The product yield and transgene instability of CHO cells need to be further increased and solved. Promoter and selection maker are essential elements on transgene expression. We investigated the effects of promoters, WPRE, and selection marker on transgene expression efficiency in CHO cells to enhance the transgene expression. In the present study, the CMV and SV40 promoters were combined with a weakened form of the Neo resistance gene NPT, and with the WPRE cis-acting element to generate expression vectors. Those vectors were transfected into CHO cells then use Flow cytometry, qRTPCR, qPCR, Western blot and ELISA to detect the expression of the transgene. The results showed that the weakened Neo resistance gene enhanced eGFP expression driven by the CMV and SV40 promoters. Moreover, a vector that WPRE combine with the Neo resistance gene considerably enhanced eGFP expression in CHO cells. However, the WPRE showed no enhancing effects when combined with the weakened Neo resistance gene. Furthermore, when the CMV promoter combined with the WPRE could result in higher cetuximab expression levels than the CMV promoter alone and could be useful for improving recombinant protein *** could not enhance the transgene expression under the circumstance of CMV and weakened Neo resistance gene. However, WPRE can improve transgene expression levels when CMV combined with the Neo resistance gene. That suggested that effects of the WPRE were selection marker-specific. The conclusion has a significant implication to industrial production of therapeutic proteins.

多维球面模型是从资源监测发展起来的 ,基于向量的多元分析方法 .向量是既有量值 ,又有方向的变量 .多维球面模型把传统的向量分析从三维扩展到任意多维 ,可以同时处理任意多个变量 .多维球面模型定义了向量除法 ,可以处理指数增长 ,因此特别适用于生物、金融、信息、经济等领域的变量 .应用于基金市场时 ,模型以基金为坐标轴建立多维基金空间 ,用多元向量表示基金市场 :以向量长度做指数表示市场的量值 ,以向量的方向表示市场状态 ;并用后前余弦的商表示市场的动态 ,称多维即时趋势 ;进而根据趋势值将基金排序 ,根据基金的序位决定售购 .在 7个月 ( 1 998.6.1 9～ 1 999.1 .2 9)的投资实验中 ,多维球面模型指导的个人退休基金的增率是 2 5.82 % .这个增率不仅超过了 2 0个基金市场的平均增率 ( 2 .64% ) ,而且超过了市场中最好的基金的增率 ( 1 8.98% ) .目前 ,尚不能用传统的风险或投机的概念来解释 MDSM的高增率 .多维球面模型假定变量无关。

Immunogenicity and protective efficacy study using combination of four tuberculosis DNA vaccines

Immune response and protective efficacy for the combination of four tuberculosis DNA vaccines were evaluated in this study. We obtained 1︰200 antibody titers against Ag85B 21d after mice were vaccinated for the first time by four recombinant eukaryotic expression vectors containing coding sequences for Ag85B, MPT-64, MPT-63 and ESAT-6. The titers of Ag85B were elevated to 1︰102400 after the second injection and decreased to 1︰12800 after the third injection. Antibody titers for MPT-64 and MPT-63 reached 1︰25600 21 d after the first vaccination, and were then decreased following the second and third injections. No antigen-specific antibody titer against ESAT-6 was detected in sera harvested from immunized mice at any time. These DNA vaccines evoked specific IFN-g responses in the spleens of vaccinated mice as well. When challenged with M. tuberculosis H37Rv, we found that the lungs of the vaccinated mice produced 99.8% less bacterial counts than that of the empty-vector control group and the bacterial counts were also significantly less than that of the BCG group. Histopathological analyses showed that the lungs of vaccinated mice produced no obvious caseation while over 50%—70% of the pulmonary parenchyma tissue produced central caseation in the vector control group. Our results indicated that the combination of four tuberculosis DNA vaccines may generate high levels of immune responses and result in better animal protection.

Novel antigen delivery systems

Vaccines represent the most relevant contribution of immunology to human health. However, despite the remarkable success achieved in the past years, many vaccines are still missing in order to fight important human pathologies and to prevent emerging and re-emerging diseases. For these pathogens the known strategies formaking vaccines have been unsuccessful and thus, new avenues should be investigated to overcome the failure of clinical trials and other important issues including safety concerns related to live vaccines or viral vectors, the weak immunogenicity of subunit vaccines and side effects associated with the use of adjuvants. A major hurdle of developing successful and effective vaccines is to design antigen delivery systems in such a way that optimizes antigen presentation and induces broad protective immune responses. Recent advances in vector delivery technologies, immunology, vaccinology and system biology, have led to a deeper understanding of the molecular and cellular mechanisms by which vaccines should stimulate both arms of the adaptive immune responses, offering new strategies of vaccinations. This review is an update of current strategies with respect to live attenuated and inactivated vaccines, DNA vaccines, viral vectors, lipid-based carrier systems such as liposomes and virosomes as well as polymeric nanoparticle vaccines and virus-like particles. In addition, this article will describe our work on a versatile and immunogenic delivery system which we have studied in the past decade and which is derived from a nonpathogenic prokaryotic organism: the "E2 scaffold" of the pyruvate dehydrogenase complex from Geobacillus stearothermophilus.

