Dual stimulus responsive borosilicate glass(BSG)scaffolds promote diabetic alveolar bone defectsrepair by modulating macrophage phenotype

The regeneration of alveolar bone is still clinical challenge,particularly accompanied with diabetes,causing metabolic disorder with a protracted low-grade inflammatory phenotype.As a result,the anticipated loading of biomaterials is highly suspicious in spontaneous modulation of cells function,which is mostly disturbed by constant inflammation.In this study,we developed glucose and hydrogen peroxide dual-responsive borosilicate glass(BSG)scaffolds loaded with epigallocatechin gallate(EGCG)to synergistically modulate the abnormal inflammation of diabetic alveolar bone defects.It was found that the release of EGCG by BSG could directly regulate the shift of macrophages from M1 to the M2 phenotype by promoting autophagy and lessening the inhibition of autophagic flux.Moreover,EGCG can also indirectly regulate the polarization phenotype of macrophages by reducing the activation of NF-κb in stem cells and restoring its immunoregulatory capacity.Therefore,the addition of EGCG to BSG scaffold in diabetes allows for a more striking modulation of the macrophage phenotype in a timely manner.The altered macrophage phenotype reduces local inflammation and thus increases the ability to repair diabetic alveolar bone,showing promise for the treatment of alveolar defect in diabetic patients.

Urban growth does not always strictly follow historical trends;the government may reshape urban growth patterns with considerations of ecological conservation or other plans.Both urban dynamic rules and landscape characteristics are the two main factors influencing the spatial patterns of cities,and obtaining an optimized spatial pattern is veiy important for sustainable urban growth.Therefore,in this study,we integrated logistic regression(LR)with the ant colony optimization(ACO)model to analyze the optimal scenario for smart urban growth.The LR model was used to discuss the relationship between urban patterns and environmental variables such as topography,development centers,and traffic conditions.Then,the urban growth probability was generated using the parameters obtained from LR.The ACO model was further integrated to optimize urban land allocation,which can meet the requirement of high growth probability,and a connected and compacted landscape pattern.This can solve the problem of urban land only being allocated by LR from being distributed fragmentarily in the space.With this integrated model,Guangzhou City,a rapidly developing area in China,was selected as a case study.The urban patterns derived from LR,as well as a simulation scenario using logistic regression-based cellular automata(LR-CA),were used in the comparison.Six landscape metrics were chosen to validate the performance of this proposed model at the pattern level.The results show that the LR-ACO model has a better performance in urban land allocation.This study demonstrated that models that couple dynamic rules and planning objectives can provide plausible scenarios for smart urban growth planning.

Early detection of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection is an efficient way to prevent the spread of coronavirus disease 2019(COVID-19).Detecting SARS-CoV-2 antigen can be rapid and convenient,but it is still challenging to develop highly sensitive methods for effective diagnosis.Herein,a lateral flow assay(LFA)based on fluorescent nanoparticles emitting in the second near-infrared(NIR-II)window is developed for sensitive detection of SARS-CoV-2 antigen.Benefiting from the NIR-II fluorescence with high penetration and low autofluorescence,such NIR-II based LFA allows enhanced signal-to-background ratio,and the limit of detection is down to 0.01 ng·mL^(−1)of SARS-CoV-2 antigen.In the clinical swab sample tests,the NIR-II LFA outperforms the colloidal gold LFA with higher overall percent agreement with the polymerase chain reaction test.The clinical samples with low antigen concentrations(~0.015–~0.068 ng·mL^(−1))can be successfully detected by the NIR-II LFA,but fail for the colloidal gold LFA.The NIR-II LFA can provide a promising platform for highly sensitive,rapid,and cost-effective method for early diagnosis and mass screening of SARS-CoV-2 infection.

Injectable bone biomaterials like bone cement should be designed and fabricated with certain biological criteria,which include:1)recruitment and polarization of the macrophages from M1(pro-inflammatory)to M2(anti-inflammatory)phenotype,2)enhance vascularization,and 3)activate osteogenic differentiation of bone marrow-derived stem cells to promote bone healing.So far,no injectable biomaterials could spontaneously regulate the entire bone healing process that involves inflammation,angiogenesis,and osteogenesis.Therefore,in this study,we designed bone cement comprised of strontium and copper-incorporated borosilicate glass(Sr/Cu-BSG)in the liquid phase of chitosan to modulate bone healing.In vitro studies showed that the controlled release of Sr and Cu ions up-regulated anti-inflammatory genes(IL-1Ra and TGF-β1)while down-regulating pro-inflammatory genes(IL-1βand IL-6)in macrophages at 3 days.Sr and Cu ions also increased the expressions of angiogenic genes(VEGF and bFGF)in HUVECs at 5 days and osteogenic genes(Runx-2,OCN,and OPN)in hBMSCs at 7,14,and 21 days.5Sr3Cu-BSG bone cement exhibited the best anti-inflammatory,angiogenic,and osteogenic properties among the bone cement groups with different Sr and Cu ratios.Short-term and long-term implantation of Sr/Cu-BSGs in femoral condylar bone defects of rats and rabbits confirmed the in vitro results,where the degradation rate of Sr/Cu-BSG matched the bone healing rate.Similar to in vitro,the 5Sr3Cu-BSG group also showed the highest bone formation in vivo.Excellent physical and chemical properties,along with its bone repairing ability,make the Sr/Cu-BSG bone cement a good candidate biomaterial for treating bone defects.

