Miniscope3D:optimized single-shot miniature 3D fluorescence microscopy

Miniature fluorescence microscopes are a standard tool in systems ***,widefield miniature microscopes capture only 2D information,and modifications that enable 3D capabilities increase the size and weight and have poor resolution outside a narrow depth ***,we achieve the 3D capability by replacing the tube lens of a conventional 2D Miniscope with an optimized multifocal phase mask at the objective’s aperture *** the phase mask at the aperture stop significantly reduces the size of the device,and varying the focal lengths enables a uniform resolution across a wide depth *** phase mask encodes the 3D fluorescence intensity into a single 2D measurement,and the 3D volume is recovered by solving a sparsity-constrained inverse *** provide methods for designing and fabricating the phase mask and an efficient forward model that accounts for the fieldvarying aberrations in miniature *** demonstrate a prototype that is 17mm tall and weighs 2.5 grams,achieving 2.76μm lateral,and 15μm axial resolution across most of the 900×700×390μm^(3) volume at 40 volumes per *** performance is validated experimentally on resolution targets,dynamic biological samples,and mouse brain *** with existing miniature single-shot volume-capture implementations,our system is smaller and lighter and achieves a more than 2×better lateral and axial resolution throughout a 10×larger usable depth *** microscope design provides single-shot 3D imaging for applications where a compact platform matters,such as volumetric neural imaging in freely moving animals and 3D motion studies of dynamic samples in incubators and lab-on-a-chip devices.

Growth promoting activity, anti-biofilm effect, and down regulation of papC and rcsA genes expression by Medicago sativa (alfalfa) extract

Consumption of Medicago sativa (alfalfa) has been linked to infection control in livestock. Therefore, studying the mechanism of this action may lead to the development of enriched food supplements. The purpose of this study was to investigate how M. sativa extract affects the growth of Lacticaseibacillus casei, Lactiplantibacillus plantarum, Staphylococcus aureus , and Escherichia coli . The effects of M. sativa extract on the bacterial cell adherence to Caco-2 cells (Human colon epithelial adenocarcinoma cell lines) and expression of genes involved in the formation of E. coli biofilm were also studied. Generally, in concentrations lower than 100 mg/ml, alfalfa extract had no antimicrobial effect on the probiotic and pathogenic bacteria. However, it not only acted as a growth promoter for two probiotic strains at 2.5% concentration but also increased the antibacterial effect of probiotics supernatants. Furthermore, alfalfa extract reduced the adhesion of pathogenic bacteria to Caco-2 cells. M. sativa extract was identified to inhibit the biofilm formation by E. coli through decreasing the genes expression, papC , and rcsA . As a result, it looks that alfalfa can improve the growth and function of beneficial bacteria, but further studies are needed to elucidate its mechanism of action.

BACKGROUND Antipsychotic drugs remain the mainstay of schizophrenia treatment;however,their effectiveness has been questioned,and it is not possible to predict the response to a specific antipsychotic drug in an individual ***,it is important to compare the effectiveness of the various antipsychotics and search for possible response *** To investigate the effectiveness of antipsychotic drugs,we examined response trajectories and predictors for belonging to different trajectory *** The Bergen-Stavanger-Innsbruck-Trondheim(BeSt InTro)trial compared the effectiveness of three atypical antipsychotics-amisulpride,aripiprazole,and olanzapine-in a prospective,semirandomized,rater-blind,head-to-head *** participants with a schizophrenia spectrum disorder diagnosis,according to international classification of diseases,Tenth Revision(ICD-10)F20–29,were *** were followed for a period of 12 mo,with assessments at baseline;after one,three and six weeks;and after three,six,nine and 12 mo.A latent class mixed model was fitted to our *** three-trajectory model based on the Positive and Negative Syndrome Scale(PANSS)total score reduction was found to have adequate fit,and the study drugs,as well as various demographic and clinical parameters,were tested as predictors for belonging to the different trajectory *** Overall,144 participants were included,and 41%completed the 12-mo study *** largest trajectory group,consisting of 74%of participants,showed a PANSS total score reduction of 59%from baseline to 12 mo(Good response group).A trajectory group comprising 13%of participants had their PANSS total score reduced by 82.5%at 12 mo(Strong response group),while the last response trajectory group comprising 13%of the participants had a PANSS total score reduction of 13.6%(Slight response group).The largest part of the total reduction for the Good and Strong response groups occurred at six weeks of treatment,

Objective This study aimed to investigate the effects of Montanide ISA-720 and Naloxone(NLX)in Hepatitis B surface antigen(HBsAg)vaccine formulation on cytokine and long-lasting antibody *** First,the HBsAg was formulated in Montanide ISA-720 adjuvant and Naloxone at 5 and 10mg/*** experimental mice were immunized three times at a 2-week interval,and then IL-4,IL-2,TNF-α,and IFN-γcytokines;long-lasting IgG antibody responses 220 days after the last shot;and IgG1/IgG2a isotypes were assessed by *** The HBsAg-Alum group exhibited the highest IL-4 cytokine response among the experimental groups,whereas NLX in HBsAg-MON720 vaccine formulation did not affect cytokine *** addition,NLX in Alum-based vaccine suppressed IL-4 cytokine response and increased the IL-2/IL-4 cytokine ***,HBsAg-MON720 was more potent than HBsAg-Alum in the induction of antibody responses,and NLX in Alum-and MON720-based vaccines induced long-lasting antibody *** NLX in Alum-based vaccine decreased IL-4 cytokine response,increased IL-2/IL-4 cytokine ratio,and improved long-lasting humoral immune responses in both vaccine ***,the adjuvant activity of NLX in the vaccine formulation depends on the type of adjuvant and the nature of the antigen in the vaccine formulation.

