Identification of the major allergen of Macrobrachium rosenbergii (giant freshwater prawn)

Objective:To characterize the major allergens of Macrobrachium rosenbergii.(giant freshwater prawn).Methods:Raw and cooked extracts of the giant freshwater prawn were *** IgE reactivity pattern was identified by sodium dodecyl sulfate polyaerylamide gel electrophoresis(SDS-PAGE)and immunoblotting technique with the sera of 20 skin prick test(SPT)positive *** major allergen identified was then characterized using the proteomics approach involving a combination of two-dimensional(2-DE)electrophoresis,mass spectrometry and bioinformatics ***:SDS-PAGE of the raw extract showed 23 protein bands(15-250 kDa)but those ranging from 40 to 100 kDa were not found in the cooked *** immunoblotting experiments,raw and cooked extracts demonstrated 11 and 5 IgE-binding proteins,respectively,with a molecular mass ranging from 15 to 155 kDa.A heat-resistant 36 kDa protein was identified as the major allergen of both *** addition,a 42 kDa heat-sensitive protein was shown to be a major allergen of the raw *** 2-DE gel fractionated the prawn proteins to more than 50 different protein *** these,10 spots showed specific:IgE reactivity with patients'*** assisted laser desorption/ionization-lime of flight(MALDI-TOF)analysis led to identification of 2 important allergens,tropomyosin and arginine ***:It can be concluded that the availability of such allergens would help in component-based diagnosis and therapy of prawn allergies.

Since the publication of the DRiP(defective ribosomal product) hypothesis in 1996, numerous studies have addressed the contribution of DRiPs to generating viral antigenic peptides for CD8^+T cell immunosurveillance. Here, we review studies characterizing the generation of antigenic peptides from influenza A virus encoded DRiPs, discuss the many remaining mysteries regarding the nature of their co-translational generation, and speculate on where the future might lead.

Natural cytotoxicity receptors(NCRs)are expressed on natural killer(NK)cells and other subsets of innate lymphoid cells(ILC).Three NCRs,NKp30,NKp44 and NKp46,are expressed on human NK cells.1 NKp46 is the only NCR shared by primates and rodents,suggesting an essential role for this receptor during evolution.2 While cellular ligands for NKp30 and NKp44 have been identified,a well-characterized non-microbial ligand for NKp46 is still ***,in a recent paper in Science Immunology,Narni-Mancinelli et al.3 reported the identification of complement factor P(CFP,also known as properdin)as a ligand of NKp46.

BACKGROUND： Zingiber cassumunar Roxb., commonly known as Phlai in Thai, has been used as a traditional medicine in Thailand for the treatment of various diseases, including inflammation and chronic airway disease. OBJECTIVE： The purpose of this study was to assess the antihistaminic effect of Phlai on skin testing DESIGN, SETTING, PARTICIPANTS AND INTERVENTION： This was a randomized, open-label, three- way crossover study. Twenty allergic rhinitis （AR） patients were enrolled. In randomized sequence, patients received a single dose of Ph/ai capsules （100 or 200 mg） or Ioratadine （10 mg） with a washout period of 1 week between each treatment. MAIN OUTCOME MEASURES： Skin prick testing for histamine and common aeroallergen （house dust mite） were performed before treatment and after 1,2, 3, 4, 6, 8, 12 and 24 hours of treatment. The main treatment outcomes were the mean wheal and flare responses to the skin prick test after treatment. RESULTS： Both 100 mg and 200 mg Phlai doses suppressed wheal and flare responses to house dust mite allergen, but only 200 mg of Phlai capsules significantly suppressed wheal and flare responses to histamine. Repeated measures analysis of variance showed that Ioratadine caused more wheal and flare suppression than Phial capsules in responses to the histamine skin prick test. However, there were no significant differences among the effects of 100 mg Phlai capsules, 200 mg Phlai capsules and Ioratadine in suppression of wheal and flare induced by the mite skin prick test. Both doses of Phlai were well-tolerated with no adverse ***： Both 100 mg （compound D 4 mg） when taken as a single therapeutic dose, inhibited AR patients. and 200 mg （compound D 8 mg） Phlai capsules, skin reactivity to histamine and mite skin prick tests in TRIAL REGISTRATION： Thai clinical trial registry （TCTR20160510001）

To the Editor:The overall prevalence of asthma among Chinese adults(aged≥20 years)was 4.2%,representing 45.7 million people.[1]With high disease burden of asthma in China,it needs to better understand the status of asthma treatment in China from physicians’and patients’*** 2021 Global Initiative of asthma(GINA)report developed two Treatment Tracks based on the choice of relievers.[2]For patients with moderate asthma(Steps 3 and 4),the recommended regimen is low-or medium-dose inhaled corticosteroid(ICS)/formoterol as daily maintenance therapy with additional low-dose ICS/formoterol as needed(known as maintenance and reliever therapy[MART])in Track 1,or medium-or high-dose ICS/long-acting b2-agonist(LABA)as daily maintenance therapy with additional short-acting b2-agonist(SABA)as needed in Track 2.