OBJECTIVE:To investigate molecular mechanisms of Jinwujiangu decoction(金乌健骨方,JWJG)in treating osteoarthritis(OA)using network pharmacology analysis.METHODS:Principal active compounds of JWJG were screened out via the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)and BATMAN-TCM,and potential targets for OA treatment were identified through Online Mendelian Inheritance in Man(OMIM)and Gene Cards databases.The JWJG network diagrams of both principal chemical compound-action targets and OA treatment target-OA disease were constructed applying the Cytoscape 3.7.2 software.The diagram of protein-protein interaction network was plotted for core analysis.Meanwhile,the common targets and relevant signaling pathways involved in both networks were analyzed using the Gene Ontology function analysis and Kyoto Encyclopedia of Genes and Genomes pathway enrichment.The predicted results were ultimately verified through animal experiments.RESULTS:Effects of JWJG were indicated in acting on key targets interleukin-6,insulin,protein kinase B,glyceraldehyde-3-phosphate dehydrogenase,and mitosis-specific MRE11-RAD50-NBS1 associated protein by regulating signaling pathways of phosphoinositide 3-kinase-protein kinase B,mitogen-activated protein kinases,tumor necrosis factor,and colorectal cancer.Meanwhile,it inhibited the over-activation of signaling pathways and the release of inflammatory factors in OA treatment.Following a signaling pathway analysis utilizing network pharmacology technique,it was demonstrated that JWJG could treat OA through the Wnt/β-catenin signaling pathway verified by animal experiments.CONCLUSIONS:The present study preliminarily analyzed the pharmacological mechanism of JWJG in treating OA on a network pharmacology approach and provided a great theoretical significance for clinical application.

Patient-derived tumor xenograft(PDX)models,a method involving the surgical extraction of tumor tissues from cancer patients and subsequent transplantation into immunodeficient mice,have emerged as a pivotal approach in translational research,particularly in advancing precision medicine.As the first stage of PDX development,the patient-derived orthotopic xenograft(PDOX)models implant tumor tissue in mice in the corresponding anatomical locations of the patient.The PDOX models have several advantages,including high fidelity to the original tumor,heightened drug sensitivity,and an elevated rate of successful transplantation.However,the PDOX models present significant challenges,requiring advanced surgical techniques and resourceintensive imaging technologies,which limit its application.And then,the humanized mouse models,as well as the zebrafish models,were developed.Humanized mouse models contain a human immune environment resembling the tumor and immune system interplay.The humanized mouse models are a hot topic in PDX model research.Regarding zebrafish patient-derived tumor xenografts(zPDX)and patient-derived organoids(PDO)as promising models for studying cancer and drug discovery,zPDX models are used to transplant tumors into zebrafish as novel personalized medical animal models with the advantage of reducing patient waiting time.PDO models provide a cost-effective approach for drug testing that replicates the in vivo environment and preserves important tumor-related information for patients.The present review highlights the functional characteristics of each new phase of PDX and provides insights into the challenges and prospective developments in this rapidly evolving field.

Background: The root of Tripterygium wilfordii(Tripterygii radix), a natural powerful traditional Chinese medicine(TCM) for various diseases treatment, has been used for centuries in the Asian countries as anti-rheumatoid arthritis(RA) agent, antioxidant agent, and anti-inflammatory agent. Its combination with other herbs in treating RA has been explored. The anti-RA effect of T. wilfordii for cancer treatment has been supported by some evidence. Aims and Objectives: To investigate the anticancer mechanism of T. wilfordii, bioinformatics databases were used to identify its active ingredients. Materials and Methods: Target proteins associated with cancer were determined using a network pharmacology analysis platform, and 25 key active compounds and 55 key targets of T. wilfordii were identified in our study. A common potential mechanism of T. wilfordii involvement in cancer was disclosed by in-depth network analysis of diseases, functions, and pathways.Finally, the analysis results of the TCM-disease target protein interaction network revealed 5 potential targets;subsequently, a total of 30targets(these 5 targets, as well as 25 previously identified compounds) were subjected to molecular docking. Results: Our results showed that the therapeutic effect of T. wilfordii in cancer is characterized by multiple components, targets, and pathways. The regulation of signaling pathways such as Kaposi sarcoma-associated herpes virus infection, colorectal cancer, small-cell lung cancer, and prostate cancer may be the important pharmacodynamic basis of anticancer therapy. Conclusion: Triptonoditerpenic acid inhibited proliferation and induced apoptosis in SW480 cells. The mechanism may be related to the downregulation of Bcl-2 expression, upregulation of Bax mRNA expression, and expression inhibition of PTGS2.

Antioxidant Activities of Crude Phlorotannins from Sargassum hemiphyllum

Brown algae are well known as a source of biologically active compounds, especially those having antioxidant activities, such as phlorotannins. In this study we examined the antioxidant activities of crude phlorotannins extracts（CPEs） obtained from Sargassum hemiphyllum（SH） and fractionated according to the molecular weights. When CPEs were administrated at a dose of 30 mg/kg to Kunming mice pre-treated with carbon tetrachloride（CCl4）, the levels of oxidative stress indicators in the liver, kidney and brain were significantly reduced in vivo. All the components of various molecular weight fractions of CPEs exhibited greater scavenging capacities in clearing hydroxyl free radical and superoxide anion than the positive controls gallic acid, vitamin C and vitamin E. Particularly, the components greater than 30 k D obtained from ethyl acetate phase showed the highest antioxidant capacities. These results indicated that SH is a potential source for extracting phlorotannins, the algal antioxidant compounds.