Inflammatory bowel disease(IBD)refers to a group of disorders characterized by chronic inflammation of the gastrointestinal(GI)*** elevated levels of nitric oxide(NO)in serum and affected tissues;mainly synthesized by the inducible nitric oxide synthase(iNOS)enzyme;can exacerbate GI inflammation and is one of the major biomarkers of GI *** natural and synthetic agents are able to ameliorate GI inflammation and decrease iNOS expression to the extent comparable with some IBD ***,the purpose of this study was to gather a list of natural or synthetic mediators capable of modulating IBD through the NO *** databases including Google Scholar and PubMed were searched from 1980 to May *** found that polyphenols and particularly flavonoids are able to markedly attenuate NO production and iNOS expression through the nuclear factorκB(NF-κB)and JAK/STAT signaling *** and probiotics can also alter the GI microbiota and reduce NO expression in IBD models through a broad array of mechanisms.A number of synthetic molecules have been found to suppress NO expression either dependent on the NF-κB signaling pathway(i.e.,dexamethasone,pioglitazone,tropisetron)or independent from this pathway(i.e.,nicotine,prednisolone,celecoxib,β-adrenoceptor antagonists).Co-administration of natural and synthetic agents can affect the tissue level of NO and may improve IBD symptoms mainly by modulating the Toll like receptor-4 and NF-κB signaling pathways.

A Review Featuring Fabrication, Properties and Applications of Carbon Nanotubes(CNTs) Reinforced Polymer and Epoxy Nanocomposites

Carbon nanotubes（CNTs） have long been recognized as the stiffest and strongest man-made material known to date. In addition, their high electrical conductivity has roused interest in the areas of electrical appliances and communication related applications. However, due to their miniature size, the excellent properties of these nanostructures can only be exploited if they are homogeneously embedded into light-weight matrices as those offered by a whole series of engineering polymers. In order to enhance their chemical affinity to engineering polymer matrices, chemical modification of the graphitic sidewalls and tips is necessary. The mechanical and electrical properties to date of a whole range of nanocomposites of various carbon nanotube contents are also reviewed in this attempt to facilitate progress in this emerging area. Recently, carbonaceous nano-fillers such as graphene and carbon nanotubes（CNTs） play a promising role due to their better structural and functional properties and broad range of applications in every field. Since CNTs usually form stabilized bundles due to van der Waals interactions, they are extremely difficult to disperse and align in a polymer matrix. The biggest issues in the preparation of CNTs reinforced composites reside in efficient dispersion of CNTs into a polymer matrix, the assessment of the dispersion, and the alignment and control of the CNTs in the matrix. An overview of various CNT functionalization methods is given. In particular, CNT functionalization using click chemistry and the preparation of CNT composites employing hyperbranched polymers are stressed as potential techniques to achieve good CNT dispersion. In addition, discussions on mechanical, thermal, electrical, electrochemical and applications of polymer/CNT composites are also included.

PLANE ELASTIC PROBLEM ON RIGID LINES ALONG CIRCULAR INCLUSION

The plane elastic problem of circular-arc rigid line inclusions is considered. The model is subjected to remote general loads and concentrated force which is applied at an arbitrary point inside either the matrix or the circular inclusion. Based on complex variable method, the general solutions of the problem were derived. The closed form expressions of the sectionally holomorphic complex potentials and the stress fields were derived for the case of the interface with a single rigid line. The exact expressions of the singular stress fields at the rigid line tips were calculated which show that they possess a pronounced oscillatory character similar to that for the corresponding crack problem under plane loads. The influence of the rigid line geometry, loading conditions and material mismatch on the stress singularity coefficients is evaluated and discussed for the case of remote uniform load.

Characteristics of machined surface controlled by cutting tools and conditions in machining of brittle material

One of the ultra-precision machining methods was adapted for brittle material as well as soft material by using multi-arrayed diamond tips and high speed spindle. Conventional machining method is too hard to control surface roughness and surface texture against brittle material because the particles of grinding tools are irregular size and material can be fragile. Therefore, we were able to design tool paths and machine controlled pattern on surface by multi-arrayed diamond tips with uniform size made in MEMS fabrication and high speed spindle, and the maximum speed was about 3×105 r/min. We defined several parameters that can affect the machining surface. Those were multi-array of diamond tips (n×n), speed of air spindle and feeding rate. The surface roughness and surface texture can be controlled by those parameters for micro machining.