[Objective]To provide a reference for dynamic .regulation of the fermentation process. [Method] Using conditions for tylosin fermentation as feature vectors, final yield during the fermentation process was predicted using the Bayesian method. [ Rgesultl The Bayesian network which learned data of tylosin fermentation could better guide conditions of the fermentation process, and better prediction results were achieved. The yield of fermentation products could be effectively predicted using the obtained Bayesian network and could well guide the fermentation process. E Conclusion] Bayesian network has good predictive performance for microbial fermentation and has good practical yield and application prospects.

TPOS Tagging Method Based on BiLSTM_CRF Model

Part of speech (POS) tagging determines the attributes of each word, and it is the fundamental work in machine translation, speech recognition, information retrieval and other fields. For Tibetan part-of-speech (TPOS) tagging, a tagging method is proposed based on bidirectional long short-term memory with conditional random field model (BiLSTM_CRF). Firstly, the designed TOPS tagging set and manual tagging corpus were used to get word vectors by embedding Tibetan words and corresponding TPOS tags in continuous bag-of-words (CBOW) model. Secondly, the word vectors were input into the BiLSTM_CRF model. To obtain the predictive score matrix, this model using the past input features and future input feature information respectively learned by forward long short-term memory (LSTM) and backward LSTM performs non-linear operations on the softmax layer. The prediction score matrix was input into the CRF model to judge the threshold value and calculate the sequence score error. Lastly, a Tibetan part of speech tagging model was got based on the BiLSTM_CRF model. The experimental results indicate that the accuracy of TPOS tagging model based on the BiLSTM_CRF model can reach 92.7%.

Objective:To investigate the zoonotic visceral leishmaniasis(ZVL) by identification of the most probable reservoir hosts using parasite isolation and analysis of a possible transmission dynamics of the disease in extra-domestic agricultural fields and rural ***:Rodents were collected from selected study sites in kala-azar endemic areas based on information for localities of kala-azar cases for screening of Leshmania infections using parasitological,serological and polymerase chain reaction(PCR) from March.2013 to January,***(Clorketam Veterinary) was used to anaesthesize the rodents according the prescribed dosage(average 2 mg/kg for intra-venous route).The blood obtained using sterile needle was dropped into sterile filter paper and allowed to air dry before sealing in plastic *** tissues from liver,spleen and skin were macerated in Locke's solution before transferring them into NNN *** and touch smears of liver,spleen,skin and bone marrow were prepared for fixing using methanol and staining by Giemsa stain for *** tissues were also ascd for DNA extractions and PCR amplification of Leishmania ***:A total of 335 rodents(13 species) were analyzed by sampling internal *** infection rate by PCR was 11.1%(6/54) for Arvicanthis nilothicas compared to 17.6%(3/17) and 12.5%(2/16) for Acomys cahirinus and Tarera(C) robustus *** all the infections were found from bone marrow samples(8/48 or 16.7%) compared with 1/91(1.1%) liver,2/87(2.2%) spleen and 0/87(0%) *** all study sites with past human VL cases,rodents and proved vectors shared similar ***:Leishmania donovani might circulate among different species of rodents in kala-azar endemic lowlands and valleys of Ethiopia by Phlebotamus orientalis and Phlebotomus *** studies to substantiate the preliminary data on the possible role of these rodents arc urgently needed.

Neuroprotective effects of human telomerase reverse transcriptase on beta-amyloid fragment 25-35-treated human embryonic cortical neurons

BACKGROUND： Numerous current studies have suggested that human telomerase reverse transcriptase （hTERT） gene has neuroprotective effects and can inhibit apoptosis induced by various cytotoxic stresses; however, the mechanism of action remains unknown. OBJECTIVE： To evaluate the neuroprotective effects and possible mechanism of action of hTERT gene transfection in human embryonic cortical neurons treated with beta-amyloid fragment 25-35 （AI325-35）. DESIGN, TIME AND SETTING： The randomized, controlled and molecular biological studies were performed at the Department of Anatomy and Brain research, Zhongshan School of Medicine, Sun Yat-sen University, China, from September 2005 to June 2008. MATERIALS： AdEasy-1 Expression System was gifted by Professor Guoquan Gao from Sun Yat-Sen University, China. Human cortical neurons were derived from 12-20 week old aborted fetuses, obtained from the Guangzhou Maternal and Child Health Hospital, China. Mouse anti-Odk5 and mouse anti-p16 monoclonal antibodies （Lab Vision, USA）, and mouse anti-hTERT monoclonal antibody （Epitomics, USA）, were used in this study. METHODS：（1） Recombinant adenovirus vectors, encoding hTERT （Ad-hTERT） and green fluorescent protein （Ad-GFP）, were constructed using the AdEasy-1 Expression System. Human embryonic cortical neurons in the Ad-hTERT group were transfected with Ad-hTERT for 1-21 days. Likewise, human embryonic cortical neurons in the Ad-GFP group were transfected with Ad-GFP for 1-21 days. Human embryonic cortical neurons in the control group were cultured as normal. （2） Human embryonic cortical neurons in the Ad-hTERT group were treated with 10 pmol/L Aβ25-35 for 24 hours. Normal human embryonic cortical neurons treated with 10 pmol/Lβ25.35 for 24 hours served as a model group. Human embryonic cortical neurons in the Ad-GFP and control groups were not treated with Aβ25-35.