Increased levels of surfactant protein D (SP-D) and lipid-laden foamy macrophages (FMs) are frequently found under oxidative stress conditions and/or in patients with chronic obstructive pulmonary disease (COPD) who are also chronically exposed to cigarette smoke (CS). However, the roles and molecular mechanisms of SP-D and FMs in COPD have not yet been determined. In this study, increased levels of SP-D were found in the bronchoalveolar lavage fluid (BALF) and sera of ozone- and CS-exposed mice. Furthermore, SP-D-knockout mice showed increased lipid-laden FMs and airway inflammation caused by ozone and CS exposure, similar to that exhibited by our study cohort of chronic smokers and COPD patients. We also showed that an exogenous recombinant fragment of human SP-D (rfhSP-D) prevented the formation of oxidized low-density lipoprotein (oxLDL)-induced FMs in vitro and reversed the airway inflammation and emphysematous changes caused by oxidative stress and CS exposure in vivo. SP-D upregulated bone marrow-derived macrophage (BMDM) expression of genes involved in countering the oxidative stress and lipid metabolism perturbations induced by CS and oxLDL. Our study demonstrates the crucial roles of SP-D in the lipid homeostasis of dysfunctional alveolar macrophages caused by ozone and CS exposure in experimental mouse emphysema, which may provide a novel opportunity for the clinical application of SP-D in patients with COPD.

Objective To identify factors affecting the efficacy of steroid-eluting sinus stents implanted after endoscopic sinus surgery(ESS)in patients with chronic rhinosinusitis with nasal polyps(CRSwNP).Methods We performed a post-hoc analysis of a randomized self-controlled clinical trial on post-operative implantation of bioabsorbable steroid-eluting stents in patients with *** logistic regression analysis was conducted to identify which of the following factors affect the response to post-operative stent implantation:sex,serum eosinophil levels,history of prior surgery,endoscopic scores,and comorbid conditions(asthma and allergic rhinitis).The primary outcome was the rate of post-operative intervention on day 30,and the secondary outcome was the rate of polypoid tissue formation(grades 2–3)on days 14,30,and *** A total of 151 patients with CRSwNP were included in the post-hoc *** was identified as the only risk factor for a poor response to steroid-eluting sinus stents on post-operative day 30,with an odds ratio of 23.71(95%CI,2.81,200.16;P=0.004)for the need for post-operative intervention and 19(95%CI,2.20,164.16;P=0.003)for moderate-to-severe polypoid tissue *** addition,the asthmatic group showed higher rates of post-operative intervention and polypoid tissue formation than the non-asthmatic group on post-operative day *** eosinophil levels were not identified as a risk factor for poor outcomes after stent *** Comorbid asthma,but not blood eosinophil level,impairs the efficacy of steroid-eluting sinus stents in the short term after ESS in patients with CRSwNP.

Assessing global fungal threats to humans

Fungi are an integral part of the earth's *** are broadly distributed in all continents and ecosystems and play a diversity of ***,I review our current understanding of fungal threats to humans and describe the major factors that contribute to various *** the 140,000 or so known species out of the estimated six million fungal species on Earth,about 10%directly or indirectly threaten human health and *** threats include mushroom poisoning,fungal allergies,infections of crop plants,food contamination by mycotoxins,and mycoses in humans.A growing number of factors have been identified to impact various fungal threats,including human demographics,crop distributions,anthropogenic activities,pathogen dispersals,global climate change,and/or the applications of antifungal drugs and agricultural ***,while models have been developed for analyzing various processes of individual threats and threat managements,current data are primarily descriptive and incomplete,and there are significant obstacles to integration of the diverse factors into accurate quantitative assessments of fungal *** increasing technological advances and concerted efforts to track the spatial and temporal data on climate and environmental variables;mycotoxins in the feed and food supply chains;fungal population dynamics in crop fields,human and animal populations,and the environment;human population demographics;and the prevalence and severities of fungal allergies and diseases,our ability to accurately assess fungal threats will *** improvements should help us develop holistic strategies to manage fungal threats in the future.
