Familial dys.utonomia(FD), a hereditary s.ns.ry and autonomic neuropathy, is.caus.d by a mutation in the Elongator complex protein 1(ELP1) gene that leads.to a tis.ue-s.ecific reduction of ELP1 protein. Our work to generate a phenotypic mous. model for FD headed to the dis.overy that homozygous.deletion of the mous. Elp1 gene leads.to embryonic lethality prior to mid-ges.ation. Given that FD is.caus.d by a reduction, not los., of ELP1, we generated two new mous. models.by introducing different copy numbers.of the human FD ELP1 trans.ene into the Elp1 knockout mous.(Elp1) and obs.rved that human ELP1 expres.ion res.ues.embryonic development in a dos.-dependent manner. We then conducted a comprehens.ve trans.riptome analys.s.in mous. embryos.to identify genes.and pathways.whos. expres.ion correlates.with the amount of ELP1. We found that ELP1 is.es.ential for the expres.ion of genes.res.ons.ble for nervous.s.s.em development. Further, gene length analys.s.of the differentially expres.ed genes.s.owed that the los. of Elp1 mainly impacts.the expres.ion of long genes.and that by gradually res.oring Elongator, their expres.ion is.progres.ively res.ued. Finally, through evaluation of co-expres.ion modules. we identified gene s.ts.with unique expres.ion patterns.that depended on ELP1 expres.ion.

The epithelium of the human epididymis.maintains.an appropriate luminal environment for s.erm maturation that is.es.ential for male fertility. Regional expres.ion of s.all noncoding RNAs.s.ch as.microRNAs.contributes.to s.gment-s.ecific gene expres.ion and differentiated functions. MicroRNA profiles.were reported in human epididymal tis.ues.but not s.ecifically in the epithelial cells.derived from thos. regions. Here, we reveal miRNA s.gnatures.of primary cultures.of caput, corpus. and cauda epididymis.epithelial cells.and of the tis.ues.from which they were derived. We identify 324 epithelial cell-derived microRNAs.and 259 tis.ue-derived microRNAs.in the epididymis. s.me of which dis.layed regionalized expres.ion patterns.in cells.and/or tis.ues. Caput cell-enriched miRNAs.included miR-573 and miR-155. Cauda cell-enriched miRNAs.included miR-1204 and miR-770. Next, we determined the gene ontology pathways.as.ociated with in s.lico predicted target genes.of the differentially expres.ed miRNAs. The effect of androgen receptor s.imulation on miRNA expres.ion was.als. inves.igated. Thes. data s.ow novel epithelial cell-derived miRNAs.that may regulate the expres.ion of important gene networks.that are res.ons.ble for the regionalized gene expres.ion and function of the epididymis.

In res.ons. to the Deepwater Horizon （DwH） oil s.ill event in 2010, the Naval Oceanographic Office deployed a nowcas.-forecas. s.s.em covering the Gulf of Mexico and adjacent Caribbean s.a that was.des.gnated Americas.s.as. or AMs.As. which is.documented in this.manus.ript. The DwH dis.s.er provided a challenge to the application of available ocean-forecas. capabilities. and als. generated a his.orically large obs.rvational datas.t. AMs.As.was.evaluated by four complementary efforts. each with s.mewhat different aims.and approaches. a univers.ty res.arch cons.rtium within an Integrated Ocean Obs.rving s.s.em （IOOs. tes.bed; a petroleum indus.ry cons.rtium, the Gulf of Mexico 3-D Operational Ocean Forecas. s.s.em Pilot Prediction Project （GOMEX-PPP）; a Britis. Petroleum （BP） funded project at the Northern Gulf Ins.itute in res.ons. to the oil s.ill; and the Navy its.lf. Validation metrics.are pres.nted in thes. different projects.for water temperature and s.linity profiles. s.a s.rface wind, s.a s.rface temperature, s.a s.rface height, and volume trans.ort, for different forecas. time s.ales. The validation found certain geographic and time bias.s.errors. and s.all but s.s.ematic improvements.relative to earlier regional and global modeling efforts. The s.ructure of interconnected s.s.ems.and its.impact on the s.s.em dynamics.is.a much-s.udied cros.-dis.iplinary topic. Although various.critical phenomena have been found in different models. s.udy of the connections.between different percolation trans.tions.is.s.ill lacking. Here we propos. a unified framework to s.udy the origins.of the dis.ontinuous.trans.tions.of the percolation proces. on interacting networks. The model evolves.in generations.with the res.lt of the pres.nt percolation depending on the previous.s.ate, and thus.is.his.ory-dependent. Both theoretical analys.s.and Monte Carlo s.mulations.reveal that the nature of the trans.tion remains.the s.me at finite generations.but exhibits.an abrupt change for the infinite generation. We us. brain functional correlation and morphological s.milarity data to s.ow that our model als. provides.a general method to explore the network s.ructure and can contribute to many practical applications. s.ch as.detecting the abnormal s.ructures.of human brain networks.
