To investigate the structure-activity relationship of polysaccharide and obtain a better antidepressant polysaccharide,the antidepressant-like activity of a carboxymethyl polysaccharide(C-MEPS2)subjected to submerged fermentation was systematically ***12-H cell and Kunming mice were used to investigate the differences and their mechanism in the antidepressant effects of C-MEPS2 and *** experiments have showed that C-MEPS2 has a better antidepressant effect than MEPS2.C-MEPS2 could exert antidepressant effects related to catecholamine synthesis with specifi c sites of TH,D2DR,and *** addition,C-MEPS2 could repair the Res-induced damage in PC12-H cell,stabilize the mitochondrial membrane potential and regulate intracellular Ca^(2+) concentration,thus reducing cell apoptosis caused by *** common dosing experiments on animals further proved that CMEPS2 could signifi cantly improve the antidepressant effect of derivatives without affecting the antidepressant mechanism of *** is speculated that it may be related to carboxymethylated modifi cation.

Marine urease with higher thermostability, pH and salinity tolerance from marine sponge-derived Penicillium steckii S4-4

Urease has a broad range of applications,however,the current studies on urease mainly focus on terrestrial plants or ***,it is quite necessary to determine if marine-derived ureases have diferent characteristics from terrestrial origins since the fnding of ureases with superior performance is of industrial *** this study,the marine urease produced by Penicillium steckii S4-4 derived from marine sponge Siphonochalina *** *** marine urease exhibited a maximum specifc activity of 1542.2 U mg protein−*** molecular weight of the enzyme was 183 kDa and a single subunit of 47 kDa was detected,indicating that it was a *** N-terminal amino acid sequence of the urease was arranged as GPVLKKTKAAAV with greatest similarity to that from marine algae Ectocarpus *** urease exhibited a K_(m) of 7.3 mmol L^(−1) and a V_(max) of 1.8 mmol urea min^(−1) mg protein^(−1).The optimum temperature,pH and salinity are 55℃,8.5 and 10%,*** urease was stable and more than 80%of its maximum specifc activity was detected after incubating at 25–60℃for 30 min,pH 5.5–10.0 or 0–25%salinity for 6 *** with the terrestrial urease from Jack bean,this marine urease shows higher thermostability,alkaline preference and salinity tolerance,which extends the potential application felds of urease to a great extent.

Towards enriching and isolation of uncultivated archaea from marine sediments using a refined combination of conventional microbial cultivation methods

The archaea that can be readily cultivated in the laboratory are only a small fraction of the total diversity that exists in *** molecular ecology methods,such as metagenomic sequencing,can provide valuable information independent of cell cultivation,it is only through cultivation-based experiments that they may be fully characterized,both for their physiological and ecological ***,we report our efforts towards enriching and isolation of uncultivated archaea from marine sediments using a refined combination of conventional microbial cultivation ***,cells were retrieved from the sediment samples through a cell extraction procedure and the sediment-free mixed cells were then divided into different size-range fractions by successive filtration through 0.8µm,0.6µm and 0.2µm *** 16S rRNA gene analyses indicated noticeable retention of different archaeal groups in different *** each fraction,supplementation with a variety of defined substrates(e.g.,methane,sulfate,and lignin)and stepwise dilutions led to highly active enrichment cultures of several archaeal groups with Bathyarchaeota most prominently ***,using a roll-bottle technique,three co-cultures consisting of Bathyarchaeota(subgroup-8)and a bacterial species affiliated with either Pseudomonas or Glutamicibacter were *** results demonstrate that a combination of cell extraction,size fractionation,and roll-bottle isolation methods could be a useful protocol for the successful enrichment and isolation of numerous slow-growing archaeal groups from marine sediments.

In eukaryotic cells,DNA damage triggers activation of checkpoint signaling pathways that coordinate cell cycle arrest and repair of damaged *** DNA damage responses serve to maintain genome stability and prevent accumulation of genetic mutations and development of *** p38 MAPK was previously implicated in cellular responses to several types of DNA ***,the role of each of the four p38 isoforms and the mechanism for their involvement in DNA damage responses remained poorly *** this study,we demonstrate that p38γ,but not the other p38 isoforms,contributes to the survival of UV-treated *** of p38γsensitizes cells to UV exposure,accompanied by prolonged S phase cell cycle arrest and increased rate of *** investigation reveal that p38γis essential for the optimal activation of the checkpoint signaling caused by UV,and for the efficient repair of UVinduced DNA *** findings have established a novel role of p38γin UV-induced DNA damage responses,and suggested that p38γcontributes to the ability of cells to cope with UV exposure by regulating the checkpoint signaling pathways and the repair of damaged DNA.

A Strategy to Optimize the Oligo-Probes for Microarray-based Detection of Viruses

DNA microarrays have been acknowledged to represent a promising approach for the detection of viral pathogens. However, the probes designed for current arrays could cover only part of the given viral variants, that could result in false-negative or ambiguous data. If all the variants are to be covered, the requirement for more probes would render much higher spot density and thus higher cost of the arrays. Here we have developed a new strategy for oligonucleotide probe design. Using type I human immunodeficiency virus (HIV-1) tat gene as an example, we designed the array probes and validated the optimized parameters in silico. Results show that the oligo number is significantly reduced comparing with the existing methods, while specificity and hybridization efficiency remain intact. The adoption of this method in reducing the oligo numbers could increase the detection capacity for DNA microarrays, and would significantly lower the manufacturing cost for making array chips.

Inteins--A Focus on the Biotechnological Applications of Splicing-Promoting Proteins

The main aim of this mini-review is to illustrate strategies and industrial applications based on inteins (INTErnal proteINS), which belong to a class of autocatalytic enzymes that are able to perform a catalytic reaction on a single substrate. However, since practical applications of inteins are strongly guided by a detailed understanding of their biological mechanisms and functions, the first part of this review will thus briefly discuss the physiological roles of inteins, describing what is currently known about their mechanisms of action. In the second part, specific biotechnological applications of inteins will be outlined (i.e. their use for (i) the purification of recombinant proteins, (ii) the cyclization of proteins and (iii) the production of seleno-proteins), paying attention to both potential strengths and weaknesses of this technology.
