequencing using RNAs extracted from human embryonic lung fibroblasts infected with HCMV clinical isolate HAN.At least three HCMV NATs,named RNA1.2 AST 1,RNA1.2 AST2,and RNA 1.2 AST3,were characterized by Northern blotting and RACE analyses.These RNA1.2 ASTs orientated from the complementary strand of RNA1.2 locus during the late phase of HCMV infection.The 5'-and 3'-termini of these transcripts were located within the opposite sequence of the predicted RNA1.2 gene.Conclusion:A cluster of novel NATs was transcribed from the opposite sequence of the HCMV RNA 1.2 gene region.
